The analysis of DNA isolated from different types of probiotic products using real-time PCR and HRM analysis

The aim of this diploma thesis was to introduce real-time PCR with high-resolution melting analysis for Bifidobacterium species. Currently a small number of publication, dealing with identification of Bifidobacterium species using high-resolution melting analysis, is available. According to publications dealing with identification of lactic acid bacteria were selected primers P1V1 and P2V1, LAC1 and LAC2, LsppUPF and LsppUPR, V3F and V3R, V6F and V6R. Using this primers bacterial DNA was amplified by real-time PCR with high-resolution melting analysis. After evaluation of the measured results efficiency of selected primers was verified on DNA izolated from complex sample of probiotic product. After further optimisation real-time PCR with high-resolution melting analysis could be suitable using selected primers for Bifidobacterium species.
Financial supervision in the EU - comparison of the CR and Great Britain

On the first April 2006 came to the turning point in the Czech Republic in the sphere of supervision and regulation over financial market, whereby approach more ideal European markets. In the master's thesis I focus on the present state of supervision and regulation of the financial market in CR, but also on system development before the 1. April 2006. I also deal with operating system on the financial market in Great Britain. The aim of my thesis is comparison of supervision and regulation in the Czech Republic and Great Britain.
Pathophysiological mechanisms of chronic kidney disease progression

The kidneys are an essential organ that maintains the homeostasis of body fluid and ions, it filters metabolites and plays important endocrine role. Renal function is controled by many mechanisms with a close interaction that can work independently without central control. These mechanisms are regulated by both systemic and intrarenal humoral systems and their physiological balance sustains the optimal kidney fuction. Any disturbance of this balance leads to the impairments of renal function and progression of renal injury and thus the kidney works inappropriately. The renal insufficiency and diseases remain significant problem despite modern medicine. This is results of several risk factors such as age, genetic predisposition and low birth weight, increase of civilization diseases - diabetes, hypertension, autoimmune disorders, but also bad habits such as unhealthy lifestyle, smoking etc. These factors can lead to the acute or chronic kidney diseases often without known cause. Than the question is an early diagnosis and optimal treatment to preserve kidney function and stop the progression of terminal renal damage. Sedláková, L. CSR Concept of the company INDUS, limited liability company with the catchment for the Vysočina region. Diploma thesis: Brno: Mendel University in Br-no, 2019. In this Diploma thesis there will be examined the actual situation of utilization of the CSR concept, it's efficiency and there will also be proposed concrete measures for improving the situation of employees of the company in terms of using CSR social pillar. The result of this dissertation based on the qualitative and quantitati-ve research will be the summary of the actual situation and also the summary of company plans for the utilization of this concept and then the design part taking into account needs and the strongest motives of the INDUS employees. Within the competition, the analysis of the industry will be prepared to compare the compa-ny's situation on the market of private security agencies.
Working morality and ethical issues at workplaces

The aim of this bachelor thesis is to find out which ethical problems are most often encountered by employees of large manufacturing companies and how these ethical problems affect their work and personal life. The theoretical part will deal with defining ethical problems and explaining basic terminology. The practical part will be realized in the form of quantitative research, which will be based on an anonymous questionnaire survey. Respondents will be employees of large manufacturing companies in the South Bohemian Region. The conclusion of the thesis will be devoted to the concrete story of the victim of unethical behavior and also suggestions to solve this problems.
Tourism in rural space - Case study of the Havlíčkova Borová village

This thesis deals with tourism in the rural area, which can be divided into various forms and kinds. Main object of this work is to chart region of Havlíčkův Brod as a tourist destination and to analyze situation in detail by form of a case study in the municipality of Havlíčkova Borová. In the theoretic part there is explanation of terms from professional literature concerning provincial tourism, kinds and forms in rural area. There is following characteristics of Region Vysočina and region of Havlíčkův Brod. In practical part there is description of selected municipality, then practical part rates its potential for activities of provincial tourism and compares data acquired by questionnaire survey. On basis of this research there were opinions and attitudes of local people found out and consequently there was rated if tourism in this municipality is used well.
Onemocnění pohybového aparátu u hříbat a mladých koní

The thesis deals with diseases of muskuloskeletal systém of foals and young horses, concretly then osteochondrosis, flexural and angular deformities, tendon laxity and physitis. The method is based on case studies and evaluation questionnaire among horse breeders. On the basis of these data was determined preventive recommendations to farmers. Given that all these diseases are multifactorial in nature and their etiology is similar, are named as developmental orthopedic disease. The main causes are genetics, nutrition, exercise management and growth rate of the individuals who are directly related to the causes mentioned. Clinical symptoms vary widely depending on severity. Likewise therapy often is not needed, but may occur also to cases which are very important to today's intractable methods. The aim is to provide recommendations to farmers how to recognize the severity of the disease and how to avoid them.
Effect of combined endothelin and renin-angiotensin systems blockade on blood pressure and chronic kidney disease regression in model of angiotensin II-dependent hypertension

and key words Enhanced activation of renin-angiotensin system (RAS) and endothelin system (ES) plays the key pathophysiological role in the progression of hypertension and the chronic kidney disease (CKD). The aim of this study was to verify wheter the combined inhibition RAS and selective inhibition ETA receptor for endothelin 1 (ET-1) will show additive renoprotective effects in experimental model CKD. This therapeutic aproach was tested on the transgenic rats with mouse renin gen (TGR), to whome ablation nephrectomy (5/6 NX) was done in the age of 6 weeks. After next 6 weeks the relevant treatment was given in drinking-water: dual RAS blockade (trandolapril 6mg/L + losartan 100mg/L) or the combination of dual RAS blockade + inhibitor of ETA receptor (atrasentan 25mg/L). Results of the first series show 100 % mortality in untreated rats with 5/6 NX to the 30th week. Both type of treatments increased the survival rate up to 30 % in 5/6 NX TGR after the 50th week. In the second series influence of treatments on the blood pressure (BP) was monitored in 5/6 NX TGR, which had systolic BP over 210 mmHg. Both treatments decreased BP to the level normotensive rats and reduced heart hypertrophy.
