Unravelling molecular mechanisms of the abasic crosslink DNA repair

4 Abstract DNA as the primary carrier of genetic information guarantees organisms to live, grow, develop and reproduce. However, this most vital molecule in the cell is subject to various damages every moment. If it is not repaired, the cell and the organism will eventually succumb to inevitable destruction. One of the most serious damages is abasic site interstrand crosslink (Ap-ICL). Ap-ICL is formed spontaneously when an abasic site covalently pairs with an adenine on the opposite strand. The lack of information on repair mechanisms, the influence of the local sequence and its stability leads to questions about the fate, toxicity and occurrence of these lesions in cells. During evolution, several mechanisms have evolved to repair these and other damages to ensure the organism's survival. A recently discovered pathway known to repair Ap-ICL is named after the DNA glycosylase responsible for removing Ap-ICL. NEIL3 DNA glycosylase is recruited to Ap-ICL by ubiquitylation of DNA helicase, which is part of the DNA replication complex. NEIL3 glycosylase contains several zinc-finger domains, that bind to the damaged DNA and ensure its catalytic function. The subject of this thesis is the diagnostic investigation of block A in the object at Rumiště 8, Brno. The work includes theoretical knowledge of technical surveys and diagnostic methods and their evaluation to identify the essential characteristics of masonry and timber structures. In the practical part is the diagnostic investigation processed. Part of the diagnostic investigation is the determination of material characteristics of the structure. At the end of the thesis is an assessment of the structure.

This master’s thesis provides us into the issues of reduction of house prices due to roof structure defect. First charter takes interest in theorethical solutions for residential houses valuation with defects.Real estate survey is introduced in chapter two. The third part of this master’s thesis is focused on possible roof structure defects and malfunctions. In last charter we can find the valuation of house with defect. The main goal of this thesis is to create a procedure for the reduction of property price with roof structure defect.
Survey and Diagnostics of Building Construction

The subject of this diploma thesis is the diagnostic survey of the masonry walls and the connecting bridge in the East Moat of Špilberk Castle. The thesis contains theoretical information about survey and diagnostics of building construction and then evaluation of diagnostic methods used for estimation of essential characteristics of masonry structures. In the practical part the diagnostic investigation of Špilberk Castle walls is processed. That includes determination of material characteristics of the studied structure. In the conclusion there is a suggestion of how to eliminate the structural defects found during the survey.

The Energy of a Flying Object Necessary for Damaging the Vehicle Windscreen

The theoretical part of the thesis entitled "The Required Energy of a Flying Object to Damage a Windscreen" covers the historical development of glass as a raw material and its further use as a key material for the automotive industry. In the following chapters, the thesis provides an analysis of a windscreen in general including the material composition of various windscreen manufacturers, the distribution according to the accessories used in the windscreens, and the various options of placing a windscreen in a car bodywork. The homologation of a windscreen and the procedure for replacing or repairing a windscreen after a damage is also covered. Lastly, the experimental part of the thesis describes the process of testing the required energy of a flying object to damage a windscreen.
Rehabilitation of sewer connections and manholes on the sewer network

Bachelor`s thesis makes a summary of today`s state of methods designed for rehabilitation of sewer manholes and laterals. The work is then comparing them and provides examples of specific technologies for each method. There are recommendations for system operatives at the ends of corresponding chapters. Prediction of the future development of this sphere of civil engineering is situated at the end of theoretical part of bachelor`s work. Practical part of bachelor’s thesis is dealing with the analysis of defects of an existing sewer lateral and three manholes, with subsequent commentary of suitability for each rehabilitation method. The result is choosing the most suitable method for rehabilitation of sewer lateral and manholes.
Suggestions for Improvement in Long - Termed Material Property Economy

This thesis is focused on analysis of production facilities utilization and maintenance in the company RAMET C.H.M. a.s. located in Kunovice. In this thesis, there is performed the analysis of present property condition, machinery and equipment used within radar antenna production. Within property maintenance, there are introduced planned and unplanned processes of servicing activities and machine and equipment repair. In a part of proposal for solution, there are recommended processes for keeping up production capabilities based on extensive analysis of size property influence on production volume. The method for reliability estimation of production chain is described for the antenna production, related to limiting production capability and costs to maintain service availability of production facilities.

This dissertation solves the problems with management of capital goods in the company FORMIRA, s.r.o. The dissertation is elaborated from the analysis of present situation in the plant, whereas the heed is oriented to the most important areas, especially the organization and service activity and maintenance of assets. The result of performed analysis is the identification weak points. Proposals are submitted in the second part of the dissertation, in order to solve the problems found by presented analysis. The economical evaluation of the proposals and determinations of assumptions for the application of extended results to the company use is performed at the end of the dissertation.
