Abstract: Very high excess of IODO-GEN(tm) reagent (115 nmol in commercially available precoated tubes from Pierce) leads to damage of oxidation sensitive peptides containing tryptophan residues. Using 5 to 23 nmol of IODO-GEN(tm) in a reaction with 1 to 4 mCi of Na[125-I] and 10 to 30 nmol of peptide leads to reproducible results and reproducible yields.
