Agglomeration of animal feed mixtures

The theoretical part of this work focuses on the description of mechanical properties of compound feed produced in cooperation with Mendel University in Brno. These properties are mainly flow properties, which are crucial for the technological processes of these mixtures. The first part of this work also contains a general information about commonly used shear devices. Jenike's Shear Cell and Rheometer FT4 from company Freeman Technology are described in more detail. Because of the complexity of the behavior of these materials, their properties are mostly found the experimental way, which is the main topic of the practical part of this work. In this section you can find a description of measurements performed on Jenike's Shear Cell and on the Rheometer FT4. Moreover measurements of strength and length of granules were undertaken. Further the analysis of powder feed mixtures on sieves is described. Materials analyzed were compound feeds for chickens. Mutually they differed in the content of mealworms. Pellets were made from the mixtures and were then also subjected to measurement.

This thesis with title „Proposal of the Company's Financial Plan“ consists of theoretical and practical parts. Theoretical part deals with theoretical ways out of financial planning. In the practical part there are performed the analyses of contemporary state of company. Afterwards there is the financial plan alone made-up for the years 2009 – 2011. The end of my practical part is formed by evaluation of created financial plans.

This diploma thesis addresses the „University Campus Bohunice - Architectural and technological project“. This thesis includes excavation, pile foundation, reinforced concrete foundation slab it also deals with in detail technological regulations on earthworks and deep-pile foundation. The thesis contains solutions to building equipment, schedule of selected stages of the construction situation with the solution of transport routes, designed mechanical assembly required for selected stages and control plan earthworks and deep-pile foundation. This diploma thesis addresses the „University Campus Bohunice - Architectural and technological project“. This project involves construction of two buildings. Building A32 with two basement floors and three floors and second building A 31 with one basement floor and three floors. This thesis includes excavation, pile foundation, reinforced concrete foundation slab, it also deals with technological regulations of earthworks and deep-pile foundation in detail. The thesis contains solutions to construction site equipment, schedule of selected stages of the construction, location of buildings and the solution of transport routes, mechanical equipment required for selected stages of project and control plan of earthworks and deep-pile foundation.

Independent front axletree of the bus

In those diploma work was solved problems of independent trapezoidal axletree for low-floor bus - NB12 CITY firm SOR Libchavy s.r.o . Work was divided into two main features: solving wheel-suspension kinematics and strenght calculation sorted parts of wheel suspension by the help of finite element method. There was effected fatigue calculation sorted parts of wheel suspension according to „Resetova metoda“ of predicting fatigue damage flowing from so – called cutaway methodics of fatigue tests at the close of this work.
Structural analysis of the interaction interface between an antibody and a protein epitope

The diploma thesis deals with the study of the mouse monoclonal antibody EEV1-2.1, which was created in the RECAMO laboratories by immunization of mice with the Hsp90 protein and subsequent fusion of splenocytes with a mouse myeloma line. The theoretical part of this work focuses mainly on the study of antibodies, especially their structure, genetics and the description of monoclonal antibodies. In this part of the work, you can also find a cross-section of modern biotechnological trends that are currently used for the production of antibodies. CHO cells and tetracycline inducible systems are also described. The aim of the thesis was to characterize the mentioned antibody. Using bioinformatics methods and prediction software to predict and describe its structure, especially then to analyze hypervariable CDR regions. Another goal was to clone the light and heavy chains of the antibody using the Gateway method. The final step was the creation of a tetracycline inducible system for efficient production of the antibody itself. The experimental part of the work initially deals with the isolation of the RNA sequence of the antibody using commercial kits. Next, the method of amplification of the given sequence using PCR with reverse transcription is described. The following is a description of the Gateway cloning method, which allows convenient insertion of the selected gene into the target vector. The basic bioinformatics methods used to study the structure of antibodies are also explained, as well as the prediction of a more complex 3D structure of the antibody, including possible interactions with the HSP90 protein. This section describes the methods of transfection of ExpiCHO cells and inducible antibody production using the tetracycline inducible system. The thesis led to the successful cloning of the light and heavy chain of the antibody and the subsequent insertion of the expression vector into the production ExpiCHO cells. The expression system based on induction by the addition of doxycycline was then successfully tested by several methods and the results show that the inducible system is not only functional, but can even lead to increased production of monoclonal antibody compared to conventional methods.

Animal cells need to preserve constant volume in the face of osmolarity perturbations to function properly. To regain their original volume after hyposmotically induced swelling, most cell types extrude intracellular electrolytes and organic osmolytes accompanied by osmotically driven water. This process is termed regulatory volume decrease and is ensured by various ion channels and transporters. Recently, much attention has been focused on the ubiquitous volume-regulated anion channels activated by cell swelling. VRACs are moderately outwardly rectifying with intermediary conductance, permeable to inorganic anions and organic osmolytes and sensitive to broad-spectrum anion channels blockers. Functional properties of VRACs in astrocytes are particularly interesting, because many brain pathologies, such as ischemia, traumatic brain injury or hyponatremia, are associated with marked astrocytic swelling and VRACs could thus constitute a possible target for therapy of cerebral edema. Furthermore, VRACs are thought to play a role in cell cycle progression, cell migration, apoptosis and intercellular communication. Neurosteroid modulation of ligand-gated ion-channel activity

The term neurosteroids refers to steroids that are synthetized in the nervous tissue from cholesterol or steroidal precursors from peripheral sources. These compounds affect the neuronal excitability by modulating the function of some ligand-gated ion channels. NMDA (N methyl D aspartate) receptors are glutamate gated ion channels involved in excitatory synaptic transmission, synaptic plasticity and excitotoxicity. GABAA ( aminobutyric acid type A) receptors mediate most of the inhibitory synaptic transmission in the mammalian brain and are targeted by many clinically important drugs. Function of NMDA and GABAA receptors can by affected by neurosteroids, both positively and negatively. The aim of this work is to summarize the current knowledge about the neurosteroid effects on the function of GABAA a NMDA receptors and suggest the physiological role and the potential therapeutic use of the neurosteroids as a regulator of some functions of the central nervous system.
