Stability and properties of combined beverages and fruit concentrates

This diploma thesis deals with the stability and basic properties of combined beverages and fruit concentrates. The theoretical part describes the production and use of fruit concentrates. Furthermore, the work deals with anthocyanic pigments and phenolic substances. The principle and instrumentation of liquid chromatography and electron paramagnetic resonance were described. In the next subchapter, the methods of determination of total anthocyanins and phenolic substances were described. The experimental part of the thesis described the determination of soluble solids, viscosity and antioxidant activity. The total content of phenolic compounds were determined by the Folin-Ciocalteua method and the total anthocyanins were determined by the pH-differential method. In the samples were determined cyanidine-3-glucoside and cyanidin-3-galactoside by liquid chromatography.
Analysis of selected nutritional parameters in the new varieties of gooseberries

Diploma thesis deals with the new varieties of gooseberries. The theoretical part characterizes gooseberry from a biological point of view, what is the content of healthful nutrients and states food technology aspects. Furthermore, the theoretical part deals with the main parameters examined nutrition: vitamin C, anthokyanům and antioxidant activity. In the experimental part was measured both weight solids content and soluble solids. Vitamin C was measured by HPLC, anthocyanins using a pH differential method and for antioxidant activity determination was selected method by Kanedy known also as DPPH.
Isolation of compounds from Cannabis sativa and their anti-inflammatory effect

The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on ubiquitous polyphenolic compounds – flavonoids, also present in the studied plant. All previously researched and confirmed effects on human body are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform and hexane yields of extract obtained from this plant. Chromatographic methods were used for isolation – thin layer and column chromatography and semipreparative HPLC. Obtained substances were characterized using UV, IR spectrophotometry and NMR. Then the anti-inflammatory activity of obtained substances and also of the ethanolic extract and its yields was observed. The result of the experimental part was the isolation of one pure substance which was identified as the cannabidiolic acid. Using the THP 1 assay, we demonstrated mild anti-inflammatory effect of the non-polar yields and isolated CBDA.
Selected nutritional parameters of some lesser known fruit

Diploma thesis is focused on the chemical analysis of the juices of black, red and white currant and jostaberry (Ribes a Ribes x culverwellii). The theoretical part is focused to the definition of fruit, fruit importance in human nutrition, description of Ribes and Ribes x culverwellii genuses, active substances in the currants and the jostaberry, description of selected chemical parameters and the determination of some of them. The procedures of determining for the following chemical parameters are described in the experimental section: determination of reducing sugars, determination of total and soluble solids, formol number, pH, titratable acidity, determination of total phenols and anthocyanins and determination of vitamin C in two samples jostaberry, six varieties of black currants, one variety of white currant and two varieties of red currants. Based on the evaluation of the obtained results, the comparison was made with different varieties and the thesis that "overall comparison of the different varieties in the content of phenolic compounds, vitamin C and anthocyanins appears to be the most promising variety of blackcurrant demon. It should however be noted that the highest content of phenolics and anthocyanins were determined in jostaberry„ has been suggested.
The selection and optimization of the method for assessment of free fatty acids

This master´s thesis deals with optimization and validation of method for determination of free fatty acids. The theoretical part is focused on possibilities of extraction, extract separation, esterification, gas chromatography determination and brief description of the most important validation parameters. All with main emphasis on applying on free fatty acids. The experimental part describes the selection and procedure of optimizing individual steps of the method. Extraction of lipids was performed by solvent mixture (diethylether/petroleumether), based on the standard ČSN EN ISO 1735, for the extraction of free fatty acid solid phase extraction was tested. The acid catalyzed esterification based on ČSN EN ISO 5509 using methanolic solution BF3 was used for derivatization of free fatty acids and a gas chromatograph with a flame ionization detector was used for their determination. Subsequently, selected validation parameters of the method were verified: repeatability, reproducibility, linearity, limits of detection and quantitation. Sunflower oil and emmental type cheese, bought in common food market, were used as model samples for selection and optimatization of the method.
Assessment of aroma active compounds in chosen fruit types

