In vitro and in vivo studies of the neuroprotective peptide humanin using 125I-radiolabeled humanin derivatives

Four new HN-derivatives were synthesized: Y-PAGASRLLLTGEIDLP (Y-des-Leu-RG-PAGA, 1), Y-PAGASRLLLLTGEIDLP (Y-RG-PAGA, 2), Y-SALLRSIPAPAGASRLLLTGEIDLP (Y-des-Leu-Colivelin, 3), Y-SALLRSIPAPAGASRLLLLTGEIDLP (Y-Colivelin, 4), their neuroprotective action was evaluated in the T-maze test and the most active ones were monoradioiodinated with 125I and the labelled peptides used for binding and biodistribution studies.
Specific binding of prolactin-releasing peptide analogues in pituitary cells

We report specific binding of 125I-PrRP31 to the following cell lines: GH3 cells (rat – somatotrophs), AtT20 cells (mouse – adenocorticotrophs) and RC-4B/C cells (rat- somatotrophs, lactrophs, adenocortitrophs and gonadotrophs). These results will serve as a basic knowledge for future structure-activity studie sof PrRP.
Labeling of peptides by Na125I-IODO-GENtm system

Very high excess of IODO-GEN(tm) reagent (115 nmol in commercially available precoated tubes from Pierce) leads to damage of oxidation sensitive peptides containing tryptophan residues. Using 5 to 23 nmol of IODO-GEN(tm) in a reaction with 1 to 4 mCi of Na[125-I] and 10 to 30 nmol of peptide leads to reproducible results and reproducible yields.
