Modulation of BELL transcription factors balance as a tool for tuberisation strengthening in selected potato cultivars

Potato (Solanum tuberosum) is the third most important crop in terms of human consumption. Hence understanding regulatory mechanisms controlling tuber initiation is significant not only from a theoretical but also practical point of view. Tuberization of S. tuberosum is a complex morphogenic process regulated by both, environmental conditions and inner plant status. Environmental conditions regulating the onset of tuberization have been known for a long time, but the molecular basis of tuber initiation has only recently been discovered, in particular using wild subspecies S. tuberosum subsp. andigena. BELL transcription factors are among the important components of the regulatory signalling network. The aim of this study was to modulate balance between the BELL transcription factors that induce and repress tuberization towards strengthening of tuber induction using two cultivars of Solanum tuberosum subsp. tuberosum, cv. Kamýk, resp. cv. Korela. To achieve this, transgenic lines carrying construct containing a part of coding sequence StBEL11, resp. StBEL29 in antisense orientation (AS) under control of constitutive promoters and lines carrying complete sequences of StBEL5 and StPTB6 genes under control of their native promoters were derived. Foliar application of zinc reduces the risk of drought stress on poppy (Papaver somniferum L.)

Especially in arid regions, drought stress is considered as one of the main reasons for yield reduction of plant. Zinc, as one of the essential microelements in crop plant, plays a crucial role in resistance to drought stress. The objective of the vegetation experiment established in 2011-2015 on locality Zaboice was to explore the effect of the foliar zinc application on the yield of poppy in interactions with weather conditions observed years. Effect of zinc foliar application was significantly dependent on the average daily air temperature (r =-0.936). Foliar application of zinc increases production of poppy seed, in the range from 6.9 to 25.5 % and the efficiency of zinc foliar fertilization increases with a widening deficit rainfall calculated as the difference between the precipitation sum and reference evapotranspiration of poppy growing season (r =-0.9072). In general, the results of the present study indicate that usage of zinc foliar application reduces the harmful effects of water deficit stress and increases resistance to drought stress in poppy plant.
Evaluation of grain yield components in selected winter wheat cultivars

This thesis deals with the yield ability of chosen winter wheat varieties during growing season 2014/2015. Comparison of the two varieties (hybrid and linear). The experiment was established in standard and reduced seed rates and also in four replicates on the plot of Faculty of Agriculture, University of South Bohemia in České Budějovice. Were examined yield components: the amount of spikes per 1 m2, amount of grains per spike and weight of thousand grains. During the vegetation observed were the amount of plants per 1 m2, each growth phase, the number of stalks per 1 m2 and plant height. Furthermore, measured were amounts of units of chlorophyll in plants using device N-Tester and the SunScan device sunlight usable for photosynthesis of plants wherein the processed values of leaf area index (LAI). At the end, compared were the theoretical and real yield and evaluated were spike length, amount of spikelets in spike and density.
The application of AFLP fingerprinting in breeding of Brassica napus

AFLP markers are widely used in breeding in some other crops, but their utilization in breeding of Brassica crops is not so frequent. AFLP markers are used for molecular characterization of particular varieties or genotypes and for evaluation of genetic diversity. The aim of this thesis was the application of this method in breeding of rapeseed and in comparative study of genetic variability of different oil seed rape cultivars of Czech, Czechoslovak and German origin. AFLP is based on selective amplification of genomic DNA. Technique consists of three basic steps: 1. restriction of genomic DNA on smaller fragments and ligation of adaptors; 2. selective amplification of fragments with specific primers and 3. electrophoresis of products and computer analysis (Vos et al., 1995). In this thesis 16 Czech (CZ) and Czechoslovak (CZS) and 16 German (G) cultivars of oilseed rape were compared. Two specific primers, EcoRI AGC and fluorescence labeled primer MseI ACC, were chosen for AFLP method. It was possible to distinguish group of CZ/CZS and G cultivars, which have origin in gene resources from Czech Republic and Germany. AFLP technique allows detection of very small differences in Brassica genome. The differences between studied cultivars exist, but they are not very distinct. They show on certain relationship and reduction of genetic basis of genetic resources, which are used in Central Europe.
Influence of cultivars on distribution of starch granules
