The master ‘s thesis focuses on the description and analysis of the ordering customer tools process in the company Mubea. The theoretical part is focused on processes, their improvement, description, and analysis. The practical part specifies the steps of the process and its shortcomings. The output is a process improvement proposal and evaluation of the benefits of the proposed changes.
Developmental disorders of the lens and anterior segment of the eye
Jedličková, Jana ; Ďuďáková, Ľubica (advisor) ; Šafka Brožková, Dana (referee) ; Cvekl, Ales (referee)
The anterior segment of the eye includes the eyelids, eyelashes, tear film, cornea, conjunctiva, iris, pupil, ciliary body, chamber angle, and lens. Cataracts and anterior segment dysgenesis are a highly heterogeneous group of diseases exhibiting all types of Mendelian inheritance. The aim of this thesis was molecular genetic analysis of 51 patients from 23 families with congenital and early onset cataracts and/or anterior segment dysgenesis, using a modern as well as conventional methods, such as next-generation sequencing, direct sequencing, bioinformatic and functional analyses including the use of artificial splicing vectors. We identified 24 causal variants in the coding, non-coding and regulatory regions of the PAX6, FTL, FOXC1, PITX2, FOXE3, and CHRDL1 genes. We confirmed the role of the n.37C>T variant in MIR204 in the development of chorioretinal dystrophy variably associated with iris coloboma, early-onset cataracts and congenital glaucoma. Splicing assay confirmed pathogenic effect of two mutations on pre-mRNA splicing of the PAX6 gene. Establishing molecular genetics diagnosis improves patient counselling and their relatives in terms of the prognosis and risk of developing secondary glaucoma Key words: molecular-genetic diagnosis, anterior segment dysgenesis, next-generation sequencing,...
Molecular analysis of Cornelia de Lange syndrom
Vokáčová, Markéta ; Schierová, Michaela (advisor) ; Jedličková, Jana (referee)
Cornelia de Lange syndrome is a rare, very heterogeneous, genetic disorder classified as a cohesinopathy. It is caused by mutations in genes that encode proteins of the cohesin complex or its regulators. So far, 5 major genes, whose defects are responsible for the syndrome, have been discovered. The mutated gene determines the type of heredity and, above all, the extent of the pathology. Due to dysfunction of the cohesin complex, not only the cohesion of sister chromatids is impaired, but also the regulation of gene expression and the repair of DNA double-strand breaks. Knowledge of molecular aspects of the disease has been enhanced by ongoing experiments with animal models of Cornelia de Lange syndrome, cell cultures from patients, and the yeast Saccharomyces cerevisiae. The detailed analysis of the biological functions of the cohesin complex may help to develop therapeutic methods. Keywords: Cornelia de Lange syndrome, cohesin complex, cohesinopathy, transcriptome change, genome integrity, topologically associating domains, NIPBL
Little Series in the Big Television - Czech Television's Online-only Content
Vašíčková, Dorota ; Szczepanik, Petr (advisor) ; Jedličková, Jana (referee)
Little Series in the Big Television: Czech Television's Online-only Content Bc. Dorota Vašíčková Abstract This master's thesis in the field of film and television studies maps the process and first results of the establishment of a new programme production line in Czech Television. The thesis applies autoethnographic methods to the authors' personal experience in order to map the changes in institutional settings resulting from the clash of public media with the realities of the internet age. The work consists of four topic-defined sections, each following the period between spring 2019 and spring 2022 when the institutional reorganization and reconstruction of the Czech Television VOD platform was taking place. The work follows on topics such as the public media values, principles of dramaturgy or the ideas of managerism, all analyzed on the background of three years of participant observation. A key role in the analysis of specific production lore is played by the possibilities granted to originally independent web-based formats by having institutional backing. This thesis aims to capture the reality of the only Czech public media television and to analyze, how an institution of the size of the Czech Television copes internally with the emergence and establishment of a new production line of its programmes.

Analysis of fluctuation in an international company
Jedličková, Jana ; Jarošová, Eva (advisor) ; Filipová, Alena (referee)
The goal of my diploma thesis is to find out the reasons of real and latent fluctuation of a specific team in an unknown international company. To reach the target I am linking up theoretical conclusions about employee's fluctuation and about other areas connected with that theme (i.e. company culture or selected personal processes) together with empirical research in the specific team. The reasons of real fluctuation are examined on former and leaving employees. Reasons of the latent fluctuation -- connected with gradual alienation leading to dissatisfaction and possible leave of the employee -- are examined thanks to five partial goals (selection of employees and psychological agreement; motivation and leadership; education and development of employees; reward system and career options). The real reasons of selected employees leaving the company are insufficient career options. Summary of findings about real and latent fluctuation and recommendations for team management are explained at the end of the study using SWOT analysis.

The thesis is also devoted to the life and works of františek Václav Míča who was the author of the first substantiated Czech opera About the origin of jaroměřice and it also touches the relevant theme of the role and assertion of the Czech lanquage in this place at the time preceding the revival era of the Czech celture.
