GEMINI – secondary school for the disabled
Zástěrová, Šárka ; Kovář, Jan (referee) ; Guzdek, Adam (advisor)
The aim of this bachelor thesis is to modify the building of the GEMINI secondary school to meet the needs of students with disabilities. The area is located in Brno, in the Lesná district. The secondary school was furnished and put into operation within one week, so the current layout and equipment is not entirely suitable for the operation of a school for disabled people. Now the school consists of two separate buildings. The main two-storey school building, which is the subject of the structural and technical part of this bachelor thesis, is located in the former kindergarten building. Two entrance staircases are added to this building from the north-eastern side and a one-storey extension with an atrium from the south-western side. This main building is connected by a covered footbridge to the building of the heat exchange station, one quarter of which is now used by the school. The new design considers the removal of the extensions to the main building, but the concept of the entrance and single storey atrium extension is retained in the new plan. A new three-storey extension with a main vertical circulation space and glazed facade has been designed and more teaching space is designed in the new single storey atrium extension. A third storey extension can be used as student accommodation. The design also considers the use of half of the building of the heat exchange station for the operation of the school. As the school building does not have a suitable layout, the design only considers preserving the existing load-bearing structure and respecting the existing lining and lintels of the window openings in the wall. The new design is now intended to enable students to move and learn comfortably and to create a pleasant living environment.
Physics problems for radiology assistants
KOVÁŘ, Jan
The thesis was examining an opportunity to apply the curriculum process theorem in the matter of general and radiology physics and their problems' expression. The first objective is to create an educational text which would elucidate an indispensable theory for radiology assistant. The educational text is to involve problem expressions of needful physical fields. The second objective was to implement the curriculum process as a quiz and so to evaluate the familiarity among the professionals in the field. Furthermore, three hypotheses were formed. The first hypothesis presumed it is possible by applying the curriculum process to delimit the collection of physics problems that will be tailored to the needs and facilities of radiology assistants. The second hypothesis was divided into two subparts: A) By comparison of a physics structure and compounds of a radiology assistant profile it is possible to process a collection of problems dedicated to general physics. B) By comparison of a physics structure and compounds of a radiology assistant profile it is possible to process the collection of problems dedicated to radiology physics. The third formed hypothesis presumed the familiarity among responders regarding the field will resemble of normal distribution. By comparing the conceptual curriculum of physics and radiology, an educational text was created, which contained exemplary statements of general and radiological physics in relation to the components of the profile of the radiological assistant. By creating the text and defining example statements, the first goal of the work was fulfilled, and the first two hypotheses were accepted. Another application of the curricular process was to compile a test, the questions of which were based on the content of the educational text. Descriptive and mathematical statistics were used to appraise the research observing the test results. The outcome of the research acknowledged the third hypothesis. The second objective was reached by the test implementation. As a certain standardisation the second, previously used test was handed to the responders concomitantly to the first test. Via a parametric test the link between both tests results was confirmed. Following correlation analysis established the strong positive correlation. The results of the bachelor's thesis show that the created educational text together with a set of example statements can be effectively used to prepare students for the profession of radiological assistant.
The role of fructose in hepatic fat accumulation
Justrová, Marie ; Kovář, Jan (advisor) ; Malínská, Hana (referee)
One of the metabolic disorders with an increasing incidence is non-alcoholic fatty liver disease (NAFLD). NAFLD is characterized by pathological accumulation of fat in the liver. According to some authors, excessive fructose consumption plays an important role in its pathogenesis. The thesis aims to interpret published findings of the relationship between fructose intake and fat deposition in the liver. In the first part the fructose metabolism is described and compared with the glucose metabolism. The other effects of fructose on fat, carbohydrate and nucleotide metabolism are mentioned. This is followed by definition of the term NAFLD, description of various stages and clinical characterization of the disease. In the final part of the thesis the results of studies concerned with the role of excessive fructose consumption in NAFLD development are summarized. Excessive consumption of fructose by healthy subjects during isocaloric diet doesn't result in hepatic fat accumulation due to effective compensatory mechanisms. However, the role of fructose in hypercaloric diet in the increase of hepatic fat content hasn't been definitely explained yet. It seems that excessive caloric intake per se is more important than the effect of fructose or its metabolites. More pronounced rise in hepatic fat content...

