Invazivní fungální infekce jsou velký problém. V jejich důsledku umírá po celém světě přibližně 1,5 milionu lidí ročně. Mezi nejčastější invazivní fungální infekce patří aspergilóza a kandidóza. Tyto oportunistické infekce mají vysoký podíl mortalit (50 % a 27 %). Diagnostika těchto infekcí bývá složitá. To často vede k jejich pozdějšímu odhalení a k většímu riziku vzniku komplikací. Pro diagnostiku a monitorování léčby fungálních infekcí se v poslední době zkoumají tzv. hypoxické sondy. Tyto sondy umožňují neinvazivní měření koncentrace kyslíku ve tkáních a mohou vést k časné diagnóze fungálních infekcí. Hypoxie může být také relevantním cílem pro doručení antifungálních látek. Z těchto důvodu byla připravena série hypoxicky-selektivních sond (prób). Sondy obsahují N-methyl-2-nitroimidazol, a fluorofor, který absorbuje v oblasti blízkého infračerveného spektra. Podle teorie mohou sondy fungovat následujícím způsobem: N-methyl-2-nitroimidazol, který se reduktivně rozkládá v hypoxickém prostředí, aktivuje sondu. Ta vyloučí fluorofor, který absorbuje v blízkém infračerveném spektru. Pomocí konfokální mikroskopie se bude sledovat, zda-li se fluorofor uvolnil selektivně v hypoxickém prostředí resp. v místě infekce. Among the most common invasive fungal infections are aspergillosis and candidiasis. Both opportunistic infections have a high mortality rate (50% and 27%, respectively). The diagnosis of these infections is often complex, leading to delayed detection and an increased risk of complications. Recently, near-infrared (NIR) hypoxia probes have been investigated for the diagnosis and monitoring of fungal infections. These probes enable non-invasive measurement of tissue oxygen concentration and can contribute to early diagnosis of fungal infections. Hypoxia can also be a relevant target for the delivery of antifungal agents. For this reason, a series of hypoxia-selective probes has been developed. These probes contain N-methyl-2-nitroimidazole and a fluorophore that absorbs in the near-infrared spectrum. We anticipate that probes will function as follows: the reducible breakdown of N- methyl-2-nitroimidazole in a hypoxic environment will activate the probe, leading to the release of the fluorophore, which absorbs in the near-infrared spectrum.
