Minors' Experience Analysis of Romanticized or Sexualized Contact with an Educator
Mašková, Kateřina ; Onder, Jakub (advisor) ; Krejčí, Josef (referee)
The diploma thesis deals with the issue of romanticized and sexualized contact of minor students and their adult educators which is being dominantly silenced in the Czech Republic. The thesis consists of a theoretical part and an empirical part. Theoretical part of the thesis presents psychological aspects of the developmental period of adolescence and of the pedagogical profession, discusses the issue of crossing professional boundaries by an educator and finally defines a specific subtype of educator sexual misconduct: educator sexual abuse of minor students in the form of a mutual sexualized type of a contact. The focus of the empirical part of the thesis is a research project framed by constructivist ontology, feminist epistemology and phenomenological metodology. The research project is an interpretative phenomenological analysis of the experience of five adult women who experienced romanticized or sexualized contact with their educator during adolescence. The purpose of the research was to gain an in-depth understanding of its participants' experiences both from their former adolescent perspective and their contemporary adult perspective and to capture any possible change of their mind during the given period concerning their own experiences. Currently, all five research participants consider...
Hypothetical protein Spr1962 as a new substrate of the signalling pathway of the Ser/Thr protein kinase StkP and phosphatase PhpP.
Mašková, Kateřina ; Ulrych, Aleš (advisor) ; Fišer, Radovan (referee)
Hypothetical protein Spr1962 as a new substrate of the signalling pathway of the Ser/Thr protein kinase StkP and phosphatase PhpP The extracellular human pathogen Streptococcus pneumoniae encodes in its genome only a eukaryotic-type serine/threonine protein kinase (StkP) and the corresponding PP2C phosphatase (PhpP) in its genome. This makes it a unique model organism for the studying of signaling pathways in bacteria. To date, several substrates of this signaling protein have been identified and characterized, including DivIVA, KhpB, FtsA, FtsZ, MacP, GlmM, GpsB, ComE and others. These proteins are involved in various cellular processes, including cell division and peptidoglycan biosynthesis. In a global phosphoproteome study based on the LC- MS approach, one of the potential new substrates was the hypothetical protein Spr1962, whose phosphorylation was detected only in the hyperphosphorylated strain with deleted PhpP phosphatase. The aim of this work was to characterize this new substrate Spr1962 and to clarify its possible function. Based on the significant structural similarity to Spr1962 the FloT flotillin protein of Bacillus subtilis, it was suggested that it might be a protein with an analogous function in pneumococci. Deletion of this gene in different genetic backrounds was found to cause an...

Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate: Kateřina Mašková Supervisor: PharmDr. Lucie Havlíková, Ph.D. Title of Diploma Thesis: Stress testing of estradiol and ethynylestradiol This diploma thesis is focused on the implementation of stress testing of two estrogenic hormones estradiol and ethynylestradiol. Both substances have been exposed to oxidation and acid and alkaline pH at increased temperature of 70řC for a predefined time period. The results of these tests should have shown a decrease or an increase of the peak area of estradiol and 17α- ethynylestradiol and a potential formation of degradation products. Peak areas and retention times of degradation products were observed by HPLC. The aim of this thesis was to determine the properties of these hormones under various stress conditions like extreme conditions of production, storage, transport and/or under various external influences. Characterization of degradation products and verification of the suitability of methods used for the analysis of degradation products were also aims of this paper. The method was partially validated. In the experimental part there has been found out that estradiol and ethynylestradiol were stable in water as well as in acetonitrile solution...
D.I. Fonvizin - the analysis of his dramas (drama "Brigadier")
Mašková, Kateřina ; Lendělová, Věra (advisor) ; Uličná, Olga (referee)
Tématem mé diplomové práce je analýza postav v komedii "Brigadýr" (1769) spisovatele D.l. Fonvizina, resp. analýza z mužské a ženské perspektivy v jejím prolínání i diferenciaci. Na drama jsem nahlížela především z hlediska mužských a ženských postav. Kromě toho nám jednotlivé příběhy a jazyková charakteristika hlavních aktérů komedie vypovídají mnohé o době, v níž autor žil a tvořil.
