Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Title, Name, Surname of candidate: Kateřina Lokvencová Title, Name, Surname of tutor: PharmDr. Hana Bártíková, Ph.D. Title of a diploma work: The impact of selected isoflavonoids on the effectiveness of anticancer therapy in MDA-MB-231 cell line. Breast cancer is the most common cancer disease in women. There is an extensive amount of therapeutic regimens for the treatment of this disease, but it is often limited by lack of efficiency or serious side effects. Isoflavonoids genistein (GEN), daidzein (DAID) and formononetin (FORM) belong among the natural substances with bioprotective effects on the human body. Some epidemiological studies have confirmed the protective effect of isoflavonoids against certain types of cancer, including breast cancer. The aim of this work was to study the effect of isoflavonoids on cancer cells and assess their ability to support the effect of cytostatics, namely anthracycline antineoplastic agents doxorubicin (Dox). For the experiments was chosen the breast cancer cell line MDA-MB-231. To study the effect of isoflavonoids on tumor cells we chose concentrations of 1 and 10 µM which is according to the bioavailability of isoflavonoids. We have shown that isoflavonoids...

Author: Bc. Kateřina Lokvencová Title: Factors related to chronic vulvovaginal discomfort - comparison of question- naires from 2007 and 2018 Diploma thesis Charles University, Faculty of Pharmacy in Hradec Králové Study program: Medical bioanalytics Background: The aim of the thesis was to get data that should help us to clarify the issue of factors related to chronic vulvovaginal discomfort and compare them with the results of Lucie Hájková diploma thesis from 2007. Methods: The questionnaire survey was focused on students and graduates of three col- leges in Hradec Králové. We put them into three groups - RVVD (women with chronic problems), VVD (women with acute problems) and control group (women with no prob- lems) and did statistical evaluation. Results: We found out that factors related to chronic vulvovaginal discomfort are family factors, vaginal douching, the use of scented panty liners, anal sex, oral sex, higher fre- quency of sexual intercourse, bigger number of sexual partners, stress and psychosocial factors. Consumption of dairy products can be protective. When we compared the data with the questionnaire survey from 2007, we found out that 40 % less women use hormo- nal contraceptives and it is replaced by less hormonal or completely non-hormonal forms of protection. We also reported...

Lokvencová, Kateřina ; Raisová Stuchlíková, Lucie (advisor) ; Matoušková, Petra (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Kateřina Lokvencová Tutor: RNDr. Lucie Raisová Stuchlíková, Ph.D. Title of Thesis: The pharmacokinetics of ivermectin in the feces of Ovis ammon Keywords: Anthelmintics, ivermectin, sheep, pharmacokinetics, excretion Infection with internal parasites is one of the most common diseases in sheep worldwide and causes significant economic losses. The nematode (Haemonchus contortus) is among to the particularly problematic representatives. Anthelmintics are used to treat helmintoses and they are divided into the several classes. The class of macrocyclic lactones includes ivermectin (IVE), which is especially popular for wide range of efficacy and low toxicity. However, the study of IVE pharmacokinetics is difficult, because it depends on many factors. Considerable attention is directed to the elimination of IVE, because its residuals in dung can adversely affect the environment and thus promote the emergence of resistance. The aim of this study was study of the elimination of IVE in sheep, especially the study of excretion profile. IVE (dose 0.2 mg/kg of body weight) was subcutaneously administered to sheep, followed by sampling of faeces at specified intervals. Ultra-high performance...

Author: Kateřina Lokvencová Title: Vaccination against papillomaviruses - Knowledge of high school students in Hradec Králové Bachelor thesis Charles University, Faculty of Pharmacy in Hradec Králové Study program: Medical bioanalytics Background: The aim of the thesis was to find out the knowledge of adolescent girls about vaccination against human papillomaviruses and HPV infection. Next, to find out how many girls are vaccinated and finally what is their attitude towards this topic. Methods: The questionnaire survey focused on female students aged 15-20 years from six high schools in Hradec Králové. Results: In total, 529 students were involved in our study. We found out, that the aware- ness of the existence of human papillomaviruses among the students is not very satisfying as well as their knowledge of the way of HPV transmission. Altogether 65 % of respond- ents know about the possibility of vaccination and 46.3 % of the girls are vaccinated. More than half (56.9 %) of respondents participate in gynecological examination, the girls from the hotel school go there the most. Students from the veterinary school have the best knowledge of HPV and cervical cancer. We found out that the students of the medical high school don't have better knowledge of this topic comparing the students of other...
