Theoretical study of enzymes related to carcinogenesis: DNA polymerase β and cytochromes P450

Present doctoral thesis contributed to understanding of mechanistic principles of two enzymes participating in the process of carcinogenesis; DNA polymerase  (pol ) and cytochromes P450 (CYP). Pol  is part of the DNA base-excision repair mechanism (BER). The primary role of pol  in, the BER mechanism, is inserting a new nucleotide into a DNA strand according to Watson-Crick base pairing rules. Pol  plays an important role in the process of carcinogenesis, approximately 30 % of human tumors express pol  mutants. The ability of pol  to discriminate between "right" and "wrong" nucleotide during the insertion process is called fidelity. We employed computational methods to elucidate molecular basis of the fidelity of pol . First, the relative free energy calculation method LRA was employed to compare differences in free energies between the "right" and "wrong" nucleotide during its insertion into DNA. The results indicated a better stabilization of transition-state of the nucleophilic substitution catalyzed by pol  in the case of the "right" versus "wrong" nucleotide. This difference resulted in an 80-fold contribution to its fidelity. Further, computational methods FEP and LIE were used to examine how mutations effect fidelity of pol . Metamorphic and structural record of the Gemer and Vepor Unit contact zone in the Central West Carpathians

The Central West Carpathians represents a crustal-scale thrust sheet stack formed by three major basement units, the Gemer, Vepor and Tatra. The basement of structurally middle Vepor Unit is dominated by Carboniferous granitoids while the upper Gemer Unit consists mainly of Lower Palaeozoic volcano-sedimentary complexes. Both units are overlain by Late Palaeozoic to Mesozoic cover sequences. During the Alpine orogenic event of Cretaceous age, the Vepor Unit was first buried during the northward overthrusting of the Gemer Unit and later exhumed during the eastward lateral escape of the Gemer. Alpine metamorphic conditions reached generally amphibolite facies conditions in the Vepor and greenschist facies conditions in the Gemer. In this work we concentrate on tectono-metamorphic record within the rock complexes incorporated into the imbricated structure of the Vepor-Gemer Contact Zone. The Vepor basement schists were affected by Variscan and Alpine regional metamorphism of medium grade. The age of metamorphism was confirmed by the Sm/Nd garnet dating (355±41 Ma - Variscan, 115±24 Ma -Alpine). Position of Saxonian granulite within the frame of European Variscides

The Saxonian Granulite Massif (the type granulite locality) is situated in the northwestern part of Bohemian Massif. It is an antiform with dome structure, which came to the surface from below SaxoThuringian Paleozoic units. It consits of high-pressure "Granulite core" with felsic and mafic granulites, which are surrounded by tectonic cover called "Schist mantle" of low-grade upper- crustal rocks. Massif is penetrated by several granite bodies. The Core and Mantle are separated by shear zone with a thickness of about 2 km. Geochronological dating of complex exhumation gives ages of about 340 Ma. The whole complex is affected by system of foliation and lineations. Massif was deeply buried, extremely heated and then exhumed with high exhumation rates during post collision extensional phase of the Variscan Orogeny.
Exhumation of mid-crustal rocks by a brittle-ductile unroofing mechanism, an example from the Veporic crystalline complex in the West Carpathians

Study of the structural and metamorphic record of the eastern part of contact zone within Vepor and Gemer units in Central West Carpathians has interpreted the burial and exhumation in the evolution of alpine orogeny. The studied area is built by granitoid rocks of Vepor basement and metapelites of cover sequences both Vepor and Gemer units. Three structural fabrics have been identified in the area, which are correlated with so far described deformation stages. Higher metamorphosed rocks with presence of two generation garnets were found out within lower metamorphosed metapelites of cover sequences. From the garnet - biotite thermometry the metamorphic conditions for the rims were established to 550-580řC. Whereas muscovite and in some case chlorite, are the only metamorphic phases in neighbouring cover rocks. Three generations of white micas are documented within structural fabrics. The oldest muscovites, magmatic in origin (Ms1), younger phengites (Phg) present in fabrics S1 and youngest muscovites (Ms2) present in fabric S2. This diploma thesis proposes the use of business intelligence working above complex economic system in a real joint-stock company for planning, cost management of cost centers and job orders and for control of managerial accounting in relation between cost centers and job orders. Furthermore thesis resolves the problems of data transition to the new information system to ensure the complexity of data entry to business intelligence.
