[cze][eng] 135 Závěr Jedním z cílů této studie bylo zavedení a optimalizace laboratorních protokolů izolace a typizace aDNA molekuly. U tří vybraných kosterních souborů se sledovaly atributy - pohlaví, přítomnost specifické DNA patologické bakterie a příbuznost. V průběhu studia aDNA jsme vymýšleli a optimalizovali jednotlivé postupy, které ve výsledku vedly k ohodnocení pohlaví nedospělých jedinců z Mikulčic a potvrzení přítomnosti Mycobacterium leprae na kostrách dvou jedinců ze Žatce. Obě výše uvedené lokality spadají do období raného středověku. Pohlaví bylo možno genetickými přístupy určit pro 78,7% jedinců souboru. Byl zaznamenán signifikantně vyšší výskyt mužských jedinců oproti ženským a to v poměru 24:13. Tyto výsledky byly porovnány s výsledky studie, která se zaměřila na morfoskopické hodnocení kosti pánevní a dolní čelisti. Vysoká shoda obou přístupů velmi pravděpodobně vypovídá o vysoké autenticitě izolované aDNA tohoto souboru. Na dvou jedincích se znaky deformit na kosterním materiálu byla úspěšně detekována část DNA specifická pro druh Mycobacterium leprae a tím i potvrzena předem vyslovená domněnka o příčinném vztahu této bakterie k zaznamenaným kostním deformitám. U posledního souboru se i přes řadu různých optimalizací laboratorních postupů nepodařilo na základě studia genetické příbuznosti pomocí...6 Coxcr-usroN The aim of our studywas to establisha laboratoryprotocolfor isolationand analysisof aDNA.
