Pesticidy jsou sloučeniny určené k hubení, prevenci a kontrole škůdců a ochraně rostlin. Jejich široké použití v zemědělství způsobuje kontaminaci podzemních, povrchových i pitných vod, protože je čistírny odpadních vod nedokážou odstranit. Mezi jedny z nejvíce používaných pesticidů patří triazinové pesticidy. Jejich používání je ale celosvětově omezováno, z důvodu negativního vlivu na lidské zdraví i kontaminaci životního prostředí a to zvláště jejich metabolity, o jejichž toxicitě nejsou zatím žádné informace. V této diplomové práci byla vyvinuta a validována metoda pro stanovení triazinových pesticidů atrazinu, cyanazinu, propazinu, simazinu a terbutylazinu a jejich metabolitů hydroxyatrazinu, desethylatrazinu, desisopropylatrazinu, hydroxyterbutylazinu a desethylterbutylazinu ve vodách pomocí LC-MS/MS s ionizačním zdrojem elektrosprejem. LOD byl v rozsahu 0,5-3,2 pg/ml, LOQ v rozsahu 1,5-9,6 pg/ml. V této práci byl proveden test stability mateřských látek těchto pesticidů, kde se ukázala mít největší vliv na jejich degradaci teplota. Provedený monitoring výskytu triazinových pesticidů a jejich metabolitů ve vodách v ČR odhalil nízké koncentrace mateřských látek v povrchových i podzemích vodách i přes jejich zákaz používání v letech 2004-2008. Byly také nalezeny vysoké koncentrace... Pesticides are the substances designed for destroying, preventing and controlling of pests and protection of plants. Their wide usage in agriculture causes contamination of ground, surface and drinking water, because wastewater treatment plants are not able to remove them. Mostly used pesticides are triazine based pesticides. However their usage is worldwide reduced because of their negative influences on human health and contamination of environment especially with their metabolites, about their toxicity are no information. A method for determination of triazine pesticides atrazine, cyanazine, propazine, simazine, terbuthylazine and their metabolites hydroxytrazine, desethylatrazine, desisopropylatrazine, hydroxyterbuthylazine a desethylterbuthylazine was developed and validated in this diploma work. The determination was realized by LC-MS/MS with electrospray ionization. LOD ranged between 0.5-3.2 pg/ml and LOQ between 1.5-9.6 pg/ml. A test of stability of the maternal substances of these pesticides was performed, in which temperature was found to have the biggest influence on their degradation.
