66 Abstract - Flexible forms of working hours This diploma thesis deals with the flexible forms of working hours and its goal is to describe this issue in intelligible and comprehensive way. It is being very interesting and current theme which is to a great extent not subject to direct legal regulations and provides its contracting parties with a big amount of freedom of contract. This fact assists in bigger flexibilization of labour market and represents a significant instrument in the fight against unemployment and economic crisis. This thesis is composed of six chapters. Chapter One introduces the term flexible forms of working hours and briefly describes the content of following chapters. Chapter Two delimits the area of labour law in relation to other branches of law and describes law principles relating to labour law. The following Chapter Three deals with legal regulations of working hours, its length, ways of allotment and international legal regulations. Chapter Four represents an introduction to the flexible forms of working hours. The basic methods of division of flexibility in the relation to the labour market are mentioned here as well as flexible forms of work are divided on the basis of typology which is used in this diploma thesis. Characterization of active layers with pH dye for fluorescent pH fiber optic sensors

Diploma thesis is deal with the problem of plant intracellular pH measurement in range of pH5-pH7. Convenient method of pH measurement is in this case local optical microdetection using so call pH transducers immobilized into matrices. Ratio-metric method of measurement of emission of excited transducer on two wavelengths is used. Tree excitation wavelengths were chosen from measured absorption spectra of pH transducers which were solved in calibration buffers (liquid samples). Time independence calibration curves (dependence of ratio-coefficient on pH) of liquid samples were established from their emission spectra for transducers BCECF, HPTS and HPCE. The requirements of sensitivity in pH range satisfied BCECF and HPTS transducers. These transducers were immobilized into xero-gel layers (TEOS, APEX and PTES) and by dip-coating method coated on the glass slide (solid samples). The diference of pH values of control medium, which were read from the calibration curves of solid samples (BCECF + TEOS and HPTS + PTES) and from pH measured potentiometrically, were 0.73. The first test measurement of pH was made by the V-taper with deposited active layer (transducer BCECF immobilized in TEOS) on its tip.

Enzymatic Sensor of Biogenic Amines with Optical Oxygen Transducer

The biogenic amines (BA) are formed by the decarboxylation of amino acids. Because of their impact on human health, they have been used as markers for assessing the freshness and quality of a wide variety of protein-containing products. We studied biosensor for BA with an optical oxygen transducer that contained a diamine oxidase from pea and ruthenium complex. Both active components were incorporated in chemically stable polymer to form sensitive films thickness of 100 - 200 μm. With aim to optimize a composition and manufacturing of the biosensor we developer a mathematical model of processes inside the sensing film. The developed mathematical model revealed qualitative dependencies of the biosensor behavior (sensitivity, response time and dynamic range) on the (i) thickness of the sensitive layer, (ii) concentrations of substrates in the solution, (iii) enzyme activity in the layer and (iv) layer permeability for the substrates.
