Use of biomarkers for assessing environmental pollution by platinum group metals

This diploma thesis in the theoretical part analyses the characteristic feature of individual platinum group elements, their toxicity, anthropogenic sources and the presence of platinum group elements in the environment of the Czech Republic. In addition, there are specified types of bioindicators focusing mainly on lichens, mosses and needles. There are described individual bioindication techniques and pre-treatment of samples for platinum metals determination in bioindicators. The subject of the experimental part is to verify the sorption efficiency and ability of platinum and palladium using the SPE method and to determine the actual platinum metal contamination in selected localities in the city of Brno. There was also the additionally subject to verify the sorption potential of selected bioindicators and determination of the environmental pollution by platinum metals using the bag technique. In the experimental part, the bioindicators of moss Pleurozium schreberi, lichen Xanthoria parietina, lichen Flavoparmelia caperata and needles Pinus nigra were used.
The presence of pollutants in matrices of bioindicators

This master‘s thesis concerns with occurrence of heavy metals in the environment, mainly in the matrices of bioindicators. Special attention is given to occurrence of mercury in the environment and its quantification in bioindicators. Mercury and its compounds are today considered as one of the most significant contaminants in the environment. Some sorts of mushrooms cumulate sizable amounts of mercury in their fruiting body and therefore we can use them to consider loading of different areas. Eight areas in Vizovice and its surroundings were chosen for these purposes and during the years 2006 – 2008 harvested 50 samples from 19 sorts of mushrooms from there. The concentrations of Hg were determined in all samples by single-purpose absorption spectrometer AMA 254. Comparison of the mercury content between individual sorts of mushrooms in the same area as well as comparison of loading of areas was performed and also bioaccumulation ability of individual sorts of mushrooms was determined.
Determination of selected xenobiotics in plant bioindicators

Monitoring of environment is basic aspect of detection of environment state. It is practice by analysis of water, air, soil and bioindicators. This thesis is aimed at elected bioindicators of plant origin, needles of pine-tree and spruce, and at determination of chosen xenobiotic, polybrominated diphenyl ethers (PBDE) in this bioindicators. PBDE are used as flame retardants and they are occurred in all components of environment, thanks to their properties and ability of bioaccumulation. In thesis is described separation of substances from sample using different extraction techniques and final gas chromatography. Experimental part is aimed at sample preparation, determination of chosen congeners PBDE and comparison of effectivity of used extraction techniques.
Possibilities of identification of essential oils in environmental matrixes

Contained substances type of essentials oils in samples of phytogenic are characterized in this thesis, namely in tree needles and in moss, theirs chemical fission, description effects on human health and theirs use in medicine. Thesis describes method of isolation and identification of essentials oils from plants. For isolation of essentials oils there are used methods solvent extraction and solid phase microextraction (SPME) and steam distillation, for identification there are thin layer chromatography (TLC) and gas chromatography (GC/MS).
Teaching of Hydrobiological Themes in the Natural Science at 2nd Grade of Elementary Schools in Central Bohemia

The diploma thesis is focusing on explaining the teaching of hydrobiological topics in natural science classes at the secondary level of primary schools, specifically in the Central Bohemian Region. The aim of the thesis is to: - introduce the issues of hydrobiological topics in natural science classes to the reader - to implement these topics into teaching at the secondary level of primary schools - to find out how these teachings are applied into practice on the secondary level of primary schools in the Central Bohemian Region - (and) to find out how much the students are interested in learning about hydrobiological topics The diploma thesis is divided into two parts, theoretical and practical. The theoretical part talks about hydrobiological theory, aquatic environment, and individual representatives of fauna and flora that occur in the studied area. It also explores the idea of anthropogenic activity (which has an impact on the aquatic environment), and the protection of waters and their habitats in the Czech Republic. The relation between the natural conditions and species composition and the prosperity of plant communities in situ is very close and suitable for monitoring potential degradation of individual natural components. In advanced science, this linkage also called bioindication, is widely used and it represents the effective and economically favourable method of pollution detection. The study of: Research of the possibility of using biomarkers for monitoring of landfills is focused of increasing knowledge about bioindication. The technical security of landfills now strictly implements the applicable laws and regulations, and therefore the risk of leakage of pollutants produced during the storage and subsequent decomposition of waste is very slight. Nevertheless the care of the possible risks of leakage of these substances and the subsequent contamination of the surrounding area must be taken. The research of verifying the possibility of using the vascular plants in monitoring the landfill was carried out in the years 2013-2015 on the landfill of municipal solid waste Petrůvky. During the three-year period, the reconnaissance of the terrain, and setting out five habitats for recording species composition and cover of vascular plants were carried out. On the basis of plant ecological requirements, and the incidence rate the bioindicative value was determined. The higher attention was paid to the plants that pointed to a higher nutrient capacity of habitat. Identified specially protected species: Filago lutescens, Veronica Dillenii, Myosurus minimus, Miosotis discolor, Ranunculus auricomus, Euphorbia amygdaloides and Filago arvensis, exhibited as important aspects for verifying the state of the environment were which indicated good natural conditons. Bioindicative properties of plant species were then compared with the results of the analysis of surface and underground water and soil, which were analyzed for a purpose of regular landfill monitoring. The phytocenological relevé created form the part of the analysed areas in 2009 was also used for the comparison of the results. The possibility of utilization of plant bio-indicators for monitoring the landfill based on performed research was determined.
