Hrádek u Nechanic - Local Architecture and Its Source of Inspiration
Sieglová, Kateřina ; Kuthan, Jiří (advisor) ; Novotná, Eva (referee)
HRÁDEK U NECHANIC - ARCHITECTURE AND ITS INSPIRATORY SOURCES Chateau Hrádek u Nechanic was built by František Arnošt, the count of Harrach (1799-1884) as a new representative residence of the county family and also as a Harrach's earldom administrative centre. The foundation-stone of the carcass was layed in the year 1839 and the ceremonial laying foundation of the chapel took place two years later. The construction was officially completed in the year 1857 but works in interiors were practically proceeding during the whole second half of the 19th century. Subject of this work is architecture of the chateau Hrádek u Nechanic and its inspiratory sources. We talk about first large aristocratic building in the whole Habsburg Imperium that was in such an integral resemblance influenced by the english Gothic Revival phenomenon - an architecture, an interiors culture and a lifestyle. As an author of building plans is despite many obscurities considered Edward Buckton Lamb (1806-1869). It makes from count František Arnošt of Harrach propably the only one developer in the Habsburg Imperium who gave an order of a project for his chateau directly to an english architect. During the construction were made changes and adjustments by the vienesse architect Karl Fischer who took also a control of bulding. At the...
The role of human papilloma viruses in ethiopathogenesis malignant tumors in sinonasal area
Sieglová, Kateřina ; Nachtigal, Petr (advisor) ; Petera, Jiří (referee)
The aim of this thesis is the comparison of immunohistochemical and molecular biological methods for the detection of human papillomavirus (HPV) in tumour tissue and their subsequent use in the study of HPV role in the etiopathogenesis of malignant tumors of sinonasal region. Several methods for the purpose of HPV detection were applied: in situ hybridization, immunohistochemical detection of expression of p16INK4a protein, classic polymerase chain reaction combined with two different enzyme immunoassay detection methods, and reverse hybridization, followed by polymerase chain reaction with real time reverse transcription. All methods (p16 DNA in situ hybridization, RNA in situ hybridization, quantitative polymerase chain reaction with reverse transcription - RT-qPCR mRNA) used for the detection of HPV performed very well when compared, showing concordance with statistical significance p ˂ 0,0001. Detection of p16 vs. DNA polymerase chain reaction (PCR) had correlation coefficient p = 0.004 in the whole group, and p = 0.002 in the subgroup of squamous cell carcinoma (SCC). Correlation coefficient between detection of p16 and overall HPV status was 0,659 for entire set and 0,864 for set of SCC, indicating significant linear dependence. Immunohistochemical detection of p16 in combination with the...
Neo-Gothic rebuilding of Castle Hluboká nad Vltavou
Sieglová, Kateřina ; Kuthan, Jiří (advisor) ; Záruba, František (referee)
Diploma thesis "Neo-Gothic rebuilding of chateau Hluboká nad Vltavou" first provides a brief summary of basic literary and archival sources, followed by description of terminology in the field of 19th century architecture together with the explanation used by the author. After that there is mentioned a situation in Czech architecture at the end of 18th and in the 19th century. Next point is the Gothic Revival phenomenon - its establishment, ways of its spreading and its influence in middle Europe including Czech countries. Own topic of this thesis is introduced by a general and building history of chateau Hluboká nad Vltavou. After that there are introduced prince Jan Adolf II. of Schwarzenberg and his wife, who managed the rebuilding of the chateau. There is also a description of their personal contacts with British culture and architecture. Main part of the thesis is dedicated to Neo-Gothic rebuilding of chateau Hluboká nad Vltavou. This chapter is divided into several subchapters, where are described changes in the chateau interior and exterior including the effort of document the direct influence of Gothic Revival, and also the adaptation of the close surroundings of the chateau. Keywords Neo-Gothic architecture, Great Britain, Gothic Revival, chateau Hluboká nad Vltavou, prince Jan Adolf II....

Detection of circulating antibodies to determination of autoimmuno diseases by means of immunofluorescence.
Sieglová, Kateřina ; Nachtigal, Petr (advisor) ; Laco, Jan (referee)
Kateřina Sieglová Detection of circulating antibodies to determination of autoimmuno diseases by means of imunofluorescence. Bachelor thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Medical Laboratory Technician Background: The aim of this thesis is to explore of coherence between the presence of antibody against anti brush border antibody (ABBA) and idiopatic bowel disease (Crohn's disease and ulcerative colitis). Methods: Evidence of connection idiopatic bowel disease with positive ABBA test is examine by two perspectives, which are used for selecting and comparising of data. The first way is comparison of the number ABBA positive patients with their diagnosis of endoscopic biopsy. The second way check the positivity or negativity of ABBA examination of all patients with diagnose of idiopatic bowel disease. Experimental part summarize by this way the results, which was done at the Fingerland institute of pathology since 2009 to 2011. Results: The survey showed that among all the ABBA-positive patients is a large group of those who don't have the morphological signs of Inflammatory Bowel Disease - 72,47%. 66.67% of patients with a clinical diagnosis of Crohn's disease, and 39.37% of patients with ulcerative colitis are ABBA positive. Conclusion: The results shows, that...
