The modification of gut microbiota composition by dietary intervention: the effect of plant-based and western-type diet

This thesis explores the relationship between diet, gut microbiota, and metabolic health, with a particular focus on their association with non-communicable metabolic diseases (NCDs) such as obesity and type 2 diabetes (T2D). The aim of the first study is to assess compositional and metabolic differences in gut microbiota between healthy lean long-term vegans and omnivores. The study reveals that while the gut microbiota composition is not significantly different between the two groups, there are significant differences in the fecal, serum and urinary metabolome. These differences may be attributed to the different availability of substrates in the diet, as the vegan diet is associated with a shift from a proteolytic to a saccharolytic fermentation program. Our results support the hypothesis of both resilience and metabolic flexibility of the adult gut microbiota. In addition to taxonomic analyses, this dissertation also includes metabolomics to evaluate the functional manifestations of the gut microbiota. The bachelor thesis Nutrition of dairy cows in the transition period deals with the nutrition of dairy cows in the peripartum period with emphasis on the prevention of metabolic disorders. The basic feeds used for the nutrition of dairy cows are described from a single-souled perspective. Characteristics of essential nutrients. Phasic nutrition and metabolic disorders caused by poor nutrition. The actual work was based on information obtained from five farms. Information was obtained on feed ration, housing method, feeding technique and incidence of metabolic disorders. From the information obtained, it can be said that most of the selected farms choose the right methods and practices in feeding dairy cows in transit. However, deficiencies were found in two holdings based on a lack of knowledge of the nutrient composition of the feed and in one case an unbalanced ration. There are no serious problems related to metabolic disorders in any of the selected holdings.

In this bachelor thesis the basic factors are described related to calving, correct nutrition and lactation of the dairy cow. The individual parts of the digestive tract and the course of digestive physiology are mentioned in the introduction. Subsequently, a section is devoted to transition period and risks that surround it. A substantial part of the work is also summary of basic nutrients found in feeds and basic components included in cattle rations.
Role of antioxidant defense in the synthesis of antidiabetic lipokines

Fatty acid esters of hydroxy fatty acids (FAHFAs) are a recently discovered group of lipokines consisting of a fatty acid attached to a hydroxy fatty acid with an ester bond resulting in a diverse group of compounds with many different biological activities. Antidiabetic and anti-inflammatory activities are the most studied. The thesis aimed to study this group of bioactive lipids to elucidate the metabolism of FAHFAs and describe the role of antioxidant defense, namely peroxiredoxin 6 (Prdx6), in their biosynthesis with the help of isotopic labeling together with in vitro and in vivo experiments. All the samples, including white adipose tissue, liver, and even human breast milk, were subjected to untargeted or targeted lipidomic and metabolomics analysis using LC-MS/MS. We used the data from isotopically labeled experiments to describe the role of 5-PAHSA in glucose uptake and to show which specific pathways are stimulated by 5-PAHSA administration and which by insulin to compare the effect of both antidiabetic agents. Nutrition of the transition cow

This bachelor thesis titled Nutrition of the transition cow deals with nutrition of dairy cows in peripartum period with regard to prevention of development of metabolic diseases. Anatomy of digestive system and physiology of digestive processes are briefly described. Characteristic of nutrients and of the most common feeds used for nutrition of dairy cattle serves as introduction to formulation of dairy rations. Metabolic diseases caused by inadequate nutrition in transition period are the biggest problem in this period that farmer has to face to. In closing main rules of feeding dairy cows before and after calving, composition of dairy rations and recommended feeding additives are summarized.

The thesis deals with the most important inherited metabolic diseases in dogs. The thesis summarizes basic findings about eight inherited metabolic diseases, including pyruvate dehydrogenase phosphatase 1 deficiency (PDP1), phosphofructokinase deficiency (PFKD), pyruvate kinase deficiency (PK Def.), cystinuria, fucosidosis, L-2-HGA aciduria, GM1-gangliosidosis and copper toxicosis (CT). Essential characteristic, clinical view, genetic background and possibilities of current diagnostics and treatment are stated for each disease. Most of these disorders have the same genetic background and clinical symptoms like in the humans, which is why dogs serve as animal models for the development of the therapeutic methods for the human population.
