Squealing noise analysis from the tram transport and its influence on the environs

Bachelor’s thesis is focused on issues with evaluations of the effects of noises from tram transport. The part of the thesis is analysis of those issues, acquisition of data in locality Starý Lískovec tram loop; follow editing data and compilation of acoustic model. I’ll do the assessment results by compilation of acoustic situation in program SoundPlan. Last part of bachelor’s thesis is formulation of conclusions and recommendations for practice.
A comparison of the normal value of infrastructure easement to the prices determined by the internal regulations of municipalities

The thesis deals with the valuation of the servitude of the engineering network by normal price and based on the internal regulations of municipalities. The theoretical part focuses on the issues related to the easements, brief historical overview, structure, possibilities of the creation and termination, as well as the description and structure of the technical infrastructure with a focus on gas conduction. The last theoretical part contains of a description of the ways and methods of the valuation of the easements. In the practical part the actual valuation of the easements based on the internal regulations of municipalities and the own calculation of the normal price are carried out.
Contemporary Press Coverage of Student Protests at the Tiananmen Square

This bachelor thesis aims to analyse how three newspapers, namely Rudé právo, The Guardian and The New York Times, presented the student protests in Tiananmen Square in 1989. The period from 13 May 1989 to 26 June 1989 was chosen for the analysis, as it fully covers the situation preceding the massacre, the massacre of 4 June 1989 itslelf and its aftermath. In order to achieve the most accurate results, qualitative content analysis and so-called grounded theory will be used. The questions that will be answered through this thesis concern the specific aspects on which the individual newspapers placed emphasis, as well as how often the events were covered and with what temporal sequence, then which language features were used, and which reporting principles were applied within the news and journalistic texts. Based on the data collected, it was found that Rudé právo, ideologically charting communist mindset in China, held similar views to the Chinese Government, i.e. that the students attempted to overthrow the Government and the regime, and during the actual suppression of the protests, the soldiers were attacked by the students, not the other way around. The Guardian and The New York Times reported on the protests in a quite opposite way, regarding the protests as pro-democracy and the military...
Chalkbrood desease detection in honeybees

Chalkbrood disease is a disease caused by the fungal pathogen Ascophaera apis, which attacks and kills bee larvae. It´s most common in weak colonies ,that have a poor cleansing soil and don´t properly remove calcareous agents. The ideal solution is to exchange the bee mother, which will increase the cleansing instinct of the bees, which then completely removes the infection. The aim of my bachelor thesis was to master the methodology of the analysis of the occurrence of Ascosphaera apis using the PCR method, to acquire routine procedures used in the laboratory including DNA isolation and to elaborate a professional research on the topic. In the theoretical part of the thesis I describe the fungal disease of chalkbrood and its influence on the infected bee. Further, I describe in detail methods that are used for the detection of diseases such as DNA isolation, PCR and gel electrophoresis. In the practical part, which I did in the laboratories of the Biotechnology Center, which is part of the Department of Genetics and Plant Production at the Faculty of Agriculture of the University of South Bohemia in České Budějovice, I describe in detail the exact methods used in the detection of this disease and the evaluation of my own results.
Lyme borreliosis diagnostics using in vitro cellular immune response testing

Lyme borreliosis is a multisystemic disease affecting skin, joints, heart and central nervous system. The disease is caused by spirochetes of Borrelia burgdorferi sensu lato complex. These bacteria are spread by ticks of Ixodes genus. In 2016 there were almost 4,000 newly infected individuals reported in the Czech Republic. Contemporary serological diagnostics of Lyme borreliosis is not sensitive nor specific enough and does not even correlate with the pathology of the disease in the early or late phases. For the correct diagnosis of the disease it is necessary to detect the pathogen and its genotype. For this reason we had aimed at two goals. Through the digital droplet PCR (ddPCR) method we detected Borrelia-specific DNA and its genotype. The detection limit of borrelial DNA was set on gDNA samples isolated from the tick. Detection threshold for the initial amount of 1 ng of tick gDNA is at the range of 10-17 g of specific borrelial DNA. Borrelia spp. coinfection was detected in 5 out of 12 tested samples. The most frequent type was B. garinii which was detected in 5 samples. Production and purification of recombinant receptor Clrb

The Natural Killer (NK) cells play a vital role in the nonspecific immunity. They are capable of efficient immunity reaction without any antigen specific receptors on their surface. NK cells recognize non-self molecules and they also recognize their molecules serving as health markers, and absence of these molecules such as MHC glycoproteins on the target cell surface. The NK cells are able to recognize viral infection or tumor transformation in the organism. If natural killer cell is in contact with a cell carrying an abnormally low MHC class I glycoproteins, it will create a signal which informs the cell is infected with a virus. NK cells trigger apoptosis of the target cell without prior stimulation, proliferation and differentiation. They also promote inflammatory responses by the production of chemokines and cytokines. The response is always the interplay of activating and inhibitory signals that the cell receives from its surroundings. The latest research shows that the targeted modulation of NK cells leads to less complications in bone marrow transplantation. They can be potentially used in immunotherapy, e.g. in the treatment of autoimmune diseases. Therefore, NK cells are a highly-studied group of cells. Changes in folklore architecture in relation to modern and postmodern

The subject of my graduation theses was the culturologic analysis of relationship betweeen human, nature and architecture. In my conception the genesis and development of architecture can be explicated by the categories "nature - culture". My pursuit was the attempt of comprehension of the human tying up with nature and architecture by traditional roots. Therefore I primarily remitted folk architecture to culturologic analyse, the phenomenon that I consider to be peculiar and eternallly revitalizing world of buildings constructed within the scope of natural ecosystem. Afterwards I turned my attention to residence houses and garden-cities expressing on my oppinion the human desire for the harmony based on the consonance of nature and architecture. The thought of the present state and future of ecologic architecture as the conclusion of my pusuit. Processing mode of my graduation theses does not aspire after entire and exhaustive treatment of given object matter, but is rather led by the attempt to embrace "the dream of everlasting comeback" to the world of nature in the context of architecture.
