Expert activities in expropriation proceedings

This diploma thesis deals with expert activities, which are following the connection with expropriation process. The work is divided into three parts, each of them is targeted by specification and contant. First are analyzed the basic related concepts of this thesis, the continuence of expropriation process and historical background of expropriation. Attention is also paid to the protection and limitation of proprietary rights. The second part deals with expert activity and status in the expropriation process. The final part of the thesis is a purely practical one. The content is related to the creation of a practice expert judgement for expropriation purpose, from which describe its structure.

This bachelor's thesis is focused on evaluating the current system of remuneration of provident financial s.r.o. employees and on subsequent drafting of proposals for possible changes. The basis is a questionnaire survey among employees in order to find out their opinions and satisfaction with the current remuneration system leading to their satisfaction.
Comparison of knowledge of the lay and professional public about the activities of the Fire and Rescue Service of the Czech Republic within the integrated rescue system.

The Fire Brigade of the Czech Republic represents one of the key components of the integrated rescue system. In the Czech Republic, the Fire and Rescue Service is it's main coordinator. It's task is to ensure the coordinated action of the components of the integrated rescue system, both in preparation for emergencies and in the implementation of rescue and liquidation work. Together with other units, it ensures a continuous emergency response, whose task is to receive reports of emergencies, evaluate them and, if necessary, take urgent action when they occur. The aim of this thesis is to quantify the knowledge of the general public about the activities of the Fire Brigade of the Czech Republic within the framework of the integrated rescue system, to quantify the knowledge of the professional public about the activities of the Fire Brigade of the Czech Republic within the integrated rescue system and to make a comparison of both quantifications and to draw recommendations. The research was carried out by means of a quantitative research survey. A non- standardized questionnaire was chosen for the data collection technique. The non- standardized questionnaire was addressed to the general and professional public. The data collection was carried out among the general public in the author's immediate vicinity and among the professional public in the fire stations of the Znojmo Territorial Department, consisting of the Znojmo Fire Station, the Moravský Krumlov Fire Station and the Hrušovany nad Jevišovkou Fire Station. The data sets obtained were investigated using one-dimensional and two-dimensional statistics (non-parametric testing, two- choice parametric testing). The results of the investigation confirmed the hypotheses of research (the knowledge of the lay public has the character of a normal distribution, the knowledge of the experts is very close to the division of Poisson's, there is a statistically significant difference between the knowledge of the lay public and the experts at the level of 0,05). The information obtained identifies shortcomings of the general public in their knowledge of the scope of activities of the Fire Brigade of the Czech Republic within the integrated rescue system. This information can then be used to create information material that will focus on the lack of knowledge of the general public or for further follow-up work and deeper investigation of individual responses.
The quality of life of the patients after surgery of the dominant upper limbs

Human health is considered to be one of the most important values but it is often damaged. Often it is by an injury of the upper limb. Injuries of the upper limb may be caused by carelessness, inattention, or by a complete accident. However, there are also non-traumatic disorders of the upper limb that limit its functions, which are indicated for surgical treatment. On the upper limb, the most frequently area that undergoes surgery is the shoulder, elbow, wrist and hand. The surgery of the dominant upper limb and the associated subsequent convalescence brings patients complications such as movement restriction, pain, dependence on the environment, difficulties in self-service and household operation, inability to work, and can significantly affect the patient's quality of life, which can be changed for a short term, for a long term or permanently. The aim of this work is to analyze the opinions of patients, who have undergone a surgery of their dominant upper limb, the changes of their quality of life both in the short postoperative period and in the long time after. The research itself was carried out through a quantitative research. The chosen data collection technique was a non-standardized questionnaire. The non-standardized questionnaire was designed for patients who have undergone a dominant upper limb surgery. Information was collected in hospitals at the Department of Surgery, Orthopedics and Traumatology. The information obtained showed in what areas of life of patients after the operation of the dominant upper limb the quality of life changed and that it varied according to the location of the injury. It also revealed that the areas in which the quality of life of patients after surgery has changed according to sex. Attention was also paid to the importance of rehabilitation after surgery. The results of the work may contribute to improving the quality of life of patients with this type of injury.
The molecular and biochemical basis of primary coenzyme Q10 deficiency

