Lectin-like receptors can modulate the cytotoxic, regulatory and dendritic cell responses by their interactions. GN8 multivalent glycoconjugates are able to internalize inside and activate NKR-P1 positive cells inducing production of IFN-gamma and expression of CD69. The final effect is a modulation of anticancer immunity with reduction of cancer growth

Environmental conditions cooperate to colorectal carcinogenesis. In an experimental model in the rat was evaluated the impact of the commensal intestinal flora on cancer development, and consequent general variations of the immunity, versus germ-free (GF) conditions. Less and smaller tumors developed in GF rats and a better developed immune response appeared to be able to be elicited in absence of intestinal bacteria

During carcinogenesis and cancer development tissues undergo to important modifications with creation of a specific cancer microenvironment. Locally, the immune system is challenged by tumor-secreted active biological molecules (eg. cytokines, growth factors, NO, lactic acid) that can impair the activation of an effective anticancer response. The use of glycodendrimers targeting immune cells of innate immunity can rescue the activation and stimulate the immune responses, operating through lectin-like receptors.

Masking oligosaccharide sequences can assist transformed cells to escape immune surveillance. N-acetyl-glucosamine coated dendrimers have binding affinity for NKR-P1 receptor and can also modify cancer cell glycosylation profile. Effects of treatment on immune cell subpopulations were analyzed in blood, lymphoid tissues and organs, and inside tumor in B16F10 melanoma bearing mice. Treatment enhanced tumor cell immunogenicity, and induced maturation,differentiation and homing of cytotoxic cells to tumor microenvironment.
