Biochemical Aspects of Artwork Restoring

The following bachelor thesis is focussed on findings on the field of art restoring in connection with its biochemical aspects. In itroduction is resumed brief history of art restoring and generally defined terms of preservation, restoring, art and paint. For better understanding basic paint techniques (aquarelle, gouache, tempera and oil painting), strata sequence and instruments and materials commonly used for art work are mentioned. Most frequently used organic and inorganic pigments are briefly mentioned, however this thesis is mainly focussed on applied organic compounds - lipids, proteins, polysacharides and terpenoids - its composition, characteristics, usagea nd possible interactions. Screening of possibilities of deep subsurface microflora to decompose selected organic compounds Abstract The aim of the study is to test the deep microflora bacterial strains for their ability to grow in oily environment, aliphatic hydrocarbons and toluene were taken as examples of aromatic hydrocarbons and where the cultivation of selected strain, were produced for testing its growth and microbial activity of selected strains in conditions simulating soil conditions; assess the usability these strains in practical remediation of contamination by oil. This thesis deals with the evaluation of possible use of selected strains of deep microflora for oil decomposition. It is evident, that some groups of microorganisms living in the Tertiary claystones at depths of 30-450 m below the surface are the biodegradable fossil organic matter type of kerogen. Chemical findings indicate that, this organic matter consists of various lengths of aliphatic chains, and thus the assumption that microorganisms decomposing kerogen might be able to disassemble oil and petroleum products. The findings of our experiment indicate that benzene and toluene, as well as kerogen are highly resistant to organic compounds and evidence of microbial degradation are rare. Mechanism of uptake of heavy metals and organic compounds by plants

Heavy metals and organic compounds represent a great problem for environment. Therefore, attention is paid to the development of techniques for removing these pollutants from the sites of pollution. One of the possibilities is that heavy metals and organic compounds are removed from soil by using fytoremediation. Fytoremediation uses the ability of some plant species to uptake toxic compounds from soil and accumulate these compounds in its tissues. These plants are able to tolerate higher amounts of toxic compounds than it's usual because of the broad range of detoxification mechanisms, which are taking place in these plants. After the root uptake, heavy metals may be chelated by phytochelatins, metallothioneins or other chelators and subsequently stored in vacuoles. Transport of metal ions and metal complexes between cells compartments is mediated by many specific transport proteins. Detoxification mechanisms of organic compounds involve transfomation, conjugation and store of these compounds in plant tissues.
NMR Aerosolomics as a Tool to Distinguish Various Types of Aerosol Samples.

In the recent study, the summer and winter aerosol samples were analyzed using NMR aerosolomics approach. The samples were collected in Prague-Suchdol during summer 2008 and winter 2009 in two different particle size fractions - PM2.5 and PM 10. Around 50 compounds were identified in each aerosol spectrum owing to the comprehensive library. The profile of 86 identified compounds, which were identified in the samples altogether, served as an input data for statistical analysis. Multivariate statistical analysis clearly discriminates the two groups studied. Furthermore, it is possible to determine the most significant compounds.

Co-Mn Mixed Oxides Preared by Magnetron Sputtering on Meshes as Catalysts for Oxidation of Organic Compounds.

The catalysts with various Mn content were examined by XRD, SEM, FUR. XPS, and Ih-TPR. tested in the gas-phase oxidation of model organic compound (ethanol) and compared with pelletized commercial Co-Mn- Al mixed oxide catalyst (Astin 2-100, Czech Republic, Co:Mn:Al molar ratio of 4:1:1 ).

Application of Raman spectroscopy for detection of sulfates of self-ignited coal heaps

This Ph.D. thesis was focused on the application of Raman spectroscopy as the main analytical method for the characterization of neo-formed minerals, notably sulfates, from burning coal waste dumps. This environment associated with subsurface fires gives rise to a variety of uncommon and rare minerals. The specific features of these minerals (metastability, hygroscopy, mixed aggregates) causes that the mineralogical investigation is a challenging task using traditional laboratory-based techniques. Advantages such as the non-destructive nature, the sensitivity to the changes in the hydration degree of sulfates, little or none pretreatment, and the option of measurements directly in the field were the main reasons for applying this spectroscopy method. The scarce availability of spectroscopic data of most gas-vent minerals can be considered as the disadvantage. Therefore, artificial prepared samples of six anhydrous sulfates, which are rarely found in nature, were analyzed by Raman laboratory spectroscopy and a miniature a Raman spectrometer, and specific Raman features as well the differences with hydrated counterparts are shown. Nomenclature of organic compounds

One of the expected outputs of organic chemistry in "RVP G" is the application of rules of organic systematic terminology for the description of compounds, with the posibility of sing trivial names. There are several handbooks oriented on nomenclature of organic compounds that are available on Czech market. However, repeated imperfections appear in them permanently: the textbooks do not comply with the hybridisation of carbon atoms, the same simple examples are repeated, the rules are so simplified that they lead to the application of incorrect terminology principles etc. The submitted thesis is dealing with the matters of the terminology of organic compounds - with created tutorial material as its outcome. It is in the form of compendium of organic terminology accompanied by the exercises. The contribution of submitted thesis is complex insight into the theme and collection of tutorial materials intended mainly for the high school teachers first of all. Nevertheless it can be used by the students of high schools or basic courses of colleges as well.
