Úvod: Imunochemické metody jsou důležitým pomocníkem klinických lékařů i odborníků v biomedicínském výzkumu. Předností imunochemických metod je jejich vysoká citlivost a přesnost. Cílem práce bylo využít imunochemických metod ke stanovení biomarkerů nádorových a degenerativních onemocnění. Byly provedeny celkem čtyři studie. Dvě studie se zabývaly hledáním nových biomarkerů karcinomu prostaty, třetí studie se zabývala sledováním FGF23 u pacientů s hyperparathyreózou a čtvrtá studie se zabývala stanovením kyseliny hyaluronové u pacientů s gonartrózou kolene. Metody a pacienti: 1. studie: Koncentrace Chromograninu A, Endoglinu, TIMP-1 a thymidinkinázy 1 (TK1) v séru a Endoglinu, SPINK-1, Annexinu, TIMP-1 v moči byly měřeny u 58 pacientů (průměrný věk 68 let, rozmezí 45 -82 let) s klinicky diagnostikovaným karcinomem prostaty a 30 zdravých jedinců (průměrný věk 64 let, rozmezí 55 - 78 let) bez relevantní urologické anamnézy a hodnoty PSA v normě. TK1 byla vybrána jako nejvhodnější marker a sérové koncentrace TK1 byly následně měřeny u 169 pacientů (průměrný věk 62 let, rozmezí 45 - 82 let) s karcinomem prostaty a kontrolní skupiny 39 zdravých mužů (průměrný věk 61 let, rozmezí 54 - 78 let). Vyšetření TK1 bylo rozšířeno o stanovení epitopu TK210. Sérové koncentrace TK210 byly měřeny u 126 pacientů... Introduction: Immunochemical methods have significant importance in biomedical research. The benefit of immunochemical methods is their high sensitivity and accuracy. The aim of the work was to use immunochemical methods in investigation of cancer biomarkers and degenerative diseases. A total of four studies were performed. Two studies were focused on the search for new biomarkers of prostate cancer, the third study was focused on evaluation of FGF23 in patients with hyperparathyroidism, and the fourth study looked at the assessment of hyaluronic acid in synovial fluid of patients with knee gonarthrosis. Methods and patients: Study 1: Concentrations of Chromogranin A, Endoglin, TIMP-1 and thymidine kinase 1 (TK1) in serum and Endoglin, SPINK-1, Annexin, TIMP-1 in urine were measured in 58 patients with the (mean age 68 years, range 45 - 82 years) with clinically diagnosed prostate cancer and in 30 healthy individuals with the (mean age 64 years, range: 55 - 78 years) without clinically relevant urological history and PSA values in the reference range.
