Study of the production of polyhydroxyalkanoates using selected thermophilic representatives of the genus Aneurinibacillus

The subject of the diploma thesis is the production of PHA copolymers by thermophilic Gram-positive bacteria of the genus Aneurinibacillus. The theoretical part is devoted to the general characterization of PHA materials, their properties, use, and also to the production of PHA polymers by these bacteria. The experimental part deals with the production of PHA copolymers using selected thermophilic members of the genus Aneurinibacillus. Firstly, the specific enzyme activity of PHA synthases was determined, and then the production of PHA copolymers was screened using selected lactones (-valerolactone, -hexalactone, -valerolactone) and diols (1,6-hexanediol, 2,3- butanediol and 1,4-butanediol). These experiments were performed with six thermophilic producers by the members of the genus Aneurinibacillus, which were isolated from compost and activated sludge. Tested microbial strains have demonstrated the ability to integrate interesting monomers into the PHA structure, including 4-hydroxyvalerate (represented up to 69.3% of the total PHA), 5-hydroxyvalerate (up to 47.1%), or 4-hydroxyhexanoate (up to 31.9%). Subsequently, a closer characterization of the obtained PHA (which were gained by the production of the best producers) using advanced methods (DSC, SEC-MALS, FT-IR) was performed. Finally, the screening of the PHA copolymers production was performed in laboratory bioreactors.
Screening of biosurfactant production in selected thermophilic bacteria

The subject of the bachelor thesis is the production of biosurfactants by selected thermophilic bacteria. The theoretical part contains a general characterization of thermophilic bacteria, describes the structures and classification of biosurfactants, characterizes their properties and uses. The final part of the theoretical part deals with the production of biosurfactants by thermophilic bacteria. The experimental part deals with the screening of biosurfactant production by selected thermophilic bacteria. Thermophilic bacteria were obtained from composts and activated sludges. Isolation is not a subject of the thesis. In total 39 bacterial isolates were cultivated and their supernatants were tested by screening methods – emulsification capacity assay and solubilization of crystalline anthracene. Based on the results of these methods, 14 most promising producers were selected. These bacterial strains were re-cultured to obtain a new set of supernatants. For more detailed screening of production by selected thermophilic bacteria, in addition to determining the activity of the emulsion and solubilization of crystalline anthracene, other methods were added: direct surface tension measurement – Du-Noüy-Ring, drop pouring method and oil spreading method. The results were evaluated and the measurements were repeated for a new set of supernatants to verify the reproducibility of the production. The results of all the tests were compared and then three most reliable thermophilic bacterial producers were determined.
Effect of natural substances from tea on G-quadruplexes and binding and transactivation properties of p53 protein

The tumor suppressor protein p53 belongs to the most important regulator of the cell cycle in case of DNA damage. Apart from stopping the cell cycle and inducing apoptosis, protein p53 interacts with other proteins and DNA. Mutation of the TP53 gene encoding the protein p53 are a common feature of human cancer. This Bachelor thesis investigates natural substances from tea which could play a positive role in the activation of protein p53 in cancer and be used to support the treatment of these diseases. In the theoretical part of the Bachelor thesis, the secondary structures of DNA were described, specifically the structure and properties of G-quadruplexes, as well as protein p53 and its binding activity to the secondary structure of DNA. Selected natural substances found in tea and food were described – gallic acid and apigenin. The aim of the experimental part of this work was to verify the ability of these substances to interact with G-quadruplexes in vitro and thus to stabilize them in the yeast model and to assess the subsequent effect on measuring the effect of protein p53 transactivation. The interaction of quaternary structures with G4 ligands was verified in vitro by using the ThT fluorescence assay and the luciferase reporter assay. It was found that G4 ligands at 30 M concentration after 20 hours of incubation did not show a significant effect on the tested yeast cultures. At a 60 M concentration of G4 ligands, an increase in protein p53 production was observed due to cellular stress caused by the presence of G4 ligands.

Revivalist, historian, apologist Matěj Procházka (1811-1889)

in English The following work is about Matěj Procházka (1811-1889), an important figure of national and ecclesiastical life in Moravia in 19th century and a member of the circle around Professor František Sušil, the-so-called Sušilova družina. The detailed biography presented here is the result of historical research into resource materials from several different archives in Moravia. This work examines three main periods of Procházka's life. He spent his childhood and student years, firstly, in his native town of Brtnice, then in Jihlava and Brno. After ordination, he served as a priest for sixteen years in different parishes of Brno diocese. Then he lived in Brno for nearly forty years while working as a secondary grammar school teacher and catechist and being involved in different activities, such as the Czech national revival, Catholic associations, education, and especially, publishing. This work maps the importance of the personage of Matěj Procházka as seen by his contemporaries and by following generations up to the present. It also brings out the overall characteristics of this priest. Knowledge about reproductive health at stage 2 of the selected primary school

This master thesis focuses on the knowledge of reproductive health among middle schoolers in the targeted middle schools in the Czech Republic. In the theoretical part of my work, I define the term of reproduction health, describe physiology of female and male genitals, disorders in reproductive health, and types of methods -natural invasive- offered by anticonception. Another part of my thesis concerns on reproductive health related to a prostitution, and a sexual abuse, and pregnancy impact on teenage girls' bodies during pubescence. I track the representation of teaching reproductive health in middle schools through RSVP and its main goals in certain areas and key competencies. The goal of my thesis was to map out the knowledge of reproductive health among the students who have been taught this knowledge through a class. As a research methodology, I used a survey that was distributed among middle schoolers in the sixth to eight grades. The results of my survey show that the eighth graders have the most amount of knowledge about reproductive health, but somehow still lower than one might expect. Level of motor ability and skills of children of older school age during the past three decades

The aim of the bachelor thesis was to compare sport performances of girls aged 11 to 18 years in Gymnasium Benešov during the past three decades. The objectives of the thesis were achieved based on the analysis of results in 10 athletic disciplines (50 m run, 60 m run, 100 m run, 800 m run, 1500 m run, 2660 m run, high jump, long jump, shot put and grenade throw) measured within the framework of physical education at gymnasium since 1988 to 2014. Data of 1083 students were used and the analysis of variance was performed. Based on the data evaluation it was found that the performance in runs at 60 m, 100 m, 800 m and 1500 m and long jump in older girls (16, 17 years old) were significantly reduced. The results also showed that the tendency towards reduced performance was more pronounced in long-lasting athletic disciplines and in older girls too.

This thesis analyses economy management of town Breclav. Aim is to evaluate the economy management of the town in period 2010 to 2014. Thesis is divided into literature review and own work. The literature review characterizes and explains basic concepts which that relate to municipalities and its management. The own work analyses assets, income and expenses. Incomes and expenses are concretely analyzed and thesis detects, which have an important role in the town budget. Farther financial analysis is accomplished through selected informative and monitoring indicators according to the methodology of the Ministry of Finance. The thesis brings debt service, indicator of the total (current) liquidity indicator and the share of liabilities to total assets. Based on the results, economy management of town Breclav can be described as very good. Thesis also includes proposal options, which can bring the development and better economy management of the town Breclav in the future.
