Mammalian Serine Racemase as a Pharmaceutical Terget
Vaníčková, Jana ; Konvalinka, Jan (advisor) ; Kotora, Martin (referee) ; Křen, Vladimír (referee)
PH.D. DISSERTATION Mammalian Serine Racemase as a Pharmaceutical Target Jana Jirásková Supervisor: Jan Konvalinka Department of Biochemistry Faculty of Science Charles University Institute of Organic Chemistry and Biochemistry Gilead Sciences & IOCB Research Centre Academy of Sciences of the Czech Republic Praha 2010 Introduction Serine racemase is a pyridoxal-5'-phosphate (PLP) -dependent enzyme that is responsible for D- serine production. D-serine is a neurotransmitter that acts, together with L-glutamate, as agonist of ionotropic N-methyl-D-aspartate (NMDA) receptors, which are important for neuronal tissue signalization. Recent serine racemase knock-out mouse studies revealed that SR produces approximately 90% of brain D-serine. SR was first isolated from a pool of rat brains about a decade ago. Its orthologs are present in mammals as well as plants and yeast. Mammalian SRs share high sequence identity, about 90%. Mouse and human SRs are similar enzymatically, suggesting that mouse SR and mouse model are suitable to shed light on human SR. SR forms homodimers in solution and has a molecular weight of approximately 37 kDa per monomer. In addition to pyridoxal-5'-phosphate, the enzyme requires divalent cations such as Ca2+ or Mg2+ , nucleotides such as ATP, and reducing agents for full activity. The...
Tactics to avoid false testimony
Vaníčková, Jana ; Konrád, Zdeněk (advisor) ; Štourač, Petr (referee)
Resumé The tactical procedures if overcoming the false testimony The subject of my diploma work is the tactical procedures of overcoming the false testimony. I also dealt with methods, which can be used for revelation and overcoming the untrue testimony. The central concepts which are connected with tactical procedures and methods are the testimony and the interrogation. The interrogation and testimony can be characterized from the psychological, criminalistic and legal view. Concerning my diploma work the criminalistic view is the most important. Criminalistics defines the interrogation as the criminalistic method, which is used (under the law) to obtain criminalisticly- and law-important information from the memory of the interrogated person. This is done by the testimony and the rights of the both of the interrogater and the interrogated person have to be strictly observed. In this method are used the tactical procedures, which can help examiner to overcome the false testimony. I evaluated all tactical procedures and methods according to their reliability and effect in the fight against the untrue testimony. I also dealt with the possibility of using the unusual tactical procedures and methods in the future. I compared all procedures and methods if they are right for overcoming the false testimony of the...

Creating optimal conditions for personal and social developement
Vaníčková, Jana ; Krištofová, Zuzana (referee) ; Šnýdrová, Ivana (advisor)
This thesis deals with personal and social development and its support in the area of education. The starting point is a humanistic view of an individual as a being with an inborn disposition to growth and self-actualization. Personal and social development is paid a great deal of attention to by almost all philosophies and religions, mostly in ideal characterizations of a human. In the area of education, personal and social development is one of the main goals of both pedagogy and andragogy. It is presently realized particularly through the development of key competencies. The aim of this thesis is to find those practical implementations of pedagogy and andragogy that contribute most to personal and social development. The essentials of each of these implementations are open, partnership-based communication, giving and receiving feedback and motivation to develop one's potential. What proves to be particularly suitable is the use of small group settings, the experience of "here and now" and its follow-up reflections.
Invertebrates in contaminated ecosystems - landfills
Vaníčková, Jana ; Hlava, Jakub (advisor) ; Husák, Tomáš (referee)
This bachelor thesis deals with invertebrates, in particular soil invertebrates, which inhabit the anthropogenically influenced enviroment at landfill of municipal waste. In the theoretical part of this work the most common types of soil invertebrates and basic ecological concepts are described. The practical part included the processing of individual samples and also sampling sites description. To obtain the material I have chosen four different landfills, where the soil samples were collected. The extraction of soil fauna was realized using Tullgrens extractors. The extracted samples were examined in lab conditions under binocular and the soil fauna were divided into taxonomic groups, according to their morphological characteristics. Sampling locations did not represent a different ecosystems. The activities are carried out standardly, according to the law. There is a part of restoration area in each locality. The aim of the practical part was to describe the composition of soil fauna communities in the landfill of municipal waste. Furthermore, this work deals with the comparison of individual sites and for a better interpretation, results are presented by tables and graphs. The observed diversity is interpreted using diversity and evenness indices. The results show that the most dominant groups forming the fellowship of soil mites without incrusted body (in the work presented as Acari in general). Further, as another of the dominant group I found rich communities of the incrusted Oribatid mites and springtails (Collembola), which are considered as a typical soil fauna. Most of these representatives was found in Sono landfill, on the other hand the minimum specimen were found in the Tušimice landfill. In the overall evaluation, samples did not represent significant differences in the composition of the soil invertebrates communities.
