Hematopoiesis in the models of zebrafish and medaka as a recipient for human HSC xenograft

Danio rerio (zebrafish) and Oryzias latipes (medaka) have recently become popular model organisms to study hematopoiesis. These model organisms present several advantages in comparison to other commonly used models, the most common being Mus musculus (mouse). The advantages are shorter generation time, large offspring production, frequent spawning, external fertilization and development, the optical transparency of embryos amenable to genetic manipulation on the background of vast numbers of transgenic lines (mainly in zebrafish) and inbred strains (in medaka). Moreover, most of the mechanisms behind zebrafish and medaka hematopoiesis are conserved in higher vertebrates. Most importantly, the optical transparency in early development and in adult mutant transparent strains allows for observation of hematopoietic stem cell (HSC) development in vivo. Therefore, it is possible to generate humanized fish using xenotransplanted human HSCs for studies of the engraftment, differentiation, and trafficking of human HSC in vivo. Currently, the most popular organism for human HSC xenotransplantation is mice. This model system is not suitable for in vivo imaging of HSC engraftment. Project of revitalization part of Kninicky brook

The diploma thesis is focused on the proposal of systematic revitalization of the selected section of the Knínický brook in the cadastral area of Veverské Knínice. It is a straight, very deep channel due to technical adjustments in the past. The HEC-RAS program verifies the capacity of the channel for the Q100 flow. In this way, it was found that the channel has very high capacity so a new route with Q1 flow capacity was designed. Due to the height conditions, the design also included three boulder chutes. Subsequently, the vegetation accompaniment and bank stands were suitably supplemented.
Single railway electrification system design

The main topic of the thesis is the design of single-track electrification of a direct current system of 3 kV and an alternating current system of 5 kV 50 Hz and then economically evaluate it over a period of 30 years. The work deals with basic traction systems and also deals with the calculation of main parameters of electric trains. The thesis describes in detail the procedure for the design of the electrification of the track and a partial introduction to the simulation programs that were used for energy calculations.
Analysis of ZED stereocamera in outdoor environment

The Master thesis is focused on analyzing stereo camera ZED in the outdoor environment. There is compared ZEDfu visual odometry with commonly used methods like GPS or wheel odometry. Moreover, the thesis includes analyses of SLAM in the changeable outdoor environment, too. The simultaneous mapping and localization in RTAB-Map were processed separately with SIFT and BRISK descriptors. The aim of this master thesis is to analyze the behaviour ZED camera in the outdoor environment for future implementation in mobile robotics.
Network Protocols Semiautomatic Diagnostics

This thesis is about semiautomatic network protocol diagnostics and creating protocol description from eavesdropped communication. Several network eavesdropping techniques  and some common programs for network analysis are introduced. Well-known network protocols are described, with focus on their communication messages. Some already existing methods for creating models from examples are mentioned and their characteristics defined. Next we design architecture of developed tool and some methods, that create protocol description. After that we explain implementation of this tool and finally the tool is tested and experimented with.
American foreign policy and Colombian peace process

This study analyzes American foreign policy with respect to Colombia since the beginning of the new millennium. The main pillar of this policy is a strategy called Plan Colombia. While the original reasoning behind Plan Colombia was to enforce better antinarcotic strategy to avoid cocaine distribution to the United States, by the time it morphed into a state-building effort. My main interest in this study is to answer the question whether Plan Colombia was successful and if so then whether it was a prerequisite for launching peace negotiations with the FARC and achieving durable peace. This study has several parts. At first, I shed a light into a situation in Colombia at the beginning of the new millennium and explain the motivation behind Plan Colombia. Next, I summarize the historical perspective and describe the most important events leading to the breakthrough which was the signing of the peace treaty with FARC. In the following section, I look more deeply into data about money transfers and I also analyze important socio-political indicators. In the conclusion, I explain why Plan Colombia was in my view a successful policy. I also discuss upon what was the success dependent.
Application of fluorescence staining for the "Scratch Wound Healing Assay" experiment

This bachelor’s thesis studies the cell culture of 3T3 cell line (mouse embryonic fibroblast cells) and cell migration. The Scratch Wound Healing Assay was used for the study of cell migration. The basic principles of experimenting, its advantages and disadvantages, as well as the practical use of this method in scientific research were presented on specific examples. Practical part of the bachelor’s thesis describes the experiment in the cell laboratory including cell culture of 3T3 cell line. The proposed experiment was tested with a sufficient number of repetitions. The statistical processing of microscopic images was performed in the MATLAB programming environment.
Transfection of somatic cells using magnetic nanoparticles and their post-transfection localization

This bachelor thesis is focused on an experiment based on magnetic particles SPIO for transfection of 3T3 cells. Nanoparticle surface was modified with PEI and Chitosan to optimize this method. The transfection experiment was performed twelve times, then the scanned data was processed and statistically analyzed in Matlab R2016a to determine the transfection efficiency. The theoretical part is focused on magnetic nanoparticles and their application in medicine, on transfection and in last part on a transfection of cells using magnetic nanoparticles.

Electrophysiological characterization of Kir2.1 membrane channel

The topic of this thesis is electrophysiological characterization of Kir2.1 membrane channel. Inward rectifier potassium channel Kir2.1 is located in muscular, heart and nerve cells and its dysfunction causes various diseases. Practical part of this stage is focused on cultivation of the HEK293T cell line that is used to transfection of the plasmid Kir2.1 and subsequent measurement of the ionic current through the electrophysiological method patch-clamp in whole-cell mode.
