Stress tester

The bachelor thesis deals with the issue of stress testing. The aim of this work is to extend the JMeter tool with amplification modules of DoS attacks. The beginning of the work introduces the issue of DoS attacks, along with a description of selected attacks. One of the points of the work is to verify the functionality and find errors of the stress tester, which is a comprehensive system for testing the information and communication infrastructure based on the JMeter tool. The last chapter deals with the implementation of DoS attacks.
Application for the Data Processing in the Area of Genome Engineering

This masters thesis has a few objectives. One of them is to acquaint with the problems of genome engineering, especially with fragmentation of DNA, the macromolecule DNA, the methods for purification and separation of the nucleic acids, the enzymes used for modification of these acids, amplification and get to know with cluster and gradient analysis as well. The next aim is to peruse the existed application and compare it to the layout of the proposed application, that is the third aim. The last one from the objectives is the implementation and the report how was the application tested by the real data. The results will be discussed as well as the possibilities of the further extension.
Significance of MLL gene aberrations in patients with acute myeloid leukemia

In acute myeloid leukemia (AML), predominantly in AML M5a, the most frequent recurrent aberration of chromosome 11 involves region 11q23. Molecular breakpoint studies of several translocations involving chromosomal band 11q23 led to the detection of a gene that was named MLL (myeloid/lymphoid leukemia). This gene is important for the proper HOX gene expression during ontogenesis and hematopoiesis. Chromosomal aberrations affecting the MLL gene occur in 5 - 10 % of AML cases and are very variable. Since that time, more than 70 different translocation partners of the MLL gene have been described. Aberrations of the MLL gene are associated with an aggresive type of the disease and its detection is needed for the treatment decision. Therefore, we investigated the occurrence of MLL abnormalities in bone marrow cells of the 66 newly diagnosed AML patients, using conventional cytogenetic and fluorescence in situ hybridization (FISH) analyses with a commercially available MLL Break Apart Rearrangement probe (Abbott VYSIS). Out of the 66 patients, we proved MLL abnormalities in 9 (13,6%): 5 (7,6%) showed translocation of MLL gene, in 3 (4,5%) we detected MLL gene amplification without any evidence of rearrangement and in 1 (1,5%) pacient only an extra copy of the MLL gene. The Bachelor´s thesis deals with the description of the techniques of nucleic acids sequencing. Based on an analysis and synthesis of findings in scientific resources, the characteristics of the methods, a brief history, advantages and disadvantages, possible modifications and future applications are described in the theoretical section, whereas special reference is placed on the principle on which these methods work. A special cha-pter is devoted to the Human Genome Project, which is considered the greatest milestone in the history of sequencing. In the practical section, animal genome of order Lepidoptera was sequenced. The entire process included DNA extraction, its purification, amplification, evaluation of quality on an agarose gel, sequencing and, last but not least, an analysis of the output data using specialized software.

The Use of the Amplified Accordion in the Contemporary Music

This bachelor thesis deals with the issues of the accordion amplification, comparing different creative and technical approaches: the ways of sound recording, different internal microphone systems and different concepts of music composition. Evaluation of the results of my work, will hopefully allow me to design and subsequently build a new internal microphone system for the live production.
Methods of Molecular Biology in Predictive Diagnostics of Her/neu in Stomach Cancer

Stomach cancer is a malignant neoplastic disease, which is caused by malignant transformation of the gastric mucosal epithelium. Elderly patients are more frequently affected, the majority of the patients are males. Stomach cancer has some typical features like minor symptomatology and early metastasis founding. Nowadays, a mild decrease of incidence is registered. Treatment of stomach cancer depends on its location and stage. Because of its relative chemo- and radioresistance, surgical resection is the only one potentially curable method, although there is high number of recurrences. An amplification and overexpression of HER2 receptor is detected in approximately 10 - 20 % patients with stomach cancer. This overexpression correlates with worse prognosis of disease. Treatment with a monoclonal antibody named trastuzumab, Herceptin® could significantly elongate their life. Trastuzumab is now widely and successfully used for the treatment of female patients with breast carcinoma. Detection of HER2 amplification is there needed. Treatment with trastuzumab can be useful only when HER2 positivity of tumor is determined. This determination should be made by a reliable test with sufficient sensitivity and specificity.