The influence of 3-month exercise program on postural stability in individuals after bariatric surgery
Mašková, Kateřina ; Daďová, Klára (advisor) ; Vilikus, Zdeněk (referee)
Title: The effect of movement program on static postural stability in individuals after bariatric surgery Aims: The aim of this diploma thesis was to find out whether a three-month regular exercise program including strenght and aerobic training has an impact on static postural stability in obese individuals who underwent bariatric surgery. Methodology: The special part was written on the basis of data obtained from posturographic measurement of obese patients who underwent bariatric surgery. The static postural stability was measured using pressure plate a MobileMat from Tekscan. Participants were measured before surgery and approximately 4 months after surgery. A total of 30 subjects aged 31-65 years with BMI values between 35-55 kg/m2 (obesity class II and III) who underwent initial and exit examination including stabilometry, were included in this diploma thesis. They were divided into 2 groups, first group participating in a three-month exercise program (n = 10; age 51 ± 7.3 years; BMI 41.7 ± 6.4 kg/m2 ) and a group undergoing bariatric surgery only (n = 20; age 48 ± 9, 4 years; BMI 43.6 ± 6.5 kg/m2 ). The exercise program included exercise three times a week: once a week one-hour strength training and one-hour aerobic group training, and at least once a week home intervention. Results: In...
The cell wall biosynthesis in gram-positive bacteria and inhibitory effect of antibiotics.
Mašková, Kateřina ; Ulrych, Aleš (advisor) ; Lichá, Irena (referee)
The cell wall of Gram-positive bacteria includes, in addition to the core peptidoglycan molecule, unique polysaccharides such as teichoic acids, capsular polysaccharides and exopolysaccharides, and covalently bound surface proteins. Together, they create a strong and durable layer that provides protection but also communication with the external environment. Peptidoglycan biosynthesis in Gram-positive bacteria can be divided into three phases: cytoplasmic, membrane and extracytoplasmic phase. The individual phases consist of specific reactions that are catalyzed by often conserved bacterial enzymes, which are potential targets for antibiotic molecules. Most known antibiotics effective against Gram-positive bacteria are aimed at inhibiting the process of cell wall synthesis. The mechanisms of action of individual antibiotics are described with varying degrees of detail. Some are known and widely used in medicine or veterinary practice, and some have so far only shown the potential to become drugs. Another use of antibiotics is in the basic research itself, especially in the study of cell wall biosynthesis and bacterial division. In this work, I have compiled a summary of knowledge about cell wall biosynthesis of Gram-positive bacteria and a list of antibiotics and a description of the mechanisms of...

The Diagnosis of Paranoid Schizophrenia from the Perspective of Parents of the Diagnosed Individuals
Mašková, Kateřina ; Šivicová, Gabriela (advisor) ; Onder, Jakub (referee)
In this bachelor's thesis, I deal with the experiences of parents whose descendants were diagnosed with paranoid schizophrenia. At the beginning of the theoretical part, I first describe several points of view which can be used when referring to psychotic states: a diagnostic perspective, non-diagnostic perspectives, and an anti-diagnostic perspective. Subsequently, I cover the diagnosis of paranoid schizophrenia both from the medical point of view, through its diagnostic manifestations, progressive forms and possibilities of treatment, and as a factor significantly affecting the quality of life. Furthermore, I focus on attitudes, emotions, behavioral patterns and needs of the parents of the diagnosed descendants. The theoretical part of the thesis is concluded with a chapter concerning with various options of psychosocial support for a family system one of whose members has got an experience with a psychotic episode. The empirical part is focused on the experiences of parents whose descendants were given the diagnosis in the age of fifteen or later. I conducted eight in-depth semi-structured interviews and for analyzing the obtained data I used the open coding method. As instances of the topics that the parents from my research sample reflected repeatedly, I would mention the disrespectful...
