Ezrin-Radixin-Moesin (ERM) proteins: The link between tetraspanin web and cytoskeleton in gametes

Ezrin, which belongs to the ERM protein family along with radixin and moesin, plays an important role in linking membrane proteins to the actin cytoskeleton. In sperm, ezrin was described so far in bovine and humans, where it probably contributes in actin polymerization during capacitation. During the acrosome reaction, actin plays a significant role in relocation of protein from the inner acrosomal membrane to the equatorial segment, which is an essential process for successful fertilization. One of the proteins that relocates to the equatorial segment, the site of initiation of gamete fusion, is protein CD9. This protein probably stabilizes protein interactions between sperm and oocyte during gamete fusion. In this diploma thesis, we focused on the role of ezrin in linking the CD9 protein to the actin cytoskeleton in mouse sperm. Using indirect immunofluorescence, we detected ezrin, CD9 and actin in acrosome intact mouse sperm. The co-localization of these proteins in the apical acrosomal region suggests their interactions. However, this was not confirmed by our co-immunoprecipitation experiments. Acne Therapy: Comparison of Treatment and Patients' Experience

In theoretical part, basic facts about acne together with current pharmacological options of its treatment have been summarised. The non-pharmacological methods have been covered as well. In experimental work, data (the occurrence of acne in the population in relation to sex, environment, season and the therapy used) obtained from patients' health cards, and information obtained from the patients through a questionnaire have been evaluated and compared.
The acrosome reaction in mammalian sperm

The acrosome reaction (AR) of the sperm is a prerequisite for egg fertilization, which takes place in the female reproductive tract. The AR allows sperm to penetrate extracellular egg coats and fuse with the egg. At first, the sperm must undergo the process called capacitation, then AR is initiated and acrosomal content is released. While it is not clear, what initiates the AR, it is probably the egg's extracellular coats - the zona pellucida and cumulus cells, secreting progesterone and some other substances, which can initiate the AR. Lately, it was demonstrated, that in the mouse the most sperms undergo the AR in the upper isthmus of the oviduct. Only a few sperms reach the ampulla, but all of them can fertilize eggs. During the AR, the acrosomal content is released into the extracellular space. It was discovered, that the release of acrosomal proteins is not synchronous, soluble components are released faster from the acrosome than are acrosomal matrix proteins. Before the acrosomal release, G-coupled receptors and tyrosine kinase receptors activate phospholipases. Protein kinases are also activated, which results in the opening of Ca2+ channels in the acrosome and sperm plasma membrane and the release of Ca2+ . My thesis deals with aspects of long-term intensive care. The goal of my thesis is to determine the basic needs of patients and family preparedness aspects of intensive home care. Other stated goals is find out the possibility of returning patients to home care and also find out what is the use of basal stimulation in long-term intensive care department. In the theoretical part of my thesis are included the chapters about definition of intensive care and home intensive care, for the full completeness is here the chapter about the state which are requesting the intensive care. The main part is dedicated to extensive immobilization syndrome in intensive care. An integral part is also a chapter discussing the basal therapy. For an easy transition to the empirical part is included a chapter outlining the nursing model according to Majora Gordon The empirical part of my thesis contains three detailed studies of nursing.Nursing case studies are carried out in intensive care department Vršovická, in chronic intensive care department of Motol hospital and long-term intensive care department of the Pardubice region, in Chrudim hospital. The adolescents' awareness about sexually transmitted diseases.

The bachelor's thesis is focused on the adolescent's awareness of sexually transmitted diseases. This thesis is theoretical and empirical. The theoretical part briefly characterizes the period of adolescence, adolescent relationship with others, historical changes and equally important moral development. The following chapter is focused on sexual diseases. It is individual subsections contain information on specific sexually transmitted diseases and diagnosis. The last two chapters deal with important terms namely HIV infection and sex education. Empirical part is implemented using questionnaires, which was administered at four schools and there is a random selection of respondents from the population. The survey was intended to explain adolescents' awareness of sexually transmitted diseases, their symptoms and the attractiveness of protection against sexually transmitted diseases.
