Mapping Survey of the locality Areal Metra company in Blansko - north part

The diploma thesis deals with the creation of map bases in the northern part of the Metra areal in Blansko in the form of a thematic map in a scale 1:250. Descriptive information for each element has been added to the map. The thesis describes in detail the process of creating a measuring network, detailed survey, calculations, testing accuracy and creation of a thematic map including descriptive information.
Comparison of sensors for temperature measurement

This work deals with temperature sensors. Basic terms are defined at the beginning of work. The following are distribution, description of construction and principle of touch sensors with electrical output. Other touch sensors are marginally described. Further is described principle of pyrometry and contactless sensors. In conclusion, work contains practical example of contactless temperature measurement.
Documentation of Barová cave - central part

The aim of this bachelor thesis is the graphic documentation of Barová cave – central part in the form of a plan and transection in a scale of 1:250. The thesis contains, including brief description of the area, all steps from reconnaisasance and completion of the existing measuring network to the measuring and processing part leading to the creation of the resulting map.
Design of control of axis with linear motor

This thesis deals with construction, connection and control axis with linear motor. The beginning of thesis is focused on principle of functioning and characteristics of linear motors. The following part describes used components of axis, their mechanical configuration and electrical wiring. Further thesis deals with creating PLC program and user interface for control axis in TwinCAT system. In last part is dealing correct software settings of axis.
Studies of nanocrystalline silicon colloidal suspensions

This diploma thesis deals with optical and biophysical properties of colloidal suspensions of porous nanocrystalline silicon (por-Si) in different media, conditions necessary to create por-Si with different luminescence peaks and their optical characterization (emission and excitation spectra). The lifetime excited state = (28,6±0,9) us on emission wavelength 600 nm for orange emitting por-Si is measured by the time resolved fluorescence. The behaviour of por-Si and grinded silicon (crystals of size about 1um) in biological environment (L929 mouse fibroblast) is studied with optical transmission microscopy, time-lapse microphotography of cell culture evolution, fluorescent microscopy, and scanning electron microscopy (SEM). Transmission pictures prove an aggregation in presence of por-Si in media. This aggregates are going to internal environment of cells. At high concentration of grinded silicon we see meronecrosis. Por-Si has had also mildly negative effect on evolution of cell culture at very high concentrations, but cells are viable and cell division proceeds normally - next cell generation shows normal behaviour. Fluorescence microscopy used on orange emitting por-Si shows that por-Si is concentrated probably on cell membranes and its presence in internal cell environment is also proved. SEM proved...

Supervisor: RNDr. Jan Krůšek, CSc., Institute of Physiology AS CR, v.v.i. Abstract: Nicotinic acetylcholine receptors (nAChRs) are ligand-gated ion channels. We use the whole-cell patch-clamp technique to study functional and pharmacological properties of muscle and neuronal nicotinic receptors. Rat neuronal receptors were heterologously expressed in COS cells and human embryonic muscle receptors were studied in TE671 cells. Lobeline, a plant alkaloid with a long history of therapeutic use, interacts with the classical agonist-binding site of nAChRs. The final result of this interaction depends on the receptor subtype, lobeline and other agonists concentrations and the time schedule of application. Generally, lobeline is a very weak partial agonist eliciting deep desensitization at several subtypes of nAChRs. In combination with other agonists, lobeline acts as a competitive antagonist or coagonist. Using point mutation procedure we studied the functional role of negatively charged amino acids in the F-loop of β2 and β4 subunits of neuronal receptors.
