Monitoring of leukemic cell line amino acid metabolism changes after Quambalarine B treatement

Leukemia is the most common cancer of children, moreover it is also not uncommon of elderly patients. Research has focused on the development of specific antileukemic drugs in recent years. Abnormalities in tumor cell metabolism that can be targeted during treatment appear to be the key. Natural 1,4-naphthoquinones, including quambalarin B produced as a secondary metabolite by the basidiomycetes of Quambalaria cyanescens, are known for their therapeutic effects. Not surprisingly, Quambalarine B has also been shown to inhibit cell proliferation in some leukemic cell lines and subsequently caused cell death. In the present thesis, I tried to observe changes in amino acid metabolism by monitoring amino acid levels in the intracellular and extracellular environment of leukemic cells after treatment with Quambalarine B using amino acid analysis with fluorescence detection. The observation was performed in Jurkat, Ramos and THP-1 cell lines, each of these lines represents another type of leukemic disease. The use of mass spectrometer for aminoacid analysis

Amino acid analysis is used across many disciplines. However, it carries certain problems such as missing asparagine, glutamine and tryptophan, due to the primary purpose for which it was developer. Nevertheless, there are many applications in which these missing amino acids need to be quantified as well. A faster but less sensitive alternative would be the mass spectrometry detection, which also makes it possible to analyze other metabolites and non-amino compounds. It is necessary to know all the possibilities and limits of the newly developed method to be applicable to the full benefit. This bachelor thesis involves both a method in which individual amino acids are separated using hydrophilic interaction chromatography and subsequently detected by mass spectrometry as well as its basic characterization and comparison with a commonly used fluorescence method commercially available from Waters.
Taste masking in orodispersible tablets. Evaluation of selected technologies.

PharmDr. Zdeňka Šklubalová, Ph.D. Student: Zuzana Matoušková Title of Thesis: Taste masking in orodispersible tablets. Evaluation of selected technologies. Orodispersible tablets (ODT) represent a modern trend in solid dosage forms. This Master Thesis is focused on assessment of the influence of excipients on properties of orodispersible tablets as well as on masking the taste of active ingredient vardenafil-hydrochlorid trihydrat. The tablet blends containing crospovidone as a disintegrating agent were prepared. The impact of hypromellose, sweeteners and flavours on the properties of tablets fabricated by the direct compression or by wet granulation was studied. Out of the examined combinations of components the formulation consisting of hypromellose, sweeteners and mint flavor had the best taste. The tablets prepared by wet granulation had longer disintegration time compared with the tablets prepared by the direct compression.

The aim of my Bachelor's thesis is to analyze the current marketing mix of the company which is focused on the service sector. Further I am going to process a SWOT analysis through which I will evaluate the internal and external situation of the company. I will also compare the SWOT analysis created by me with analysis processed by management of the company or by other external party. This comparison will enable the company management to look at the company from more angles. After a detailed analyzing of all parts of the analysis I will suggest certain recommendations for change in the marketing mix. The management and a key employees of the company will select an innovative measure from these proposals. This measure will be implemented.
Financial statement of the selected operator to focus on finding profit and optimization of the income tax

The main target of my diploma work is the evaluation of the financial statements course and the statements in a certain company, the calculation of the income tax and the draft tax strategy for the business entity. At the beginning of the practical part I describe the individual ithems of the financial statements for the years 2010, 2011 and 2012. The following is findings profit, its subsequent transformation and the calculation of taxes of corporate income. The last part of the practical part is tax optimalization and design strategy. I suggested 3 options: tax optimalization by way of depreciation, the donations to good causes and employment of workers with reduced working capacity.
