Microfluidic circuit

Microfluidics is a science dealing with flow of a very small amounts of fluids and is mainly connected with medicine and biochemistry. One of a main problem, which this science is facing, is a difficult mixing of fluids due to occurrence of laminar flows. This bachelor thesis contains a design of a microfluidic circuit with Staggered herringbone microfluidic mixer and subsequent implementation of the experiment according to the performed design.
Methods of mounting of acoustic emission sensors on the surface of tested structures

This bachelor thesis is focused on the issue of mounting acoustic emission sensor to cylindrical and sferical surface of tested tube segments which are made of non-ferromagnetic material. This work includes overview of current state of cognition in the basics of acoustic emission testing needed for understanding the issue. The folowing part includes the overview of commonly used practices for installing transducers. In the part of mechanical design drafts of mounting fixtures are discussed along with testing and comparision of their characteristics.
Changes in selected postural stability tests in hemiparetic patients after botulinum toxin application

The subject of this bachelor thesis is to compare changes in selected clinical and posturographic tests of postural stability, before and after application of botulinum toxin A to selected lower limb muscles in patients with spastic hemiparesis. The theoretical part deals with the introduction of general information related to posture, such as clinical tests of postural stability, their significance and use, as well as the issue of falls. The next part concerns spastic paresis, its origin, causes, consequences and also clinical scales that evaluate spasticity. The last theoretical part summarize the treatment options of spastic paresis. The practical part describes in detail the kinesiological analyzes of patients and the results of clinical and posturographic tests. Functional Reach Test, Timed Up and Go and Mini BESTest were selected from the clinical trials. Within the posturography, the Weight Bearing Squat test, Modified CTSIB and Limits of Stability were selected. The overall results of this research were not entirely clear, as the individual tests focus on different aspects of postural stability. Some tests were better after botulinum toxin application and some of them before botulinum toxin application. However, impairment of postural stability was in relation to the hemiparetic half of the body. Research has also shown that vision is a key aspect of postural stability in hemiparetic patients. This research could serve as inspiration in the treatment of postural stability not only in hemiparetics, but also in stability disorders in general.
Disc friction loss in centrifugal pumps and hydraulic turbines

Dizertační práce je zaměřena na výzkum proudění v mezidiskových spárách hydraulických strojů s důrazem na čerpadla s nízkými specifickými otáčkami. Navzdory tomu, že tekutina v mezidiskových prostorách čerpadel zaujímá velmi malý objem, může výrazně ovlivnit proudění v celém stroji. To má následně vliv na provoz, charakteristiky a celkovou účinnost, zvlášť v případě čerpadel s nízkými specifickými otáčkami, protože stav proudění určuje vznikající diskové ztráty. Rovněž ovlivňuje rozložení tlaku a tím výslednou axiální sílu. Porozumění proudění v mezidiskových spárách je zatím omezené, současné poznatky vychází z teorie odvozené pro značně zjednodušený případ disku rotujícího v hladké válcové spáře. Použitelnost pro popis proudění ve skutečných hydraulických strojích je diskutabilní. Za účelem výzkumu proudění v reálném odstředivém čerpadle bylo navrženo a sestaveno experimentální zařízení umožňující pozorování proudění v zadní mezidiskové spáře pomocí optických metod, měření krouticího momentu a vznikající axiální síly. Také byla popsána metodika pro numerické simulace tohoto druhu proudění umožňující podchytit nestability a vírové struktury. Numerické simulace vhodně doplňují experimentální měření, rozšiřují rozmezí studovaných provozních podmínek a pomáhají popsat jevy, které by šlo měřením jen obtížně odhalit.
Aerodynamic properties of different front wing specifications on formula race car

This bachelor’s thesis focuses on the aerodynamic properties of front wings on Formula 1 cars. The first chapter discusses the basic aerodynamic principles of fluids and gases. The second chapter discusses the historical evolution of front wings in Formula 1. Third chapter talks about the latest aerodynamic regulations and their benefits for the sport. In the last chapter there is a practical part with an air flow simulation on four different wings and their aerodynamic properties are compared.

A defect in this protein's trafficking from cytoplasm to cytoplasmic membrane is thought to play a role in development of Sjögren's syndrome - a chronic autoimmune disease. Prolactin-inducible protein is a cytoplasmic protein which was found to be contributing to this disease's pathogenesis. This project was aimed to study the proposed interaction between aquaporin 5 and prolactin-inducible protein. Both recombinant proteins were overexpressed in the yeast Pichia pastoris and purified using low-pressure column chromatography on an ÄKTA purification system. Glycosylation of prolactin-inducible protein was confirmed using specific glycoprotein staining. Two coelution tries were run on a HisTrap column. Parkinson's disease (PD) is the second most common ageing-related neurodegenerative disorder after Alzheimer's disease and the prevalence in population is increasing. The characteristic movement disorder is caused by selective dopaminergic neurons loss, while the mechanism of this neurodegeneration is not well understood. Increasing evidence points out the key role of vicious cycle of microglial overactivation and oxidative stress, while the questions "where it begins" and "how to stop it" remain without clear answers. Surrounding structures including proteins may be damaged in case the content of a phagocyte is spilled to the vicinity. Catechins belong to a group of flavonoids whose antioxidant properties were demonstrated in many studies. My diploma thesis was focused on a potential effect of eight naturally occurring derivatives of catechin towards oxidative damage of human serum albumin (HSA) by hypochlorous acid. I used two spectrophotometric (determination of carbonyl groups and chloramines) and three electrophoretic methods (native PAGE, SDS-PAGE, immunoblotting). Dependence of an antioxidant effect on its concentration (0-3 mM) was observed during determination of chloramines and obtained data were used for calculation of EC50 values of individual catechins using program GraphPad Prism. Lenka Javorská Supervisor: RNDr. Lucie Škarydová, Ph.D Title of diploma thesis: Detection of His-tag and FLAG-tag recombinant proteins Recombinant proteins are proteins produced in genetically modified organism by recombinant DNA technology that enable insert DNA sequence of target protein in expression cell. This technology allows preparation of actually any protein in any quantity without difficulties associated with demanding purification of native protein from tissues. Recombinant proteins are produced often in form of fusion proteins with suitable peptide tag (e.g. His-tag, FLAG- tag) that can help with their detection and enable simple purification. The aim of this diploma thesis was to introduce and optimize universal method for the detection of recombinant proteins labelled with His-tag (6x histidine) and FLAG-tag. The model recombinant protein with His-tag was enzyme carbonylreductase 1 (CBR1) produced by expression system Escherichia coli.
