This Master's thesis deals with the processing of a business plan for establishing a family guesthouse in Vysočina region in the village Tři Studně. The theoretical basis of the thesis includes development and growth of the company, company strategy, external and internal environment analysis, customer analysis and SWOT analysis. The practical part analyzes the current situation, the description of the pension and its surroundings. The design part calculates the cost of the project, predicts sales and average cost of accommodation.
Evaluation of the Financial Situation of a Company and Proposals for Improvement

This bachelor thesis evaluates the financial situation of the company IMOS Brno, a.s. between the years 2011-2015 and the formulation of proposals for its improvement. At first are summarizes the theoretical knowledge of financial analysis that will be used to develop in the practical part. In the next part are captured the basic information about the company and analyzes the current status of the selected company. Analysis of the company is then used in the last part for the creation of proposals to improve the current financial situation.
Whole genome synthesis: methodology and applications

Genomes contain the information needed to support life and viruses. Understanding this information is no small task but an essential one for the understanding of life and its evolution. Genome sequencing can provide useful information about the meaning of genetic information, although it is now clear that sequencing alone cannot provide all the answers. For deeper comprehension of life's intricacies, synthesis of genomic DNA will extend our current knowledge and offer a platform for further applications. The immense growth in the methodology of DNA synthesis in the last decades facilitated the construction of the first cellular genome in 2008. Continuous advances in genome synthesis and engineering are the driving forces of synthetic biology. The history, current state and future applications of the whole genome synthesis are presented in this thesis. Analysis of Histone Deacetylase 6/Kinesin Interactions

Intracellular transport is provided by two major types of molecular motors kinesins and cytoplasmic dynein. Kinesin-1 is a molecular motor that transports molecules and organelles along microtubule tracks anterogradely. Specific protein-protein interactions are required to activate kinesin-1 as the free kinesin exist in an autoinhibited state. The activation of kinesin-1 induces its conformational change, enables microtubule binding and ATP hydrolysis necessary for the directional cargo transport. HDAC6 is a multifunctional protein composed of several domains. It plays an important role in many microtubule dependent processes as HDAC6 is a major tubulin deacetylase. It has been shown that HDAC6 manipulation (inhibition/genetic ablation) affects transport along microtubules but the exact mechanisms are unknown. The effect can be caused either by deacetylation microtubules or direct interaction with molecular motors. This thesis is focused on characterization of interactions between kinesin-1 and HDAC6 that have not been described so far. To this end, we expressed and purified various constructs of kinesin-1 and HDAC6 and tested their interactions by microscale thermophoresis (MST) and hydrogen deuterium exchange (HDX) to determine affinity and interaction sites, respectively. Phylogenetic and ecological impact to the microstructures of mollusc shells

This bachelor thesis summarizes the available information on microstructures of mollusc shells. The aim was to provide an overview of the different types of microstructures for specific specimens from different environments, as well as to obtain as much information as possible about the ecology of these animals. The first part summarizes the basic information on the shell formation. The following chapter summarizes known microstructure types of mollusc shells. Microstructures of molluscs living in terrestric, freshwater and marine environment are discussed. The influence of temperature changes on the shell microstructure of V. viviparus, the occurrence of vaterit within C. fluminea shells, the extreme environment of hydrothermal vents and cold seeps, as well as extreme temperatures on rocky shores, have been mentioned in more detail. The microstructures of related molluscs are compared with regard to ecology to determine whether the occurrence of microstructures in a particular group is given by the phylogeny of these animals, or if the changes in microstructures can be affected by different ecology. The effect of Ca2+ ion on the enzymatic activity of human glutamate carboxypeptidase II

Human glutamate carboxypeptidase II (GCPII, EC 3.4.17.24) is a homodimeric membrane glycoprotein. GCPII has been studied as a marker of prostate carcinoma and a therapeutic target of neurodegenerative disorders. The extracellular region of the protein is composed of three domains, apical, protease and C-terminal. There are two zinc ions in the active site that are essential for the enzymatic activity. A calcium ion is located between apical and protease domains near the dimeric interface approximately 20 Å away from the active site. Consequently, the Ca2+ ion in unlikely to participate in substrate hydrolysis. The aim of this thesis is to elucidate the function of Ca2+ in GCPII using a combination of molecular-biological, biochemical and biophysical approaches. To this end we prepared series of GCPII variants with mutations in calcium-coordinating amino acids. The mutant constructs were expressed in insect S2 cells and purified by combination of affinity and size exclusion chromatography. Enzymatic activity and thermostability of the mutants were decreased significantly. Furthermore, mutated proteins were aggregation prone and formed a monomeric GCPII species. Level of prevention tumors in breasts in the Strakonice region

The number of women newly diagnosed with breast cancer is about seven thousand annually, which means slightly increased incidence. The causes of this disease are not quite clear. What is known, however, are the risk factors. Primary prevention focuses mainly on the suppression or elimination of risk factors. Secondary prevention aims at diagnosing the cancer at an early clinical stage, which enables more efficient treatment. The first preventive mammographic screening was introduced in the Czech Republic in 2002. It is a preventive examination of mammary glands by means of Mammogram, which is covered by health insurance for all women over the age of 45 once in two years. The survey on the level of prevention of breast cancer in the region of Strakonice was conducted by means of a questionnaire which was distributed among the patients of general practitioners in the selected towns and districts. The objective of the paper was to find out whether the women in the Strakonice region are informed of the risk factors of the disease. Another topic addressed was the breast self-examination. Women under 45 depend purely on it, as they are not entitled to free screening covered by health insurance. Women have been found to carry out breast self-examination quite rarely, and therefore, it would be beneficial to familiarize these women with the self-examination technique via general practitioners, gynecologists, or through the media. Self-examination should be commonly conducted by every woman over 20. Another goal of the paper was to find out what experience women had with imaging screening technologies in mammography, especially whether women over 45 undergo regular preventive mammographic screening. Also, how accessible such an examination is in the Strakonice region. At present, there are only three mammography centers in the South Bohemia region, none of which is in Strakonice. It was found that 82% of respondents over 45 take part in the screening program, and 78% of them would appreciate if a mammography center were established in Strakonice.
