Identification DNA of Plant and Animal Species in Food by Polymerase Chain Reaction
Šmíd, Jiří ; Hanák,, Petr (referee) ; Timko,, Jozef (referee) ; Kuchta, Tomáš (advisor)
We were developing detection methods for three food allergens of plant origin. We used real-time PCR for soy detection in food oriented on gene lec, that is coding lektine specific for soy. On this target sequence were oriented PCR system with primers Le2F and Le2R and TaqMan probe Le2P. Detection limit (2,75 pg), practical detection limit (0,02 %), inclusivity and exclusivity were determined. Whole system were quantified. Real-time PCR for pistachio detection were based on primers and probe for gene COR. Detection limit (3,5 pg), practical detection limit (0,002 %), inclusivity and exclusivity were determined. For almond detection we were not succeed system, that fulfil all qualitative parametres.
A complex study of biological effects of some soya products
Ručková, Michaela ; Němcová, Andrea (referee) ; Márová, Ivana (advisor)
Soybeans and products thereof are considered as a very contradictory legume across layman and expert opinions. Some see it almost as a “superfood” while others avoid it due to its biologically active content with an unclear effect on the organism. As a lot of research on soybeans was already performed the content varies considerably and it is not easy to grasp the issue correctly. The objective of the research part of the thesis is a structuralized study of already published scientific knowledge to clarify the current state of the art. Experimental part of the thesis focuses on characterisation of soy and its products in terms of active ingredients content and determination of antioxidants and antimicrobial effect. After that, the cytotoxic effect on human heterogeneous colorectal adenocarcinoma epithelial cell line and mouse melanoma cell line is determined by in vitro testing. All obtained information and results of experimental part could possibly serve as a starting point for further study of soy and soy products, both in the dissertation and in the submission of a grant project.

PCR detection of genetically modified soybean in food and feed
CHADIMOVÁ, Nikola
Genetically modified organisms which genetic equipment has been specifically targeted of have been acquiring properties they did not have before. One of the main reasons for this aimed genetic modification in plants is herbicide resistance. Especially in soy case is used the modification to resist to glyphosate. The safety of the food which has been made of genetically modified crops or products of animal fed with GM feed has not been proved so far. This bachelor thesis focuses on methods for detection of GMOs in food and feed. Also, it focuses on verifying the data concerning the presence of GMOs presented by companies on food or feed packaging. The research part of this work contains information about genetic engineering, the issue of genetically modified organisms and also focuses on the description of techniques needed to detect GMOs, especially in processed foods, but also in feeds available on the Czech market. The methodical part of this bachelor thesis focuses on the detection of CaMV 35S promoter occurring in DNA crops that have undergone genetic modification. The presence of a specific transgenic material involved the detection of a particular glyphosate-resistant gene, which is particularly known under the trade name Roundup. There were used basic molecular biological methods to analyze GMOs.
PCR detection of genetically modified soybean in food and feed
JOSKOVÁ, Markéta
The aim of this thesis was to summarize in the theoretical part the issue of genetic modifications and their main use. Another task was to describe various methods used for detection of genetically modified plant products and their properties. The practical part is focused on the isolation of DNA from various foods and feeds containing soy. Using the PCR method, samples were tested for the presence of soy DNA, the CaMV promoter, and the Roundup resistance transgene. A total of 42 samples were tested and 5 of them contained transgenic soy.
Comparison of Czech and world superfoods
Soldánová, Lucie ; Vojáček, Jan (advisor) ; Slimáková, Margit (referee)
v angličtině k bakalářské práci na téma "Srovnání českých a zahraničních superpotravin" ,Lucie Soldánová, Karlova Univerzita, 1. lékařská fakulta, obor Nutriční terapeut. This bachelor thesis focuses on the topic of superfoods and aims to discover how our crops differ from those from abroad. The work consists of a theoretical and practical part. In the theoretical part of my thesis, I firstly analyze five most well known foreign (avocado, soy, goji, chia seeds and quinoa) and Czech (various seeds, poppy seeds, walnuts, blueberries and vegetable juices) superfoods. Then I compare them on four levels- a nutritional, economical, ecological and ethical one. In the practical part I asked several questions related to the problematics of superfoods and with the use of sent out questionnaires investigated whether people know superfoods, what is their opinion on them, if they buy them and whether they think about their origin and deeper problematics of the import of exotic crops. I discovered that more than 75 percent of those questioned are aware of superfoods. Whole 40 percent of people were somewhat sceptical of these foods and only 18,5 percent expressed full trust in them. Almost 73 percent of respondents buy superfoods at least once a week, which reflects their more frequent consumption, while the...

Methodology for analysis of dairy products and their soy analogues for the presence of RoundUp Ready soybean
Tesařová, Zuzana ; Šídová, Dana ; Vráblík, Aleš ; Hodek, Jan ; Ovesná, Jaroslava
Dairy products may be subject to adulteration, where there is a relatively expensive raw material (e.g. cream) replaced by the addition of vegetable fats. Milk can be replaced in dairy products for example by soybean extract. Because soy belongs among strong allergens and the incidence of allergy is rather high. Therefore it is necessary to have a method for its detection. This work introduces the method for detection of soy, including analysis of genetic modified RoundUp Ready soybean. Detection of soybean in dairy products is based on analysis of amplification products melting profiles by High Resolution Melting (HRM) method. Analysis of geneticaly modified RoundUp Ready soybean is based on DNA detection via Polymerase Chain Reaction(PCR).
