Sledovanými látkami v této diplomové práci jsou ženské pohlavní hormony, jejichž prob- lematika v environmentální analýze nabývá na významu. Vliv hormonálně aktivních látek na životní prostřední a člověka má rostoucí tendenci. Již v 80. letech 20. století byly pozorovány deformity na rybách vyskytujících se v řekách. Mezi sloučeniny vykazující tyto účinky patří kromě steroidních hormonů také pesticidy, tenzidy a plastifikátory. Estrogeny, jak přirozené tak syntetické, jsou sloučeniny s vysokou hormonální aktivi- tou. Tyto sloučeniny zapříčiňují rakovinu prsu, abnormality v oblasti pohlavních orgánů a snížení početu spermií. Jsou také příčinou feminizace, hermafroditizmu a klesající plod- nosti u zvířat. Stoupající riziko znečištění vod nutí analytické laboratoře vyvíjet nejdokon- alajší instrumentální metody ke stanovení steroidních látek. Vliv estrogenů na životní prostředí byl pozorován již při koncentracích v pg/ml. Tato práce se zabývá vývojem metody pro kvalitativní a kvantitativní analýzu deseti látek steroidní povahy ze skupiny ženských pohlavních hormonů pomocí ultravysokoúčinné kapalinové chromatografie ve spojení s hmotnostním spektrometrem typu trojitého kva- drupólu s možností tandemové hmotnostní detekce (UHPLC-MS/MS). Separace byla provedena na koloně BEH Phenyl. Jako mobilní fáze byl... In this study a new analytical method for determination of endocrine disruptors is pre- sented. The endocrine-disrupting phenomena is an area of growing concern. Potential ef- fects of endocrine-disrupting pollutants on wildlife and humans has been increased. Since 1980s deformities in fish are observed. Compounds associated to these effects include nat- ural compounds, such es estrogens, progestins and phytoestrogens, synthetic estrogens and a wide variety of organic pollutants, inluding pesticides, surfactants and plasticizers. Among them, estrogens are extraordinary due to their high estrogenic potency, with phys- iological active concentrations ranging from fg/ml to pg/ml. Endocrine disruptors might cause breast cancer in woman, abnormalities in the human reproductive system and differ- ent expressions of biological disorders in wildlife such as hermaphroditism, feminization, and decreased fertility. The presence of numerous endocrine-disrupting compounds in sur- face waters and sediment has been primarily atributed to their incomplete removal in the sewage-treatment process. Determination of these chemicals is required in order to assess their environmental impact.
