Motivation of children from children's home for studying and professional preparation

(in English): The thesis deals with the motivation of children from children's home for studying and preparation for their future profession, especially with the influence of educators on this motivation. The aim is to present to the readers how the education of children growing up in a children's home is carried out and by what it is affected. And to find out how educators influence children's motivation for studying and preparation for their career. The theoretical part describes the institutional care in the Czech Republic and its legislative basis. In more detail, the text then focuses on children's homes, the children who grow up there and the educators who work there. It also focuses on the motivation and education of children in children's homes by comparing the results of recent research studies. The empirical part is based on qualitative research. The results can serve as an incentive for more extensive research and at the same time help educators and other pedagogical workers in educating and motivating children in institutional care.

Roles of sperm morphology variation on proteomic content in muroid rodents

We can find a whole range of morphological differences between the spermatozoa of different animal species, whether at the level of shape, size or intracellular arrangement. It has been proven that these differences are likely to be driven by selective pressure caused by sperm competition. This can cause morphological adaptations that give sperm a selective advantage and increase the individual's success in fertilization. The main goal of my diploma thesis was to determine whether individual morphological variability of spermatozoa is detectable at the proteome level and possibly to determine what changes at the proteome level occur in spermatozoa with changing morphology. Three species of rodents, Mus musculus, Apodemus flavicollis and Microtus arvalis, were selected for this study, and a thorough morphometric measurement of their sperm was performed. Because the greatest variation was detected in the dimensions of the apical hook, only acrosomal proteins were analyzed by mass spectrometry, and then the proteins that distinguished the most sperm with long and short apical hooks were determined by discriminant analysis and machine learning methods. The role of an educational counsellor at elementary school

(in English): The bachelor's thesis deals with the personality and activities of an educational counsellor at elementary school. It presents a system of counselling services at schools in the Czech Republic, acquaints with individual members of this system and their main job description. The text focuses on the role of an educational counsellor, on the personal and qualification prerequisites needed to perform this role. Furthermore, on individual activities that are an integral part of his/her work at elementary school and the related ethical principles. The practical part evaluates the results of the survey that was used to determine the most common activities and methods of work of educational counsellors today and their overall perception of this profession.

Tumor in vitro chemosensitivity and resistance assays (CSRA) using flow cytometry

In vitro chemosensitivity and resistance assay determine the sensitivity of a specific tumor after a specific treatment administration in an experimental setup. A heterogeneous population of cancer cells is exposed to various approved anticancer drugs in short-term ex vivo and their combination thereof. The effect of each drug is then determined based on the viability of specific tumor cells allowing for individual patient treatment using a precise combination of drugs. This approach is an example of the personalized medicine principle, which is focusing on the adjustment of diagnostic procedures and treatment of a specific patient. Therefore, its goal is to avoid treatment failure in patients with poor response to the statistically most effective treatments based on randomized clinical trials. The number of viable cells determined by the flow cytometry provides very accurate statistics for multiparametric analysis. A necessary prerequisite is the presence of dissociated cancer cells in a single cell suspension. This is different from cloning methods, where tumor colonies grow on agar media, or from histocultures, which are specific with its three-dimensional tissue cultivation.
