Professor Einstein byl na základě získaných výsledků screeningových studií vybrán pro podrobnou fytochemickou práci za účelem co nejširšího spektra AmA. Z 34,3 kg čerstvých cibulí bylo získáno 31,7 g přečištěného alkaloidního extraktu, který byl rozdělen pomocí sloupcové chromatografie se stupňovitou elucí benzinem, chloroformem a ethanolem v různých poměrech na téměř 500 frakcí. Tyto frakce byly spojeny do 27 podfrakcí, z nichž bylo pomocí preparativní TLC, vakuové sloupcové chromatografie a krystalizace izolováno 25 alkaloidů v čistém stavu. Všechny látky byly identifikovány pomocí GC-MS, ESI-MS, NMR, optické otáčivosti a literatury a jedna z nich byla identifikována jako nový nepopsaný alkaloid lykorinového strukturního typu. Všechny alkaloidy izolované v dostatečném množství byly podrobeny testům jejich biologické aktivity spojené s Alzheimerovou chorobou (inhibice hAChE, hBuChE, POP,... Professor Einstein was chosen based on results of previous screening studies for detailed phytochemical work for the purpose of isolation of the widest range of AmA. From 34,3 kg of fresh bulbs was obtained 31,7 g of purified alkaloidal extract, which was processed using column chromatography with stepwise elution by light petrol, chloroform and ethanol in different ratios to almost 500 fractions. These fractions were fused into 27 subfractions, which were processed by preparative TLC, vacuum column chromatography and crystallization. Finally, 25 pure alkaloids were isolated. All compounds were identified by GC-MS, ESI-MS, NMR, optical rotation and literature. One compound was identified as a new unpublished alkaloid of lycorine structure type.
