Tato diplomová práce se zabývá optimalizací a validací metody pro stanovení volných mastných kyselin. Teoretická část je zaměřená na možnosti extrakce, rozdělení extraktu, esterifikaci, stanovení na plynovém chromatografu a stručný popis nejdůležitějších validačních parametrů. Vše s hlavním důrazem kladeným na aplikaci na volné mastné kyseliny. V experimentální části je uveden výběr a postup optimalizace jednotlivých kroků metody. Pro extrakci lipidů byla zvolena extrakce směsí rozpouštědel (diethylether/petrolether) vycházející z normy ČSN EN ISO 1735, pro zisk frakce volných mastných kyselin byla testována extrakce na pevnou fázi. Pro derivatizaci byla vybrána kysele katalyzovaná esterifikace vycházející z normy ČSN EN ISO 5509 za použití methanolového roztoku BF3 a pro stanovení methylesterů mastných kyselin byl použit plynový chromatograf s plamenově-ionizačním detektorem. Následně byly ověřeny vybrané validační parametry metody: opakovatelnost, reprodukovatelnost, linearita, meze detekce a meze stanovitelnosti. Jako modelové vzorky pro výběr a optimalizaci metody byly použity slunečnicový olej a sýr ementálského typu zakoupené v běžné tržní síti. This master´s thesis deals with optimization and validation of method for determination of free fatty acids. The theoretical part is focused on possibilities of extraction, extract separation, esterification, gas chromatography determination and brief description of the most important validation parameters. All with main emphasis on applying on free fatty acids. The experimental part describes the selection and procedure of optimizing individual steps of the method. Extraction of lipids was performed by solvent mixture (diethylether/petroleumether), based on the standard ČSN EN ISO 1735, for the extraction of free fatty acid solid phase extraction was tested. The acid catalyzed esterification based on ČSN EN ISO 5509 using methanolic solution BF3 was used for derivatization of free fatty acids and a gas chromatograph with a flame ionization detector was used for their determination. Subsequently, selected validation parameters of the method were verified: repeatability, reproducibility, linearity, limits of detection and quantitation. Sunflower oil and emmental type cheese, bought in common food market, were used as model samples for selection and optimatization of the method.
