Design of the off-road vehicle

The main goal of this diploma thesis is to create a design of an off-road vehicle. The vehicle has become a concept vision of an ecological off-road of the future fueled by hydrogen fuel cells. The secondary goal is: to bring new innovative form elements into conventional design solution of off-road vehicles and to highlight the problem with ecology, environmental protection and limited resources of fossil fuels.

OFDM implementation in FPGA

The thesis is focused on designing OFDM modem, which should be implemented into the FPGA device. The advantages of using OFDM signals in order to provide high baud rates together with high multipath immunity has provoked a mass expansion into media systems such as DSL, DVB, Wi-Fi, WLAN, etc. . Thanks to this technology we can quarantee high modulation rates with minimal negative disturbance eects. The rst part is dedicated to characterise OFDM signals, their generation and the algorithm producing the OFDM which is implemented in DSP devices. For the purpose of using the fastest algorithm, the Fast Fourier Transform using Cooley-Tukey algorithm was shown. Before we can implement OFDM modem into the FPGA device, we have to simulate its correct function. Because there is no OFDM analyser available at our departement, its necessary to prove its correct function by simulations. Therefore a large part of this thesis is focused on simulations using Matlab and ModelSim, in order to show comparison between the theoretical, and simulated results. Between the theoretical and practical simulations there is a part which shows the brief characteristics of available FPGA devices. Detailed view is presented just for the Virtex II device, which the implementation is made for. As a suitable FPGA device, we have chosen Virtex II XC2V1000 which is available for students. In the last part the measured results were shown to prove the corect function of the modem. Programming the FPGA using VHDL language is realized in the software ISE Xilinx (distributed in Xilinx software support). All programmed scrits and data used in this thesis are included on distributed media.

These motivation instruments could help cut down stress and improve communication in the company. The aim of this thesis is on the basis of analysis current status to purpose concrete motivation instruments, which will increase job performance of employees.

Tato diplomová práce se zabývá detekcí prvků uživatelského rozhraní na obrázku displejetiskárny za použití konvolučních neuronových sítí. V teoretické části je provedena rešeršesoučasně používaných architektur pro detekci objektů. V praktické čísti je probrána tvorbagalerie, učení a vyhodnocování vybraných modelů za použití Tensorflow ObjectDetectionAPI. Závěr práce pojednává o vhodnosti vycvičených modelů pro zadaný úkol.
Receiver for remote control

This semestral project deals with the plan of peripheral involvement of the receiver remote controller, which will serve as a control module for controlling the turning antenna rotator. Part of the work is focused on the issues of wireless remote controllers, the modulation method used for transmitted order and transmission protocols. Then there is solved the problem of the right choice microcontroller for controling the entire application, including a description of its basic properties. The work also solves the overall interconnection of control units, power supplies and antenna rotator itself.
Role of glycosylation of ionotropic glutamate receptors in mammalian neurons

Glutamate is the most abundant excitatory neurotransmitter in the mammalian central nervous system. There are two distinct types of glutamate receptors, ionotropic and metabotropic, present in the mammalian excitatory synapses. My thesis is focused on the ionotropic glutamate receptors, which play critical roles in learning and memory formation. The main subtypes of ionotropic glutamate receptors are α-amino-3-hydroxy-5-methyl-4- isoxazolepropionic acid (AMPA), N-methyl-D-aspartate (NMDA) and kainate receptors. All types of the ionotropic glutamate receptors, which are assembled as tetramers, contain many glycosylation sites, which can be modified by glycans or monosaccharides. The glycans and monosaccharides attached to the ionotropic glutamate receptors have been shown to regulate key processes such as folding of the subunits, transport to the cell surface as well as their functional properties. Recent literature also suggests that many neurological and psychiatric disorders such as schizophrenia exhibit abnormal glycosylation of ionotropic glutamate receptors. Structural determinants of regulation of surface delivery of NMDA receptors in mammalian cells

N-methyl-D-aspartate (NMDA) receptors are ligand-gated ion channels activated by agonist glutamate and co-agonist glycine. They play a key role in mediating the fast excitatory synaptic neurotransmission in the mammalian central nervous system. To create a functional heterotetrameric receptor, the presence of two GluN1 subunits combined with GluN2 or GluN3 subunits is necessary. Previous studies confirmed the importance of M3 transmembrane helix and extracellularly localized cysteines in regulation of surface expression of functional NMDA receptors. The aim of my thesis is to elucidate an influence of clinically relevant mutations in M3 transmembrane helix and the role of all known cysteines that form disulphide bonds on surface delivery of NMDA receptor expressed in heterologous monkey kidney fibroblasts cell culture (COS-7). Using molecular biology methods, immunocytochemistry and microscopy I found that the clinically relevant mutations M641I and Y647S in GluN1 subunit and also the mutations of particular cysteines forming disulphide bonds caused substantial decrease of surface expression of NMDA receptors. Functional and pharmacological properties of GluN1/GluN2 and GluN1/GluN3 subtypes of NMDA receptors

(EN) N-methyl-D-aspartate receptors (NMDARs) are ionotropic glutamate receptors and they play a critical role in excitatory synaptic transmission in the mammalian central nervous system (CNS). Hyperactivity or hypoactivity of NMDARs can lead to a wide spectrum of pathological conditions and psychiatric disorders, such as Alzheimer's disease, Parkinson's disease, Huntington's disease, epilepsy, schizophrenia. NMDARs form a heterotetrameric complex made up of GluN1, GluN2(A-D) and/or GluN3(A, B) subunits. Different subtypes of NMDARs could have various effects on disease pathogenesis and therefore it is crucial to investigate the specific role of each subunit in the regulation of normal NMDAR functioning. The regulation of NMDARs occurs at different levels, from early processing, including synthesis, assembly, quality control in the endoplasmic reticulum (ER), trafficking to the cell surface, to internalization, recycling, and degradation. In this dissertation, we mainly focused on determining the roles of extracellular and transmembrane regions of different subtypes of NMDARs in the regulation of their function. The role of RNA demethylase FTO in differentiation, regulation of energy metabolism and sensitivity to streptozotocin of neuronal and glial cell models

Fat mass and obesity associated (FTO) demethylase is responsible for erasure of the most abundant epitranscriptomic mark in eukaryotic mRNA, the N6-methyladenosine (m6A) residue. Together with other m6A erasers, writers (methyltransferases) and readers it forms an m6A regulatory pathway that controls the amount, location and biological effect of m6A. The dynamic regulation of the brain's m6A methylome during neurodevelopment is essential for maintaining cerebral functions. In addition, preclinical research suggests that the m6A regulatory pathway regulates energy balance in a tissue- and cell type-specific manner. The FTO gene has been associated with lifelong risks of obesity and metabolic syndrome as well as regulation of total body energy intake and expenditure. However, little is understood about the function of the m6A pathway in control of brain energy metabolism. That is of interest in pursuit of understanding Alzheimer's disease, as this illness is characterized by profound disruptions in cerebral energy metabolism and mounting evidence suggests that disrupted brain bioenergetics may play a role in the disease's early genesis, before the appearance of clinical symptoms.
