IL-25: Novel target to treat allergic diseases
Lišková, Kateřina ; Hrdý, Jiří (advisor) ; Boháčová, Pavla (referee)
Interleukin (IL) 25 is a proinflammatory cytokine that promotes a Th2-type cell response. Its alternate name is IL-17E, and along with 5 other members, IL-25 belongs to to the family of cytokine IL-17. The family is based on the similarity of their amino acid sequences. The source and target cells of IL-25 include many different cell types. IL-25 is not only produced by many types of immune cells, but epithelial and Paneth cells are involved in its production as well. Its receptors form heterodimers composed of 2 subunits - IL-17RA and IL-17RB. Both receptor proteins are required for IL-25 mediated activities and occur in other IL-17 family members. IL-25 also plays an important role in allergies - one of the most common diseases in developed countries. Cytokine IL-25 has been studied primarily in asthma. However, other very common types of allergies, such as food allergies, atopic dermatitis and allergic rhinitis, can not be overlooked. Even in these cases, the role of IL-25 is not negligible and is studied. Based on the knowledge of IL-25 biology and its role in allergies, this cytokine may be an important therapeutic target in the treatment of allergic diseases. One possibility is, for example, the use of neutralizing antibody and subsequent blocking of IL-25 activity. Key words: interleukin-25,...
Crisis intervention via electronic communication (on-line chat, on-line videochat)
Čechová, Kateřina ; Havránková, Olga (advisor) ; Lišková, Kateřina (referee)
This diploma disertation deals with the topic of on-line crisis intervention (KI further on in the text) in the Czech Republic (CR), specifically the service functioning of the online chat and videochat. This topic has not been paid attention to so far. The topic concerned is a rather new service area which provides service to the clients in a difficult situation on the internet and which has been working for past few years in a few KI centres (workplaces). No professional has tried to search it systematically or describe it in more detail yet. Considering the fact the internet is a widely used medium in our population nowadays, a high future potential of these services can be expected in the field of psychosociology. Therefore this diploma disertation aims to give a detailed description of the on-line chat and videochat functioning and necessary connections at KI centres (workplaces) in order to create a quidebook to the basics of these services for specialists and amateurs who are interested in this topic. Secondly, this diploma disertation is written to inform about the process of implementing the on-line chat into KI centres (workplaces) at three contemporary providers in CR, and to make the readers understand what the whole process is about. At the same time, this is to compile all the data for a...
Study of decomposition of organic matter in soil using the method of "tea bags"
Lišková, Kateřina ; Sedláček, Petr (referee) ; Enev, Vojtěch (advisor)
The main aim of this master thesis was to introduce an optimal method which could be used for study of a decompostion of an organic matter in a soil. For this purpose was chosen based on a literaly review so-called tea bag method. With regard to the completeness of the work, a physico-chemical characterization of the soil environment and HS isolated from it was also performed. The degraded organic material was then further characterized using thermogravimetric analysis, elemental analysis, FT-IR spectroscopy, and the total content of water-extractable polyphenolic substances in the samples was also determined.
Teaching Mendelian inheritance by didactic game
Lišková, Kateřina ; Janštová, Vanda (advisor) ; Mourek, Jan (referee)
This diploma thesis focuses on the education of genetics to grammar schools and students' comprehension of genetics. This abstract scientific discipline is considered difficult to learn so I decided to prepare educational materials with the aim to improve students' understanding of the topic. The main aim of the thesis was to prepare and test a didactic game focused on Mendelian inheritance and compare the effectivity of this kind of interactive education compared with the classical explanatory style of education. Other aims included evaluating the influence of other variables; comparing the difficulty of individual terms in genetics and the connection within chosen triplet of terms; and identifying the most common misconceptions. The data was collected in four classes of upper graders at two grammar schools by newly prepared questionnaire focused on demographic and knowledge. The results showed that the didactic game was as efficient as the classical explanatory method at creating short term knowledge Students had a bigger problem with explaining the connection among the terms than defining the individual terms. The most complicated term reported by the students was chromosome. It was the only term in which there wasn't any improvement between pre-test and post-test. Part of this thesis is also...
Noun-verb distinction in Czech Sign Langugage
Lišková, Kateřina ; Adam, Robert (advisor) ; Macurová, Alena (referee)
Some signs denoting substances and signs denoting events are formally and semantically related in the Czech sign language. These signs can be distinguished by different parameters. This study looked at the parameters of the relative duration of a signing, the use of mouthing, the juxtaposition of signs and the frequency and mode of motion. Signs denoting substances and signs denoting events can be clearly distinguished by those features n Czech sign language. Key words Czech sign language, sign languages, nouns, verbs, noun-verb distinction, word classes
Value - based education using Method Persona Dolls
Lišková, Kateřina ; Poche Kargerová, Jana (advisor) ; Vondřičková, Klára (referee)
This thesis deals with Method Persona Dolls, which evolves children's approach and values, it is pre-eminently focused on empathy development, creativity, self-confidence, positive evaluation of others and forms of pro-social behaviour. The theoretical part is focused on the description of curriculum reforms and characteristic of curriculum documents in general, particularly on Framework Educational Programme for Basic Education. The theoretical part further deals with this method from the point of view of theoretical principles, methods of its use and contribution, but from the point of drawback as well, which can occur when using this method. The method is also analysed from the point of view of curriculum documents as a possible tool for key competencies development. In addition, it is concerned about emotional intelligence and grade-schoolers emotional development, values and emotion description. Furthermore, the concept of value-based education of Cyril Mooney will be concerned as next item. Both methods are compared and I search for possible linkage between Method Persona Dolls and strategies used in value education. The action research will form the research part of the thesis, in which the empathy development and communicative competence of pupils using the Persona Dolls Method is...
Isolation and fractionation methods for determination of humic substances content in Leonardite
Lišková, Kateřina ; Kalina, Michal (referee) ; Enev, Vojtěch (advisor)
The main aim of this thesis was to isolate humic substances (HA and FA) from the natural sample of Leonardite, to characterize their structure and physical and chemical properties. Among other things, the humic acid standard supplied by IHSS, isolated from Leonardite, originally from Dakota, USA, was also studied. The following thermal and spectrometric methods were used to characterize the samples of isolated humic substances: thermogravimetric analysis (TGA), elemental analysis (EA), ultraviolet-visible spectrometry (UV / Vis) and infrared spectrometry with Fourier transformation (FTIR). The atomic ratios were determined from the results of the elemental analysis. Absorption coefficients were calculated from the measured UV/Vis spectra and were used for the physicochemical characterization of these biocolloid compounds. The measured infrared spectra were used to identify the individual functional groups.
