The analysis of selected parameters of water quality

Cílem této diplomové práce je provést analýzu vybraných ukazatelů kvality pitné vody a posouzení faktorů, které ji ovlivňují. V první části jsou zachyceny vybrané mikrobiologické ukazatele, tak jak je popisuje literatura, včetně platných norem a vyhlášek. Druhá část popisuje průběh provádění vlastní analýzy, z poskytnutých materiálů akreditovanou laboratoří společnosti AGRO-LA, spol. s r. o., až po vyhodnocení získaných výsledků.

Evaluation of the Technical State of Water Treatment Plants

The subject of this thesis is to create evaluation criteria of technical condition of water treatment plants. The thesis is divided into four parts. In the first part of this thesis are presented evaluation systems by performance indicators drinking water treatment plants, which are then applied in real time. There are detailed descriptions of individual performance indicators. Each evaluation conducted certain period of time. After the expiry of the period under review was to evaluate and create charts to display results. In the next chapter the thesis focuses on the identification data. First, it was necessary to find out all necessary information about the water treatment plant. Necessary information means information relating to building construction, human resources, mechanical parts, technological equipment and other entities with links to the operation of the water treatment plant. All these data were sorted for better overview and facilitate setting evaluation indicators. The third part of this thesis is focused on the development of indicators that are used to determine the current status of the water treatment plant. As a first step was established 19 indicators used to evaluate the condition of the entire water treatment plant. Followed by a description of the individual indicators and then determine the individual evaluation criteria. In the last part of this thesis was carried out by the proposed criteria on selected water treatment plants. They were selected three water treatment plants located in the Czech Republic, which held a personal tour of each water treatment plant. This was followed by an overall assessment of the technical condition of water treatment plants.

How to prevent the proliferation of Legionella pneumophylis in hot water in the hospital Czech Krumlov

This diploma thesis deals with microbiological quality of hot water in a hospital in Cesky Krumlov as well as hot water piping condition with respect to Legionella. It also deals with arrangements for improving the quality of hot water in this hospital. The subject and target of this thesis are the findings of the mentioned parameters based on own observations, technical documentation and analysis of warm water performed by an accredited laboratory. The hospital in Cesky Krumlov, the construction of which began in 1909, was built in 1911 as the 'Nemocnice Císaře Františka Josefa I.' As the time went by, new buildings were built up. The hospital was renamed to 'Všeobecná veřejná nemocnice' in 1942. In the following years the hospital expanded until the current condition. Water quality and presence of Legionella is a world-wide issue, which we must fight against. The thesis is divided into two parts. First, theoretical part, which includes both general and specific knowledge about Legionella, its health risks, the environment in which it occurs, possibilities of eliminating, suitability of the material for hot water pipes, gained in professional literature and a part of the Czech legislation dealing with public health protection, the law 'No. 258/2004 Coll.' in actual version and quality of drinking water and hot water. In the second, empirical part, I focus on the situation of the hospital in Cesky Krumlov from 2009 till 2015, concerning newly built boiler room and the whole distribution system of hot water regarding Legionella. Particularly I focused on materials used for ecologisation of the gas boiler room and materials used for hot water pipes and actual state of water distribution in individual buildings of this health care facility. I mentioned the analysis of the water samples performed by an accredited laboratory with corresponding technical documentation, which concerns the water distribution system, provided by the medical facility. I used secondary data analysis to evaluate these documents. Regarding the results from 2009 to 2015 the microbiological quality of warm water doesn't meet the limits defined in the Attachment No. 3 of the above mentioned law Order 252/2004 Coll. Limit value for colony forming units of Legionella is set to maximum of 100 CFU. In most of the samples this limit was exceeded by series of units up to hundred thousands of units. The last results in 2015 prove improvement of the unfavorable situation. In that year the analysis was tested three times. First in February, where the CFU units were 'only' in hundreds, with the top value of 550 CFU in the Postacute Care ward. Second analysis was performed in June that same year. Results were acceptable. The highest value was 64 CFU. Last analysis in the hospital was performed in December. Increased concentration of Legionella was again in the Surgery building. There were established of 1400 colony forming units. There were 300 CFU in the Postacute Care ward and the limit value of 100 CFU was in the ward of Internal medicine. The answers to research questions 'How to prevent reproduction of Legionella peumophylis in the water distribution system of the hospital in Cesky Krumlov?' and 'How to ensure a sufficient amount of quality hot water in the medical facility?' are included at the end of the stated recommendations. A risk factor causing the fast reproduction of Legionella is the old water distribution system in most of the buildings and low water temperature of water leaving the boiler, which is 55°C. All mentioned results and evaluations are included in this thesis as well as attached pictures, which show the environment and situation within the Health Care Institutions.

Comparison of HPLC a GC-MS method for pesticide determination in soil

Physical and chemical properties of pesticides may vary significantly. Pesticides can be acidic, neutral or alkaline. Some compounds containing phosphorus, other sulfur, nitrogen or halogens. These atoms can be important for the detection of pesticides. Some of the compounds are volatile, others on the contrary not evaporate, which is also one of the important properties for chromatography. The diversity of pesticides actually makes it impossible to create a universal method, which is very impractical in terms of quality and time determination. Because that pesticides are one of the most chemicals in most countries. In the food and drinking water are limits on pesticides in order to avoid negative impact on public health. For residues in soil or surface water limits are set. (Lutz et al., 2006) The objective of this thesis is to evaluate the possibility of using two types of chromatographs with different detectors for the analysis of pesticide. Pendimethalin which was chosen for this work, will be analyzed usinga liquid chromatograph with UV detektor and gas chromatography with mass detector. Both this method will be compared as well two extraction methods (Soxhlet extraction and shaking), and two extraction agents (methanol and acetonitrile). Another point of the work is to provethe hypothesis that, for the determination of pendimethalin in soil is by gas chromatography using a mass spectrometer in comparison with the high performance liquid chromatography time and more expensive, but will achieve a higher yield and a lower detection limit of the pesticide.

