Úvod Metoda MALDI-TOF hmotnostní spektrometrie je relativně nová metoda identifikace mikroorganismů, založená na analýze části buněčného proteomu. V posledních letech prodělala prudký rozvoj a postupně doplnila a pak i částečně nahradila do té doby dlouhodobě ověřené rutinní biochemické metody. Stala se tak nedílnou součástí mnoha mikrobiologických laboratoří a s jejím rozšířením souvisí i včasné nasazení efektivní terapie a ve výsledku zkrácení doby léčby pacienta. V těch případech, kdy potřebujeme detailnější charakterizaci mikroorganismů, můžeme dnes využít celé spektrum dalších, především molekulárně- biologických metod, polymerázovou řetězovou reakcí (PCR) počínaje, přes RFLP (polymorfismus délky restrikčních fragmentů), sekvenační analýzu amplikonů části genu pro 16S rRNA (16S rRNA), až po celogenomovou sekvenaci. Cíl práce Pomocí metody MALDI-TOF MS i jiných metod (PCR, RFLP, rozšířená biochemická identifikace) charakterizovat a/nebo reidentifikovat vybrané čtyři skupiny infekčních agens (Campylobacter spp., Plesiomonas shigelloides, Citrobacter spp., Staphylococcus intermedius/Staphylococcus pseudintermedius). Zvolená agens byla klasickými metodami identifikace (morfologie, mikroskopie, biochemické testy) testována v letech 1950-2015; hlavním cílem disertační práce bylo porovnat efektivitu původních... Background MALDI-TOF MS (Matrix-Assisted Laser Desorption Ionization Time-of-Flight Mass Spectrometry) method for identification of microorganisms appeared not long ago. It is based on analysis of cell proteome. The method has undergone rapid development recently and it gradually complemented or even substituted classical routine biochemical methods. It became highly important part of microbiological laboratories. An increase in its use contributed considerably to starting of effective therapies timely and resulted in shortening of time needed for medical care. Numerous laboratory techniques, that are available nowadays, can be applied for more detailed characterization of isolated microbes, when it is necessary. The molecular-biological methods are of the most importance, namely polymerase chain reaction (PCR), restriction fragment length polymorphism (RFLP), 16S rRNA gene sequence analysis or whole gene sequencing (WGS) analysis.