This bachelor thesis deals with the assessment of aroma active compounds in less known berries, gooseberries. In the theoretical part the general characteristics, biological signs and properties of selected varieties, chemical composition and structure of the components, drug effects, the possibility of processing and use of gooseberries are mainly described. The main aim of this thesis was to determine aroma active substances in selected varieties of gooseberry fruits using SPME-GC method. Aroma active compounds were first extracted by solid-phase microextraction and then assessed by gas chromatography.
Comparison of vitamin C content between less known kinds of fruit

This thesis deals with the determination of vitamin C in different varieties of cornelian cherry (Cornus mas) and mountain ash (Sorbus aucuparia). The theoretical part describes the botanical characteristics of both plants, the extension in the CZ and in the world, methods of reproduction, the content of meaningful health substances and the importance of both plants in food-processing industry. Attention is paid to vitamin C, which is in their fruits in high quantity. Conclusion of the theoretical part discusses various methods of determination of vitamin C. The aim of the experimental part was to comparison of the vitamin C content in different varieties cornelian cherry and mountain ash. The chosen method for the determination of vitamin C by high performance liquid chromatography (HPLC) with UV detection was first tested on standards. Selected validation parameters of this method was then determined. Validated method was used to determine the content of vitamin C in nine varieties cornelian cherry and six varieties of the mountain ash. For comparison were also tested some samples of the determination of total vitamin C after reducing dehydroascorbic acid by using dithiothreitol (DTT).
Introduction of the method for assessment of conjugated linoleic acid (CLA)

This work deals with the optimization of the method for determination of conjugated linoleic acid (CLA) using gas chromatography. The summary about formation and occurrence of CLA in animal materials, its biological effects in human organism and methods suitable for its determination is introduced in the theoretical part of this study. The experimental part verify, if the gas chromatography is applicable method for assesment of CLA in selected biological matrices. The chosen method was introduced and verified at FCH of Brno university of technology. At the end the applicability of this method to CLA determination is discussed here.
Positive effects of conjugated linoleic acid on human health

This bachelor thesis deals with the characteristics of the fatty acids, their structure, nomenclature, properties, reactions and analysis. It is also dedicated to the history, sources, effects on human health and the correct ratio of omega-fatty acids in the body. The main issue of this work is conjugated linoleic acid, which is known to its countless positive effects, which can include weight loss and lower level of cholesterol, anticarcinogenic effects, prevention of osteoporosis and arteriosclerosis, etc. The work is focused on its characteristics, structure, occurrence in food, chemical production and a wide range of preparations available on the market. Last but not least, deals with the possibilities of its determinations, including gas chromatography, high performance liquid chromatography, gas chromatography-mass spectrometry, NMR spectroscopy, and combined methods.
Analysis of czech garlic varieties

This master thesis deals with analysis of Czech garlic varieties in comparison with two varieties from abroad. Alliin and carbohydrates were determined by high performance liquid chromatography with diode-array detector or evaporative light scattering detector. Crude proteins were calculated from total content of nitrogen that was determinated by Kjeldahl method. Elementary analysis was performed by inductively coupled plasma optical emission spectrometry. In theoretical part of this thesis is describes botanical characterization and cultivation of garlic, chemical composition of garlic, especially groups of substances which were analysed in this thesis. Principle of used methods is described too. The experimental part of this work deals with sample and calibration solution preparation and process of analysis. Obtained results from analysed substances are summarized in the tables, discussed and compared with results of analysis of other authors. Between Czech and Spanish/Chinese garlic are slight differences. Garlics contain 24,66 g/kg dry weight of alliin. Content of crude proteins in Czech garlic is comparable to Chinese garlic but the Spanish garlic has just half of amount crude proteins. Elementary analysis also includes significant differences. Czech varieties contain higher amount of phosphorus, iron and zinc. On the contrary, content of potassium in foreign garlic is higher than in Czech Republic and the analysed amount is 15 g/kg dry weight.
