Use of interspecific ICSI method for assessing the quality of the retained biological material from genetic resources
Vacková, Kristýna ; Benc, Michal (advisor) ; Barnetová, Irena (referee)
Intracytoplasmic sperm injection, or ICSI, is a commonly used method in assisted reproduction centers and is also an effective tool for a fertilization research. The interspecific ICSI is the suitable method for production of zygotes, which are formed by association of gametes of different, distantly unrelated species. It can bypass natural interspecies barriers and create so-called interspecific zygotes. This can be used to evaluation properties of genetic material. In this work, this method was used for evaluation of the quality of stored sperm originating from male goats of the white shorthair goat genetic resource. The aim of this work was to evaluate the effect of cryopreservation on the quality of goat sperm, which was frozen in three different ways at two different breeding stations. Freshly collected semen served as a control group. The heads of xenogeneic goat sperm were injected into mature mouse oocytes. The injected oocytes were allowed to develop to the zygote stage, in which both female (mouse) and male (goat) pronuclei was formed. In male pronuclei, DNA damage was assessed by immunofluorescence staining with anti-γH2AX antibody, a marker of DNA breaks. The results obtained by measuring the level of γH2AX fluorescence intensity in the paternal pronuclei show that the values differ...
Differentiation potential of human dental pulp mesenchymal stem cells
Vacková, Kristýna ; Voltrová, Barbora (advisor) ; Steklíková, Klára (referee)
Nowadays, dental pulp mesenchymal stem cells (dpMSC) are very popular amongst stem cells used for the study of cell therapy and regenerative medicine, due to their broad differentiation potential and simplicity of isolation. They are isolated from third molars which are often removed during routine surgeries in dentistry and would otherwise be discarded without any benefit as a waste material. Moreover, there are no ethical problems connected with their use. DpMSC fulfil all criteria established for mesenchymal stem cells, proliferate sufficiently and have a long lifespan in vitro. Besides their ability to differentiate into osteogenic, adipogenic and chondrogenic lineage, dpMSC are capable to differentiate into neurogenic, myogenic and hepatogenic lineage and into melanocytes and endothelial cells. Especially, dpMSC are used on animal model organisms for in vivo regeneration of tooth, bone, muscle, neuronal and liver tissue. Currently, several clinical trials are investigating the regenerative potential of dpMSC in humans. Apart from application in stomatology for tooth or bone regeneration, they can also be used for the treatment of the neurogenic diseases and for many other applications, on account of their origin in neural crest. The aim of this work was to write a literature review focused on...
Rights and obligations of parties to consumer contracts with a focus on distance contracts
Vacková, Kristýna ; Elischer, David (advisor) ; Dvořák, Jan (referee)
This thesis deals with the rights and obligations of parties to consumer contracts, where on the one hand is the consumer and the entrepreneur on the other. The thesis focuses on consumer distance contracts. The rule of law is protecting the consumer as a weaker party, and so the legislation strengthens the consumer's position. This tendency is even more significant for distance contracts, given that the distance nature of contracting may further disadvantage consumers. The thesis is divided into three thematic sections, preceded by definitions of terms that are crucial to the chosen topic of the thesis. The individual thematic sections of the tesis are subsequently: information obligations of entrepreneurs, withdrawal from the contract and rights arising from a defective performance. The first part deals with the various information obligations of the entrepreneur with a division into the information obligations resulting from the Civil Code and then from other legal regulations. The summary of this section is a simple list of mandatory information demonstrated on the model case. The second part is devoted to analyzing a specific institute for consumer distance contracts, namely withdrawal from the contract without giving a reason within a fourteen-day period. The third part defines the rights...
