Testing of the possible applications of the flow-through dual amperometric detector

In this work, the possibilities of using an electrochemical dual detector in a flow arrangement were investigated using flow injection analysis. The dual detector was first tested for potassium ferrocyanide for its reversible oxidation at the generation electrode and subsequent reduction at the detection electrode. The possibilities of determination of nitro substances were explored, when hydroxylamine was formed by the reduction of nitro groups on the generation electrode, which was subsequently oxidized on the detection electrode. Thanks to determination in the oxidation potentials, it was possible to avoid the noise caused by oxygen reduction and the detection limit for dinitrophenol 0.47 μmol∙dm−3 was achieved. The possibilities of using a dual detector for detection in reduction potentials by minimizing the influence of oxygen, by reducing it on the generation electrode, were verified. Furthermore, the possibility of using a dual detector for the determination of dopamine in the presence of ascorbic acid based on the reversible oxidation of dopamine and the irreversible oxidation of ascorbic acid was investigated. Creation of electrochemical sensors for DNA analysis

The objective of this work was creation of an electrochemical sensor for DNA analysis using thick layer technology. Development of sensors is described from the design face with particular steps of realization along with theory. Measurements with results and evaluation of possible application in common use are described.
Electrochemical sensor for ascorbic acid detection

This work deals with electrochemical detection of ascorbic acid using screen-printed three-electrode sensors. The work is divided into three parts, where the first describes the theory of thick-film technology, including the types and compositions of the pastes, fabrication and firing. The second part deals with the methods, measurement principles and the electrochemical detection of ascorbic acid. Experiments were done using screen-printed carbon electrodes in a solution of phosphate buffer and ascorbic acid. In conclusion the results are evaluated.

Biosensors based on modified carbon nanotubes

The aim of this work is to describe biosensors for detection of substances in liquids using modified carbon nanotubes. This work reports on knowledge about nanotubes and electrochemical analysis methods which were used. The matter of direct electrochemical detection of insulin using CNTs modified thick layer planar carbon nanotubes is discussed. The elementary working electrodes were created by using of commercial carbon paste and were modified by thin film CNTs, CNTs/ruthenium oxide and CNTs/chitosan then. The best results of the insulin detection were achieved with the working electrodes modified by low concentration non-purified CNTs which had low elementary response in electrolyte and good response to increasing concentration of insulin in the concentration range from 0, 25 to 10 mol/L. The other modifications caused increasing of the electrode elementary response, but they did not significantly affect the detection.
Testing of the electrochemical properties of flow-through diamond detector

In the presented bachelor thesis, the functionality of a flow-through multitubular boron- doped diamond electrode (mtBDDE) was verified on two selected neurotransmitters, DA and NE, using the HPLC technique with isocratic elution. An aqueous solution of Britton-Robinson (B-R) buffer was chosen as the mobile phase and a Kinetex EVO C18 reverse column (150 x 4.6 mm) as the stationary phase. The flow rate of the mobile phase was set at 1 ml/min. For electrochemical detection, mtBDDE was used and UV detection at 280 nm was applied as an assistant to the main detector. The electrochemical response of the DA and NE solution (cΘ = 1.10-4 mol/dm3 ) was investigated in the form of hydrodynamic voltammograms over a potential range from 0.1 V to 1.3 V in different pH ranges of the mobile phase (2.5; 4.0; 6.0; 8.0 and 10.0). It was found that the potential wave shifts to lower potentials with increasing pH of the mobile phase, while the response of individual analytes does not change significantly with pH. Under optimal conditions, i.e., mobile phase B-R buffer at pH = 6 and a potential loaded on the mtBDDE of 0.6 V, the repeatability and reproducibility of the measurement results were verified. Voltammetric and amperometric determination of homovanillic, vanillylmandelic, and 5-hydroxyindole-3-acetic acid

Presented dissertation thesis is focused on the development of electrochemical methods for the determination of three important tumour biomarkers, namely homovanillic acid (HVA), vanillylmandelic acid (VMA), and 5-hydroxyindole-3-acetic acid (5-HIAA). First part of the study is focused on electrochemical behaviour of these analytes in batch arrangement using differential pulse voltammetry (DPV) at screen-printed carbon electrodes (SPCEs). It has been proved that presented method is sufficiently sensitive for monitoring above mentioned analytes. Moreover, it can be used for determination of HVA and VMA in mixture. Obtained limits of detection (LODs) were 0.24 µmol·L-1 for HVA, 0.06 µmol·L-1 for VMA, and 0.12 µmol·L-1 for 5-HIAA. The requirements to speed up the analysis and at the same time to reduce its price initialized our study of the determination of tested biomarkers in flow systems. Firstly, flow injection analysis with amperometric detection was investigated for the determination of all three biomarkers at the same SPCE, and then an analogous determination of structural more similar pair, HVA and VMA, was performed at a boron doped diamond electrode (BDDE). Preparation and Characterization of Silver Amalgam Particles on Gold Electrode Surface

This work is focused on the preparation of silver amalgam particles by its direct electrodeposition on gold surfaces using double pulse chronoamperometry from solution containing soluble Ag+ and Hg2+ ionts. During the electrodepositions of silver amalgam particles, different: i) parameters of chronoamperometry, ii) Ag/Hg ratio in solution and iii) gold electrodes were studied. All the prepared amalgam particles were characterized by scanning electron microscope with energy dispersive X-ray spectroscopy and electrochemical methods. Voltammetric application of the gold electrodes decorated by sliver amalgam particles was confirmed by analysis of model organic nitrocompound using cyclic voltammetry.
