Effects of light on the growth and miliacin content in common millet

This diploma thesis focuses on the effect of light on the growth and content of miliacin in millet panicum (Panicum milliaceum L.). A field trial on the ground of the Faculty of Agriculture and Technology at the University of South Bohemia in České Budějovice was set up with varieties Unikum, Kornberg and Hanácka Mana. Plant samples were taken at the time of clustering and maturity, and the height and weight of individual parts were determined together with the content of miliacin. Next, a lab trial with Unikum variety was set up, when the seeds were germinated for 11 days under the different wavelengths of colored spectrum of visible light and then the germination rate, the length and weight of the obtained sprouts were determined, and the analysis of the miliacin content was performed. The amount of miliacin in a dry matter was determined using gas chromatography coupled to mass spectrometry.
Evaluation of qualitative and quantitative parameters of entomopathogenic fungus \kur{Beauveria bassiana} related to the inoculum production temperature

Bachelor thesis is focusing on the entomopathogenic fungus Beauveria bassiana strain GHA, which is contained in the commercial bioproduct Botanigard. Botanigard is a product used in biological control in many countries around the world to control a wide range of pests of various plant species. In this thesis, "in vitro" (radial growth, spore production) and "in vivo" parameters (virulence) of the GHA strain of Beauveria bassiana were tested at various temperatures on different nutrient media. The most optimal temperature for radial growth of the GHA strain was 25 °C, on media with a nitrogen to carbon ratio of 20:20. The GHA strain produced the largest amount of conidia on the medium with a nitrogen/carbon ratio of 20:40 at temperature 25 °C. On the contrary, the smallest conidia production was obtained on medium PDA and nitrogen/carbon medium in ratio 5:10. The temperature of 25 °C was also the most optimal for the infection of larvae of Tenebrio molitor. Thesis also deals with the comparison of blastospores production of GHA strain with other strains of B. bassiana isolated from different geographical areas. The strain isolated in Argentina had the highest production of blastospores on MEB medium.

