Center for seniors in Brno-Medlánky

The task of the diploma thesis is to design a senior living facility in Brno – Medlánky. This task was preceded by a pre-diploma project within the subject NGA056 – Specialized project, developed in the previous semester, which consisted of preparing an urban study with a proposal for urban structure including transportation solutions and the placement of various functions - residential areas, commercial areas, public buildings, civic amenities, a sports hall, a church, and further development of the senior living facility. The architectural study focuses on creating a suitable environment for long-term stay for seniors, meaning comfortable living, outdoor relaxation areas, adequate technical infrastructure, and a variety of services. The design stems from the concept of a compact, clear, and barrier-free base space with social areas and facilities connecting three pavilions of housing units. This ensures privacy in the housing units while providing ample space for social interaction. Access to the terrain linked with a park located on the southern side of the plot is easy from both the base floor and the housing units, providing direct contact with greenery in outdoor spaces. The main entrance leading to the base on the first floor is designed with a representative atrium, reflecting the grandeur of the entire complex. Technical and operational facilities are expanded to cover the capacity of a potential additional fourth pavilion. The entire complex reflects the requirement for peaceful and high-standard living in old age, respecting contemporary architecture and the needs of today's population. As a result, it becomes a place for contented aging.
RECONSTRUCTION AND REBUILDING OF CITY BREWERY IN VYŠKOV

The bachelor´s thesis focuses on the solution of today's unused Vyškov brewery, which is part of the city's history. The proposal presents a new use of existing buildings and revitalization of the entire complex. The main task is to revitalize this area and find suitable uses that will co-operate and complement the original main function of the site.
Multifunction centre in Opava

Master´s thesis deal with the supporting steel structure of multi-storey multifunction center in Opava. The complex consists of three sub-objects of three and five storeys with a total floor plan dimensions 48 m × 82.27 m. Five-floor building is designed with ground plan dimensions 42 m × 30 m with the distance of the poles 6.0 m x 6.0 m. The median three-storey building is designed with ground plan measuring 42 meters x 22.27 meters to the distance of the poles 6m x 5m. Ultra three-storey building is designed with ground plan dimensions 42 m × 30 m with the distance of the poles 6m x 6m. Construction of the complex height is 5m.
Intensive City House

The topic of diploma thesis is to design a "city house", in a form of the intensive urban structure, using the area with the greatest efficiency in terms of housing, services and jobs on a relatively small area. The main function of the new complex is housing supplemented by services, as well as, leisure and education facilities. The art school, the gallery, the office space and other facilities serve not only the residents of the "intensive house", but also the rest of the public, with the possibility of getting new jobs at an accessible distance, which is the main task of an intensive structure. The aim is to create a multifunctional urban building that revitalizes the inefficiently used area and at the same time brings green spaces designed for relaxation that are missing in this area.

Zuzana Kotková - Ph.D. Thesis: MRI Contrast Agents for Modern Applications 1 Abstract Over the last few decades, Magnetic Resonance Imaging (MRI) has become one of the most powerful medicinal methods for the diagnosis of various diseases (Fig. A1a ). In several types of examinations, contrast agents (CAs) are used to increase the quality of images (Fig. A1b ). These contrast agents are usually based on paramagnetic metal complexes of macrocyclic ligands. In dependence on the measured nuclei, two basic groups of CAs are considered - 1 H MRI CAs and, recently, also 19 F "hot-spot" CAs. The 1 H MRI CAs are based on Gd3+ complexes of macrocyclic ligands with one water molecule coordinated to the central metal ion (Fig. A1c ) and the effect of the CAs is to shorten the relaxation times of water protons. Characterization of the binding interface between transcription factors FOXO4 and p53

This work deals with the study of human transcription factors FOXO4 and p53. FOXO4 is a member of the "O" subfamily of FOX transcription factors. Genes encoding FOXO proteins are evolutionarily conserved across species. FOXO transcription factors regulate the expression of genes involved in the control of metabolism, cell cycle and cell proliferation, cell survival and stress resistance. They are considered tumour suppressors because of their ability to arrest the cell cycle and induce apoptosis. However, their function in tumorigenesis appears to be more complicated, as recent studies indicate a poorer prognosis for the development of tumours that express higher levels of FOXO4. The p53 protein is a thoroughly studied naturally occurring tumour suppressor. The cellular response after its activation is somewhat similar to that of FOXO4, it can also block cell cycle progression or induce apoptosis depending on the cell type and severity/type of cellular stress. Both FOXO4 and p53 appear to be key molecules affecting aging. Under stress conditions, p53 and FOXO4 interact with each other and together increase the expression of p21 protein, thereby inducing the transition of cells to a senescent state. Conception of an Object as a Complex of Perceptions in David Hume's Philosophy

Práce pojednává o objektu (jakožto komplexu percepcí) a s daným tématem souvisejícími otázkami v kontextu filosofie Davida Huma, jak je obsažena především v první knize jeho díla Treatise of Human Nature a rovněž s přihlédnutím k dílům Enquiry Concerning Human Undestanding a An Abstract of A Treatise of Human Nature a dalším titulům primární a sekundární literatury. Tematizováno je v tomto kontextu tedy nejprve Humovo pojetí percepcí jako takové (a to na základě první kapitoly Treatisu, Of ideas, their origin, composition, connexion, abstraction etc.). Poté je v souvislosti s předchozím pojednáno i o Humově pojetí identity (především objektu-tělesa) a jeho koncepce individuace, resp. i stálosti a koherence. (a to na základě čtvrté kapitoly Of scepticism with regard to senses). Na konci práce jsou kontrastovány různé možné typy objektů, jež lze (dle autora této práce) chápat jako komplexy percepcí a poté je nabídnuto schéma hypotézy o individuaci tzv. materiálních těles (bodies) v prostoru.
Biophysical characterization of the CAR/RXR protein complex with its binding partners

The constitutive androstane receptor (CAR) plays an important role as a xenosensor in the organism and is therefore widely expressed in the kidney, liver, gallbladder and in the small intestine epithelium, where the biotransformation of xenobiotic occurs. CAR is also an important factor in the elimination of bile acids and bilirubin. CAR has also been shown to have an important role in regulation of glucose and lipid metabolism. Impairment of lipid and glucose metabolism is a common cause of cardiovascular and metabolic diseases such as atherosclerosis, type 2 diabetes, obesity and insulin resistance. These diseases are called metabolic syndrome and result in severe organ damage. CAR respective its complex with RXRα (retinoid X receptor α) has become a promising biological target for drug discovery for metabolic syndrome. The major aim of this study was a structural characterization of the CAR/RXRα protein complex together with the agonist IV676*HCl, which would describe in detail the interaction of this small molecule with the receptor. Obtained structural information would be used to design improved agonists.