This diploma thesis focus on disorders of coenzyme Q10, which lead to decrease of its concentration in certain patient`s tissues (CoQ10 deficiency). This thesis introduces two molecular-genetics methods for CoQ10 deficiency analysis. The first one is a screening method - High Resolution Melting Analysis, which is a useful tool for screening of a large group of patiens with possible mutations in PDSS1, PDSS2, COQ2, COQ9, CABC1 and APTX genes, which encode some enzymes of CoQ10 biosynthetic pathway. The second one is direct sequencing of coding sequences of these genes including intron sequences neighbouring exons. This work also describes a model of CoQ10 deficiency applied in vitro using cultured human skin fibroblasts and HEK293 cells. In this case CoQ10 deficiency is evoked by CoQ10 biosynthesis inhibitor - 4 aminobenzoic acid (concentration 1 mmol.dm-3) . 4 days treatment by 4 aminobenzoic acid caused decrease in CoQ10 level in fibroblasts and HEK293 cells (60 - 70 % of control, resp. 41 % of control in case of HEK293 cells), although cell viability and morfology of mitochondria remained unchanged. This thesis - Contrastive Topics in Czech - describes some of syntactic and semantic characteristics of the experessions, called Contrastive Topics. Following theory of Functional Generative Description (p. Sgall, E. Hajičová and J. Panevová), author is trying to Bnd a ways to explain and verify several hypotheses of FGD by searching data of Prague Dependency Treebank. Prague Dependency Treebank is collection of czech sentence s, annotated on three levels of depth, namely the morphemic layer, the surface shape of sentences and tectogrammatical layer. The deep syntactic structure annotations in the Prague Dependency Treebank, the so-called tectogrammatical tree structures, capture three basic aspects of the underlying structure of sentences: the dependency tree structure, the kinds of dependency syntactic relations, and the basic characteristics of the topicfocus articulation. PREIMPLANTATION GENETIC HAPLOTYPING IN GENETICALLY RISK FAMILIES Abstract of Irena Borgulova's PhD study Page 1/1 ABSTRACT Preimplantation genetic diagnosis (PGD) is at the intersection of assisted reproduction and clinical genetics. PGD precedes prenatal diagnosis because consists in biopsy of a single embryonic cell and its examination excluding genetic risks before embryo transfer back to mother uterus. Methods within PGD can offer all spectrums of possible investigations of a single cell, whether focused on monogenic disorders, chromosomal aberration or abnormality of whole genome. Monogenic diseases in embryos can be detected by direct or indirect linkage analysis. Indirect linkage analysis has the advantage compared to direct analysis that it is able to indentify pertinent aberration of examined chromosome. Indirect linkage analysis is characterised by preimplantation genetic haplotyping (PGH) which is prime and important constituent of PGD cycle. PGH is based on family anamnesis for determination of pathologic/ high-risk (mutation-associated) haplotype and healthy/ low-risk (without mutation) haplotype by comparison with the haplotypes of other family members. Dynamics of acrosome reaction during intra-specific sperm competition in rodents.

Dynamics of acrosome reaction during intra-specific sperm competition in rodents Sperm acrosome integrity is disturbed in promiscuous species field mice (Apodemus) and more than half of the spermatozoa undergoing spontaneous acrosome reaction (AR) before binding to the zona pellucida. In Muridae it is documented a generally high rate of spontaneous AR, and the percentage increases in promiscuous species up to 60 % in 60 min capacitation in vitro. The acrosome integrity positively corellates with presence of CD46 protein which absence in wood mouse is fenotypicaly same as in CD46 knock-out mouse leading to accelerated spontaneous AR. It is necessary to clarify whether for mouse sperm it is essential the primary binding of intact sperm to zona pellucida of the egg or whether it is preferred secondary sperm binding after spontaneous AR. In this context, the question is whether there is a relocalization of the key fusion protein IZUMO in sperm during spontaneous AR. IZUMO relocalization was monitored by immunofluorescence at specific times of capacitation in vitro during spontaneous and induced AR. Effect of cryopreservation on mouse sperm.

Cryopreservation or freezing of sperm in the reproductive biology is still actual topic. Today is the only method used for sperm storage, whether for the purposes of assisted reproduction, or for scientific purposes. However, this method has a negative impact on such stored cells and is therefore still a subject of many studies. Among the main causes of sperm damage in rodents there are inappropriately selected cryoprotective agents, poor or no elimination of oxidative stress generated during cryopreservation, as well as a poorly chosen speed and temperature of freezing. Correctly chosen procedures and the composition of the media in which sperm are stored, can significantly affect the quality of sperm. This bachelor theses focuses on the influence of cryopreservation on mouse spermatozoa. The main impact of this method includes the effect of freezing on sperm DNA, plasma membrane, acrosome, and sperm motility.

Průmysl 4.0 je velmi aktuální a často diskutované téma. Publikace Nové technologie a BOZP přináší čtenáři základní přehled o vybraných technologiích, které souvisejí s Průmyslem 4.0. Úvodem popisuje základní principy Průmyslu 4.0, které jsou uplatňované též ve vybraných oblastech pracovních činností. Pro účely této publikace bylo vybráno 5 oblastí pracovních činností, které jsou podle dostupných dat v České republice dlouhodobě považované za nejrizikovější. Jedná se konkrétně o oblast zpracovatelského průmyslu, logistiky, zemědělství, lesnictví a stavebnictví. Publikace obsahuje přehled technologií, které jsou v daných oblastech používané, případně přehled technologií, u kterých je v blízké budoucnosti možné očekávat jejich rozmach. Technologie, kterým byla pozornost věnována, by měly vést ke zvýšení bezpečnosti na pracovištích a zlepšení zdraví pracovníků. Opomenuta není ani oblast inteligentních osobních ochranných prostředků a chytrých pomůcek. Závěrem je nastíněné téma bezpečnosti na pracovišti související s využitím nových technologií a stručný právní úvod týkající se Průmyslu 4.0.
