Characterization of plasma during organosilicone thin film deposition using haxamethyldisiloxane monomer

The aim of this thesis is plasma diagnostic during deposition of thin films based on organosilicones. Hexamethyldisiloxane was used as a precursor for plasma deposition in the presence of oxygen, and the diagnosis was performed by using optical emission spectroscopy. The theoretical part summarizes the basic characteristics of plasma and processes occurred at plasma polymerization and deposition. It also deals with thin films and their use in coating materials. A relatively large part is devoted to the specification of the physical-chemical nature of the used analytical method – optical emission spectroscopy. Finally, the principles of rotational, vibrational and electron temperature calculations are described. The contemporary deposition process was carried out in continuous and pulsed mode of radiofrequently excited capacitively coupled discharge. The effects of monomer flow rate, plasma power and duty cycle on the deposition process were studied. In the individual spectra, atomic hydrogen lines of Balmer’s series as well as an atomic oxygen line were identified. Molecular bands of CO have been identified as Angstrom’s system and 3. positive system. In the case of the identified fragments, the intensity dependencies on the monomer concentration in mixture, plasma power and duty cycle were detected. Electron temperature of plasma was also calculated using the relative intensities of atomic hydrogen lines of Balmer’s series. Vibrational and rotational temperature could not have been determined because no suitable fragments for their determination were found in the spectrum. Based on findings mentioned above, partial composition of plasma and some of its properties were estimated. The subject of the further research will be determination of the exact content and structure of the thin films and investigation of other plasma characteristics. Plasma deposition is influenced by many factors, and the study of their optimal combination for the most efficient deposition process is a goal for the future research.

Sperm protein profiles of different mammalian species

Proteins are a substantial equipment of the spermatic cell; therefore, the characterization of sperm proteins is crucial for explanation of molecular mechanisms in the reproduction process. We isolated sperm proteins from different mammalian species - pig, bull, human, mouse, dog and cat. Extracted proteins were separated by SDS-electrophoresis and protein/glycoprotein profiles from epididymal or ejaculated sperm were compared. Additionally, we tested cross-reactivity of antibodies prepared to sperm boar proteins on spermatozoa of other mammalian species using immunofluorescent technique. Our future plan is to compare the protein profiles of sperm during their functional development (epididymal, ejaculated, capacitated) in various mammalian species and identify species-specific sperm proteins with zona pellucida binding activity.

Role of Psb28 proteins in the biogenesis of the Photosystem II complex in the cyanobacterium Synechocystis sp. PCC 6803

The thesis focuses on the role of Psb28 proteins, namely the Psb28-1 and its homolog Psb28-2, in the biogenesis of the Photosystem II complex (PSII) in the cyanobacterium Synechocystis PCC 6803. The aims of this work were to localize the proteins within the cells, and to determine their function. A fraction of both Psb28 proteins was identified in the monomeric PSII core complexes but most proteins were found in the unassembled protein fraction associated with thylakoid membranes. Psb28-1 was mostly detected as a dimer while Psb28-2 as a monomer. Psb28-1 also differed from Psb28-2 by its higher affinity to the PSII core complex lacking CP43 antenna. Characterization of Psb28-less mutants suggested regulatory function of the proteins in PSII biogenesis in connection with chlorophyll biosynthetic pathway. Analysis of preparations isolated using FLAG-tagged versions of Psb28 proteins showed their association with Photosystem II - Photosystem I supercomplexes, especially under increased irradiance, and supported a role of Photosystem I in the PSII biogenesis.

Underground research site Bukov - geotechnical characterization of the site

Podzemní výzkumné pracoviště Bukov (PVP) je navrženo jako testovací lokalita pro zhodnocení vlastností a chování horninového masivu, typově blízkého vybraným kandidátním lokalitám, v hloubce odpovídající předpokládané úložné hloubce finálního hlubinného úložiště vysoce aktivních odpadů v České republice. PVP Bukov, jehož výstavba byla zahájena v roce 2013, je realizováno v jižním křídle uranového ložiska Rožná, na severovýchodním okraji strážeckého moldanubika, poblíž jeho styku se svrateckým krystalinikem. Důlní díla pracoviště jsou situována na 12. patře jámy Bukov-1, v hloubce přibližně 600m pod povrchem.