Kovář, Jan ; Frýbort, Pavel (advisor) ; Kokštejn, Jakub (referee)
Title: Analysis of attacking playing systems Czech football team U19 on the elite qualification European championship. Objectives: In this bachelor thesis I will focus on the identification of attack systems in football and the detailed analysis of players' participation in shoot attempts. Method used: Observational analysis with the method of indirect observation. This is a joint research. Results: From indirect observation records I found out that the most used attack is fast attack (77,3%). Continual attack (13,3%) and combined attack (9,63%) was used rarely. The most attacking playing system that was finished was fast attack (23,1%). The most participate players at finished attacks were middle defenders, midfielders and forwards. Keywords: Fast attack, football, combined attack, continual attack
Regulation of triglyceride metabolism in circulation in postprandial phase.
Zemánková, Kateřina ; Kovář, Jan (advisor) ; Kazdová, Ludmila (referee) ; Zlatohlávek, Lukáš (referee)
Increased triglyceride (TG) concentration has been generally accepted as a risk factor for ischemic heart disease and, therefore, lowering TG is therapeutic target that should reduce cardiovascular disease risk. Traditionally, concentration of TG is measured in the fasting state (8-12 hours after an overnight fasting) mainly because the rise in TG levels after meal leads to the high variation in TG values. However, human beings spend larger portion of the day in a postprandial state and postprandial hypertriglyceridemia may then play a substantial role in determination of cardiovascular disease risk. The increased and prolonged postprandial lipemia has been found in patients with coronary heart disease. Moreover, recent data from Copenhagen Heart Study point out that the non-fasting TG concentration is associated with cardiovascular disease risk more tightly than the fasting TG concentration. Importantly, concentration of non-fasting TG is substantially affected by individual behavioural habits such as diet composition and physical activity. It remains to be determined whether it would be appropriate to identify individuals at higher risk of cardiovascular disease due to increased postprandial TG using tolerance test analogous to glucose tolerance test. The protocol of standardized fat tolerance...
Influence of tetrabromobiphenol A (TBBPA) on reproduction parameters, spermatogenesis and expression of selected genes in mice in vivo.
Žatecká, Eva ; Pěknicová, Jana (advisor) ; Kovář, Jan (referee)
Tetrabromobisphenol A is a substance that is widely used in industry as a fire retardant and is currently among the most widely used; its consumption is 210,000 tons / year and is still growing. It was shown that TBBPA can be released into the environment and it was found even in the human body. TBBPA has negative effects on different cell lines in vitro, including TM4 Sertoli cells. It was shown that it can affect the thyroid hormone system as well it might be able to influence the androgen / estrogen hormone system. Due to these facts, TBBPA was added to the list of potentially dangerous substances to human health. In this study, the effect of TBBPA on the reproductive parameters, spermatogenesis and activity of selected genes in mice in vivo was examined. First, we perform two generation in vivo study with outbred mouse line - CD1. In experimental animals we detected increased number of apoptotic cells in the testes, and also we observed changes in the expression pattern of selected genes encoding proteins that play an important role during spermatogenesis. In contrast, sperm quality and reproduction was not affected by TBBPA. As a complementary study was perform one generational in vivo study with inbred mouse line - C57BL/6. This study mostly confirmed the results obtained from previous in...
The role of caspase 2 in apoptosis induction in tumor cells.
Schmiedlová, Martina ; Kovář, Jan (advisor) ; Horníková, Lenka (referee)
Within the cell, caspase-2 probably fulfills several functions. Caspase-2 can be involved in apoptosis induction, DNA repair as well as cell cycle regulation. Caspase-2 has the character of initiator and also executioner caspase. A stimulus for caspase 2 activation can be oxidative stress or DNA damage. Caspase-2 is activated by cleavadge during an interaction with protein complexes. One of protein complexes,i.e. PIDDosome, is made of protein PIDD, RAIDD and pro-caspase-2. Withine the PIDDosome, caspase-2 is activated. Activated caspase-2 occures in a short S form and in long L form. L form of caspase-2 has proapoptotic effects and S form of caspase-2 has antiapoptotic effects. Caspase-2S has been only detected on mRNA level but not on protein level. The main role of caspase-2L is apoptosis induction in normal and tumor cells. Caspase-2 in tumour cells is activated by extrinstic as well as intristic apoptotic pathway. Apoptosis induction by caspase-2 is for example studied in connection with breast cancer treatment with taxanes. Caspase-2 ability of apoptosis induction in cancer cells independently of p53 protein is employed in cancer treatment including overcoming the resistance to apoptosis induction which is based on loosing p53 activity. Caspase-2 is involved in apoptosis induction by different...