Analyze of drinking water marketing

The aim of this bachelor's thesis is to chart marketing mix of drinking water in the Czech Republic in respect to the trends in the world. Together with this aim this thesis intend to compare these two marketing mixes of bottled drinking water and tap water and make the analysis of them to create recommendations about marketing of tap water. This bachelor thesis is divided into two parts. The first one contains theoretical chapters about marketing, marketing mix, marketing research and SWOT analysis. Second part contains definitions of terms drinking water, bottled drinking water, tap drinking water etc. This part also contains history of bottled drinking water and mostly analysis of marketing mix of both types of drinking water. In the conclusion of this bachelor thesis are made some recommendations about marketing of tap water to enhance the position of tap water on the market.

Analysis of biological features of Capsella bursa-pastoris (L.) Med.

The aim of this bachelor thesis is to determinate selected biological characteristics of shepherd´s-purse, which have impact on the establishment and expansion of its populations in field crops. In particular, development of primary dormancy was studied and the effect of factors that influence the dormancy and germination of seeds was determined. Shepard´s purse is an annual or biennial herb of the cabbage family, which is classified as winter annual weeds. Primarily infest the winter and spring crops, rapeseeds, potatoes or forages. It appears from lowland to foothills, so biological features of individual population can be very different. The ways of control are shallow tillage or herbicidal treatment. After harvesting of ripe seed the first experiment was set to define the primary dormancy. Remaining seeds were stored in two different types of environment. Part of seeds was stored in the dry at 20 °C. Others seeds were stratified at 5 °C. Effect of storage conditions on primary dormancy was tested every two weeks. Seeds were germinated for 1 week in grow chamber under the influence of various factors such as different light mode, type of water and type of media for germination. The results were processed by multiple-factor analysis of variance. Fresh seeds showed a strong primary dormancy at constant temperatures of 10 °C and 20 °C. Treatment with fluctuating temperature (20/10 °C), filter paper, drinking water and stored in dark had the highest germination rate (10.75 %). Seeds stratified at 5 °C had increase in germination in first weeks, but lost it during storage. On the other hand, seeds stored in dry at 20 °C firstly were in dormancy, but they lost it gradually during storage.

Water Resources and their Technical Construction

Nowdays, the renewal of water supply infrastructure is becoming increasingly hot topic. Gradual renewal is one of the key points for long-term maintenance of quality and trouble-free operation. The most basic element of the entire water supply system are water resources, which shouldn't be neglected during the planning of a renewal. This thesis deals with technical construction of drinking water resources and it is mainly focused on ground-water resources. The main part of the thesis is the design of a technical assessment system for underground drinking water resources for public water supply. The purpose of such system is to create a tool for technical assessment that will identify current technical status of multiple water resources and will compare them. This comparison enables to create more effective renewal plan in future.

Sanitation installation in an apartment building with commercial space

This thesis describes the design of plumbing in an apartment building with commercial space in Ilava. The theoretical part is devoted to the design of the container hot drinking water and it´s possible alternatives. The work also focuses on possible solutions diploma work. Computational part of a project proposal contains sewage and rainwater drainage, water supply systems and connecting the building to the existing network hardware. It is a residential house, which is in part intended for commercial premises and the second part is intended for housing. The building has four floors and no basement.

Sanitation installation in student dormitories

Diploma thesis describes the design of plumbing systems in student dormitories. The theoretical part discusses the types of waste water, focusing on gray water and its subsequent use. The experimental part is focused on measurement drinking water consumption and drinking water discharge in existing buildings student dormitories and apartment building. The work also contains other variants of possible installation solutions. The project is part of the selected solutions developed a variant form of project documentation for construction.

Effect of probiotic bacteria Lactobacillus paracasei subsp. paracasei on the development of acute colitis in BALB/c mice.

Inflammatory bowel disease - ulcerative colitis and Crohn's disease - are chronic relapsing intestinal disorders of unknown etiology. Acute ulcerative colitis was induced in BALB/c mice by 3 % dextran sulfate sodium (DSS) administrated in drinking water for 5 days. The effect of Lactobacillus paracasei subsp. paracasei RL14-P, a bacterium with potential probiotic influence, on the development of inflammation was evaluated. Lactobacillus was administrated intragastrically to mice on days 1, 4 and 8 and the administration continued for next 5 days during DSS treatment. Control groups received instead of lactobacillus MRSC medium or phosphate-buffered saline according to the same schedule. Clinical symptoms such as diarrhoea, rectal bleeding and loss in body weight were evaluated daily. Cytokine secretion (IL-6, IL-10 and TNF-α) was determined in supernatants of 48 h cultivated splenocytes, mesenteric lymph node lymphocytes and colon pieces by ELISA. The impairment of colon descendens was evaluated histologically. Lactobacillus paracasei subsp. paracasei RL14-P administrated mice remained healthy without signs of the intestinal inflammation. We did not find the shortening of colon (a sign of inflammed intestine) in lactobacillus-treated mice. We observed that administration of probiotic bacteria...
