Molecular mechanisms and components controlling the Wnt signaling pathway output

Beyond its essential roles in embryonic development, the Wnt-mediated signal transduction cascade is critically implicated in homeostasis of adult tissues. In the gastrointestinal epithelium, the threshold of active Wnt signaling is kept in a physiological range by a spectrum of regulatory networks and loops, thereby balancing the opposing processes of cell fate determination, proliferation and stem cell self-renewal. Furthermore, compelling evidence undoubtedly link an aberrant Wnt activity to the onset of bowel cancer. Understanding the principle causes and effects secondary to excessive Wnt signaling can provide valuable insights into the pathology of the malignant transformation of the colorectum. The proposed thesis attempts to focus on novel modes of the Wnt pathway modulation; both general and context-specific nuances of the Wnt level adjustment are thereby delineated. The results are presented in three distinct research publications and one review article. Depiction of society in historical television series: Bridgerton versus The Knick

The diploma thesis examines the depiction of social classes, men, women and ethnic minorities in historical television serials. It is concentrated specifically on a qualitative analysis and comparison of two serials, namely Bridgerton (2020) and The Knick (2014- 2015), of which both fall within the quality television genre. The thesis deals with the ways of representation of the societies of their respective periods, aiming especially at identifying the incorrect depiction of the historical period in question, if any, and at finding similarities with the problems concerning the today's society (such as addressing the issues of social inequalities and manifestations of the identity policy). The theoretical part of the diploma thesis describes the phenomenon of a television serial, namely that concerning the historical serial and quality TV genres, the latter being the kind of television serial work distinguishing itself by comprehensive and innovative procedures, elaborate narratives, and emphasis put on the development of characters. Attention is also paid to the field of the cultural studies, mainly the hegemony theory and the concept of ideology, media and their representation, as well as the study of media audience. Functional role of Islet1 in pancreatic development

1 Abstract Diabetes mellitus is characterized by the dysfunction and reduction of insulin-producing cells, resulting in hyperglycemia, which in long term harms the organism. For future therapy, it is crucial to understand the function of various factors participating in the differentiation and maturation of endocrine pancreatic cells. The aim of this study was to unravel the functional role of ISL1 during the development of the pancreas. ISL1 is expressed in all endocrine cells of the islets of Langerhansbut its function remains unclear, especially during early pancreatogenesis. As the global deletion of this gene is embryonically lethal, we used the tissue specific deletion of Isl1 in Neurod1 possitive cells using the Cre-loxP system. In this work we studied the effect of this deletion on the structure of islets of Langerhans, the formation of endocrine cell types and relative expression of genes during early pancreatic development. A defective achitecture of islets together with postnatal absence of α-cells was found in the Isl1 deletion mutant. Also, the expression of genes important for the specification of α-cell lineage and their subsequent function was decreased. Corporate Culture and Personnel Activities in a Selected Business

The topic of this diploma thesis is Corporate Culture and Personnel Activities in a Selected Business. The aim of the thesis is to characterize the corporate culture with the help of specification of its crucial cultural dimensions in connection with the personal activities and proposing changes leading to the desired state in the selected business. The research was carried out at the company of an engineer Jan Hutkay. The thesis is divided into four main parts - literary research, aim and methodology of work, characteristics of the current state in the business and discussion, summarized evaluation and proposals for changes. Literary research is focused on explanation of important terms related to corporate culture and personnel activities. It is followed by the summarization of the aim of the work and the research methods. Quantitative research methods used in the work included the questionnaire survey in the form of the Personnel Activity Questionnaire and the VSM 94 a 2013 tests. Among the qualitative methods used for the purposes of the thesis belong observations, unstructured interviews and Color Semantic Differential Tests were used from qualitative methods. The current situation in the company was characterized by the analysis of the individual questionnaires, which was supplemented with the information obtained through interviews and observations. After a thorough analysis and interconnection of the results of all research methods, it has been found out that corporate culture tends to deserve the dimensions of low power distance, femininity, collectivism, and acceptance of risks and changes. The undesirable resultant dimensions include the short-term business orientation and the dimension of indulgence. The final part of the thesis focuses on designing measures that stabilize the desired cultural dimensions and, above all, change the undesirable cultural dimensions. The results of the personnel activities' questionnaire which identified the main shortcomings and problems were used to suggest specific changes. The introduction of the proposed changes directs the enterprise closer to the knowledge economy, which improves its performance and competitiveness.

Protection of economic competition - block exemptions

1 Abstract Block exemptions are very specific institute of competition law but although they have existed in European law for many decades, their application is not well-known in public. Certain activities in an economic competition were generally considered distorting the free competition and thus being undesirable so the EU institutions decided to prohibit them unless otherwise provided. The cartels and state aid generally bring imbalance and unfair conditions in the internal market, but there are certain circumstances which justify their usage and allowance in some cases. The actions, which are allowed due to their specific circumstances and under certain conditions, are provided in the form of so called block exemptions. Block exemptions are issued in two areas of competition law, agreements between undertakings and state aid. Agreements between undertakings, i.e. cartels, which affect the business in the internal market, are prohibited by Art. 101 (1) TFEU and the block exemptions determine the categories of cartels which can be allowed. On the other side, the state aid is only considered as incompatible with the internal market in Art. 107 (1) TFEU. Phosphoinositides and their effectors in regulation of the Wnt signalling pathway

Phosphoinositides (PIs) make up only a small proportion of overall amount of lipids in cell membranes. However, their function mediated through protein effectors is indispensable for cell signaling, vesicular trafficking, cell movement and other important aspects of cellular life. In this bachelor thesis function of PIs is described in relation to Wnt signaling pathway. Proper execution of several steps of the Wnt signaling pathway requires the presence of PIs. Retrograde transport of Wntless (Wls) from the plasma membrane (PM) back to the Golgi apparatus (GA) in Wnt producing cells or internalization of Wnt receptors in Wnt receiving cells are only two examples. All processes are tightly regulated and malfunction of enzymes processing PIs can cause their deregulation resulting in disruption of the Wnt signaling pathway. As deregulated Wnt signaling is a known cause of serious diseases including cancer, understanding the crosstalk between PIs and Wnt signaling could help in designing novel strategies for therapeutic intervention.
