1 SOUHRN Tato disertační práce se zabývá hledáním potenciálních léčiv, konkrétně přípravou a studiem biologické aktivity derivátů přírodních látek - chalkonů. Součástí práce je příprava meziproduktů - 5- alkylovaných pyrazin-2-karbonitrilů a 1-pyrazin-2-ylethanonů. Jako finální produkty byly syntetizovány substituované chalkony, několik sérií pyrazinových analogů chalkonů a Michaelovy adukty chalkonů s různými thioly. V této části práce byly použity známé metodiky reakcí. Produkty byly identifikovány teplotou tání, resp. teplotou varu, IČ a NMR spektry, příp. MS spektry. Jejich čistota byla ověřena elementární analýzou. Celkem bylo připraveno  22 meziproduktů (z toho 9 dříve popsaných na našem pracovišti)  62 finálních látek (z toho 19 dříve popsaných)  1 vedlejší produkt Podařilo se připravit 57 dosud nepopsaných látek. Syntéza 12 sloučenin byla neúspěšná. Konečné produkty byly testovány na antimykobakteriální, antifungální, antialgální a antiagregační účinky. Chalkony byly zkoušeny také na inhibici jedné z cysteinových proteas - papainu a v návaznosti na tuto část práce byla provedena předběžná studie reakční kinetiky adice chalkonů s látkami obsahujícími thiolovou skupinu. Minoritní díl této práce tvoří část zabývající se syntézou 2-benzyl-2-hydroxybenzofuran-3(2H)-onu jako přírodního auronolu. V této... 1 SUMMARY This dissertation thesis deals with searching for potential drugs, particularly with preparation and biological activity study of natural compounds derivatives - chalcones. The work includes preparation of intermediates - 5-alkylated pyrazin-2-carbonitriles and 1-pyrazinyl-2-ethanones. Substituted chalcones, several series of pyrazine analogues of chalcones and Michael adducts of chalcones with different thiols were prepared as final products. In this part of the work, known reaction methodologies were used. Products were identified by melting point, resp. boiling point, IR, NMR spectra. Their purity was checked by elemental analysis. In the case of liquids, elemental analysis could not be performed, and MS spectra were recorded instead. In total were prepared  22 intermediates (9 of them have been described in our department)  62 final products (19 of them have been already described)  1 by-product 57 Unknown compounds were prepared. Synthesis of 12 compounds was not successful. The final products were tested for their antimycobacterial, antifungal, antialgal, and antiaggregating effects.