Anna Rejtarová Title: Possibilities of Occupational Therapy Intervention in Patients after Acquired Brain Injury with Apraxia Abstract of the Thesis: The bachelor's thesis is devoted to the topic of apraxia and the possibilities of interventions for patients with apraxia after acquired brain damage. Apraxia is a disorder of motor planning, and this condition affects the performance of activities that the patient was able to manage before the acquired brain damage. This often limits his self-sufficiency and quality of life. With the help of special interventions, it is possible to help to recover faster and return to normal life. The thesis contains theoretical and practical part. The theoretical part focuses on acquired brain damage, in this case cerebrovascular accidents, the neurological aspect of apraxia and then to a greater extent on the types of apraxia, evaluations and the possibilities of intervention. Kateřina Rybářová Title: The Inter-rater Reliability of the Nine Hole Peg Test in Patients with Multiple Sclerosis Abstract: Introduction: The Nine Hole Peg Test (NHPT) is used by occupational therapist as a standardized tool which evaluates fine motor skills and manual dexterity. The aim of this thesis was to establish the inter-rater reliability and variability of the results of the NHPT administered according to the Czech extended version of the manual in patients with multiple sclerosis (MS). Methods: 33 probands with MS (27 females, 6 males) were tested by NHPT. Video recordings taken during testing were analyzed by two independent raters. Pearson correlation coefficient was used for statistical data processing. Recording sheets from the testing were analyzed in order to determine invalidated trials for each subtest and reasons for the invalidation. Results: A very high positive correlation was established for inter-rater reliability (0,999902 to 0,999965) and variability of the results (0,999902 to 0,999965). The invalidation of trials was most frequent in the 2nd trial of the dominant hand (24.24% of 33 trials). The least frequent was in the 3rd trial of the non-dominant hand (6.06%). Title: Pilot Usability Testing of the Box and Block Test in Occupational Therapy by People after Stroke: Creating a Video Manual Abstract: Stroke often results in impaired upper limb function. Occupational therapists use standardized tests to objectively assess hand dexterity. An example is Box and Block Test (BBT), which assesses mainly fine and gross motor skills. The tested subject's task is to sequentially move as many blocks as possible from one side of the test box over partition to the other side using only the tested limb within one minute. For this test, only working versions of Czech translations of instructions by Mathiowetz et al. from 1985 were used in the Czech Republic. Therefore, in 2021 Rybářová et al. created Extended Czech Version of Manual for BBT with instructions in Czech. It also contains new more detailed rules that should ensure greater objectivity in the standardised implementation of BBT. The name of the thesis: Interrater Reliability of the Box and Block Test in People after Stroke Introduction: Czech extended version of the Box and Block Test (BBT) manual has been developed recently, thus the reliability needed to be reestablished. The BBT is often used in occupational therapy. Aim: The main aim is to determine the interrater reliability of BBT administered according to this manual in people after stroke. A secondary objective is to determine the variability of BBT results measured by the same rater. The research question is to determine the percentage agreement between the two raters. Methodology: 20 people after stroke were tested using the BBT. A video was made of the attendance testing. This was independently scored by two raters, A and B, whose scores were correlated to determine interrater reliability. The percentage agreement of these values was also determined. Values measured by the same rater from the attendance testing and from the video were correlated to determine the variability of the results. Title: Inter-rater Reliability of the Purdue Pegboard Test in Adults after a Stroke Abstract: The Purdue Pegboard Test (PPT) is a standardized tool which evaluates fine motor skills, finger and hand dexterity. This test is used to assess and evaluate not only neurological patients. The official Czech translation of the manual has not been available in the Czech Republic yet. To unify the administration of PPT, a new Czech extended version of the manual for PPT: Model 32020A was created. It includes new evaluation rules, unification of scoring and interpretation of tested individuals' performance and also provides exact instructions for administration of recommended three trials of each subtest. The main aim of this study was to determine inter-rater reliability of the PPT administered according to new Czech extended manual version. Therefore, 31 Czech-speaking adult patients after a stroke were tested by PPT at the Rehabilitation Center Kladruby. The evaluation of these patients, based on the analysis of the video documentation obtained from their testing, was performed by two independent occupational therapists. Title: Creating a Video Manual of Proper Administration of the Purdue Pegboard Test According to the Czech Extended Version of the Manual for the Purdue Pegboard Test (PPT): Model 32020A Abstract: A common consequence of a stroke is a fine motor skills dysfunction. The widely used Purdue Pegboard Test (PPT) measures the dexterity of the fingers of each hand separately or both together. The tested subject should insert as many components as possible into the holes in the test board during five time-limited subtests exactly according to the instructions written in the manual. In the Czech Republic, various working versions of the translation of the manual have been used so far, containing verbal instructions necessary for the administration of the test. In October 2021, the Czech Extended Version Manual for Purdue Pegboard Test (PPT): Model 32020A was published. In addition, it contains new rules for solving and evaluating situations (e.g. falling of a peg on the ground, changing hands during making assemblies), which therapists have not yet dealt with uniformly. Comprehensive Rehabilitation of a Child with Mild Clinical Course of Canavan Disease with the Emphasis on Intervention in Occupational Therapy

This Bachelor's thesis deals with rare Canavan disease. The aim of the thesis is to describe the disease generally as well as to show overall development of specific individual with Canavan disease focusing on rehabilitation, specifically occupational therapy, and to carry out comparison analysis between two individuals (Canavan disease child and his peer) resulting in occupational therapy plans. A complex view of Canavan disease knowledge and findings is described in the theoretical part, with a focus on a mild form. Moreover, there is an overview of rehabilitation in the field of metabolic diseases included. The occupational therapy within leukodystrophies is also summarized. Psychomotor development of specific individual with Canavan disease alongside with comprehensive rehabilitation overview is documented including detailed occupational therapy intervention in practical part of the thesis. Based on specific individual and his healthy peer skills diagnostics and comparison, goals are set in terms of both short-term and long-term occupational therapy plan with ambition of specific individual self-sufficiency maximization. Analysis of Painful Shoulder Therapy for Stroke Patients in Acute and Subacute Phase

Title: Analysis of Painful Shoulder Therapy for Stroke Patients in Acute and Subacute Phase Abstract: The aim of this thesis is to describe the possibilities of occupational therapy interventions aimed at preventing the development of painful shoulder syndrome or eliminating its manifestations. The theoretical part contains a summary and explanation of the basic concepts and description of individual facilitatory methods that occupational therapists can use when working with this syndrome. In the practical part there are four case reports of patients who have suffered a stroke and already been diagnosed with painful shoulder snydrome, or been suspected of developing this syndrome. The best methods for these patients were chosen and applied to affect the painful shoulder syndrome and the deficiencies that have arisen. According to the results, these patients have achieved a reduction in pain, increased self-sufficiency and quality of ADL and last but not least, an increase in range of motion, which also affects the quality of life.
