Readiness of Selected Companies for Industry 4.0

The Diploma thesis is based on the analysis of four companies on the topic of implementation of Industry 4.0. The aim of this work is to study a current situation of the industry 4.0, to compare it with the literary theory and the to analyse it in each of the companies. It is crucial to explore the existing elements of the industry 4.0, attitudes and values that management acquires on these topics. These topics are analysed through research investigation, interviews, and observation in these companies. Subsequent research is a questionnaire survey. The analysis of the implementation of Industry 4.0 illustrates progress and necessary steps to remain competitive in the market. In companies there are detected some interesting points in progress or some defects of implementation.
The Analysis of the Organizational Culture in a Company

The Bachelor thesis is based on the analysis of organizational culture in Löttco company, which is an engineering business. The aim of this work is to study a current situation of the organizational culture, to compare it with the literary theory and then to analyse it in the business. It is necessary to examine the existing working environment, values, standards and the rules through the interview process with the management and current employees. A subsequent research is a questionnaire survey, which forms the basis for quantitative evaluation of the organizational culture. The analysis of the organizational culture illustrates how important part of company it is, how it influences the pace of work and the long-term sustainability of the business in the market. In the company there were detected some defects of the culture, the possibilities of changes were suggested for rising a healthy working environment.

Diploma thesis deals with the state of organic farming in the Vysočina Region, with focus on Třebíč district. The theoretical part includes principles of organic farming, current legislation, the problem of organic food and control system. It also describes the state of organic farming in the Czech Republic and in the Vysočina Region. Natural conditions and agricultural production are also mentioned. Another part is focused on comparison of organic agriculture in individual districts, which constitute Vysočina Region. The practical part deals with comparison of typical activities in the Třebíč district with other districts and describes important subjects. The last part is a questionnaire survey focused on two companies, organic farm DoRa and Tata Global Beverages Czech Republic. The conclusion constists of a summary of detected realities.
Analysis of marketing strategy of restaurant "Na Pekárně"

The thesis deals with marketing strategy of restaurant Na Pekárně. There are analyzed the opportunities, threats, strengths and weaknesses of the restaurant through a SWOT analysis, political-legal, economic, socio-cultural and technological environment through PEST analysis, microenvironment in relation to customers, competitors and suppliers, current marketing strategy of restaurant and used marketing tools. To obtain detailed information about the company was used marketing research in the form of interview and Mystery shopping. Based on the results of the analysis will be suggested a suitable marketing strategy for the future and potential improvements. Agroturistické využití Vinařství Sádek

This bachelor thesis deals with the utilisation of agricultural tourism of the viniculture Sádek. There are described the types of tourist travel as well as explained fundamental terms such as agricultural tourism and eco-agricultural tourism in the forepart. Another part is devoted to the region of Třebíč and to its climatic, soil and hydrometeorological conditions. There are also mentioned wine regions, wine agricultural tourism and Moravian wine trails in this part. A significant part of this thesis is devoted to the viniculture of Sádek, to its attractions and to offers for general public. There are also highlighted the attractions of towns and villages in vicinity of the Sádek corporation in this part. In the conclusion you can find my suggestions and ideas on behalf of viniculture advancement as well as appreciation, SWOT analysis and of the present state of the cooperation. The suggestions are focused mainly on agricultural tourism development and increase of visitor numbers.
Study of antimicrobial peptide induction in Brassica napus

Development of cheap, effective and environmentally friendly protection of industrial plants is a key problem of mankind, where using of antimicrobial peptides could be a reasonable solution. As a source for isolation of these peptides canola (Brassica napus) was chosen, which was treated by functional analog of salicylic acid – benzothiadiazole (BTH) – to stimulate the immune system. To isolate the responsible antimicrobial peptides, crude buffer extract was obtained. Next steps were precipitation, centrifugation and molecular weight filtration. The further isolation procedure comprised hexane extraction, SPE-C18 (Solid Phase Extraction) to separate hydrophobic and hydrophilic fractions and reversed-phase chromatography. Antimicrobial activity was tested by diffusion method. Fractions were tested on gram-positive and gram-negative bacteria and on yeast Candida scotii.
Isolation and study of the properties of recombinant pseudoosmotin

Antimicrobial peptides represent new clinical compounds with antimicrobial action and they could participate in solving the problem of ever-increasing resistance of microorganisms to the traditional antibiotics. The aim of this work was to study optimal conditions for expression, isolation and purification of recombinant pseudoosmotin from Escherichia coli BL21 (DE3). Pseudoosmotin was isolated from membrane fraction of bacteria and was purified by the affinity chromatography using Ni-NTA. Isolated pseudoosmotin did not show antimicrobial activity against studied microorganisms.
Larvae of flesh fly Neobellieria bullata as a source for novel antimicrobial peptides

The antibiotic resistant strains of microorganisms are increasing threat for public health. Antimicrobial peptides from natural sources display a wide spectrum of activity. In our laboratory, several peptide fractions with antimicrobial acivity against gram-positive and gramnegative bacteria and fungi were isolated from larvae of flesh fly Neobellieria bullata. The presence of peptides was proved by tricine electrophoresis, molecular weights and sequences determination by MS and Edman sequencing, respectively. In comparison with database sequences we obtained structures present mainly in insect and amphibian antimicrobial peptides, but total matches were not higher than in 36%. Upon the obtained sequence motifs we designed structures of several synthetic peptides. These were synthetised on the Rink amide MBHA resin. Synthesis was performed according Fmoc/PyBOP-DIEA protocol. Antimicrobial activity of these peptides was determined.
